To improve the thermostability and organic solvent stability of the lipase, chemical modifications of the enzyme were performed.
Lysine residues of $\underline{Candida}$ $\underline{rugosa}$ lipase were acetylated and succinylated, and activity and stability of these modified enzymes were examined in reversed micelle system. UV difference and flurosence spectroscopic studies were also performed to get some information about structural changes of the modified enzyme.
Both actylation and succinylation resulted in increase of substrate affinity, thermal stability and organic solvent stability of the enzyme. And there was no significant structural changes by the modification.
From the results of the modification, at least one of the cysteine, histidine and tyrosine residue was involved in active site of the enzyme and the histidine residue is most probable. Lysine residue was not involved in active site of the enzyme.
효소의 열안정성과 유기용매에 대한 안정성을 높이기위하여 효소의 화학적 변형을 유도하였다.
$\underline{Candida}$ $\underline{rugosa}$ 리파제의 라이신 잔기를 acetic anhydride와 succinic anhydride 로 화학적 변형을 유도하여, 결과로 나타난 효소의 활성과 안정성 변화를 역미셀(reversed micelle) 반응계에서 조사하였다. UV흡광법과 형광 흡광법이 변형된 효소의 구조분석을 위해 사용되었다.
Acetylation 과 succinylation 두 경우 모두가 효소의 기질친화도, 열안정성, 유기용매 안정성을 증가시켰으며, 이 때 큰 구조변화는 없었다. Acetylation에 의해 효소의 활성과 유기용매 안정성이 동시에 증가될 수 있었으며, 이 때의 열안정성에는 큰 변화가 없었다. 하여, acetylation에 의하여 얻을 수 있는 생물반응공학적 이점이 보다 많았다.
위의 결과들은, 효소의 구조적 변화에 의해서보다는 효소표면의 전기적 성질변화에 의한 것이라 추정된다.
한편, histidine, cysteine, tyrosine잔기 중의 최소한 하나가 효소의 활성잔기에 해당되며, 다른 보고를 참고해보면 histidine이 가장 유력하다. Lysine잔기는 활성잔기에 해당되지 않았다.
