An oligo (dT)-primed cDNA coding for the gastrin precursor was constructed from poly(A)-containing RNA isolated from feline antral mucous cells. The cDNA was inserted into the EcoRI endonuclease site of lambda gt10 vector. Clones containing gastrin sequence were selected by hybridization to a single-stranded 32p-labeled DNA probe synthesized from the porcine gastrin cDNA by primer extension method. The nucleotide sequence of the cDNA insert (475 nucleotides) revealed 315 nucleotides in the entire mRNA coding region, 46 nucleotides in the 5'-untranslated region, 96 nucleotides in the 3'-untranslated region, and a poly(A) tail of 18 nucleotides. Gastrin is located near the carboxyl end of preprogastrin. A considerable nucleotide sequence homology between the coding region of feline preprogastrin and that of other mammalian species has been found; the feline sequence shows 83% homology with porcine sequence, 86% with human, 80% with rat, and 91% with canine sequence. There always have been three dibasic amino acids in other mammalian preprogastrin published previously. Surprisingly, the first dibasic amino acid, Arg-Arg (57-58 residues), is replaced with Arg-Trp though the other two dibasic amino acids exists. The 14 nucleotide sequence following polyadenylational signal (AAUAAA) is very highly conserved (84%) among mammalian gastrin cDNA.
고양이의 antral mucous cells 에서 분리한 mRNA 와 oligo(dT)를 이용해서 가스트린 전구체를 포함하고 있는 cDNA를 만들었다. 이렇게 만들어진 cDNA를 lambda gt10 운반체내의 EcoRI 절단부위에 넣었다. 돼지의 가스트린 cDNA로부터 합성된 한가닥의 표지된 DNA 탐침과 혼성시켜서 가스트린 서열을 가지는 clones을 골라냈다. 삽입된 cDNA (475 뉴클리오티드)의 염기서열을 정해 보니 번역부위에 315 뉴클리오티드를 가지고, 5' 비번역 부위내에 46 뉴클리오티드, 3' 비번역 부위내에 96 뉴클리오티드, 그리고 18개의 poly(A) 꼬리를 가지고 있었다. 가스트린은 전구체의 카르복실 부분에 있었다. 고양이와 다른 포유동물의 가스트린 전구체에 해당되는 부분의 염기서열을 비교해 보니 매우 큰 유사성을 보였다; 고양이는 돼지와는 83%, 사람과는 86%, 쥐와는 80%, 그리고 개와는 91% 의 유사성을 보였다. 전에 발표된 다른 포유동물의 전구체는 언제나 3 개의 쌍염기 아미노산을 가지고 있다. 놀랍게도 첫번째 쌍염기 아미노산인 Arg-Arg (57-58 잔기)은 Arg-Trp으로 바뀌었다. 그렇지만, 다른 두개의 쌍염기 아미노산은 계속 존재한다.
