Mus81-Mms4 and Fen1 are DNA endonucleases involved in processing of a variety of DNA structural intermediates formed during DNA transactions such as replication and ho-mologous recombination. Recently, it was reported that Mus81 physically interacts with Rad27 (yeast Fen1) and they are mutually stimulatory in a manner dependent on specific pro-tein-protein interactions. In this paper, we investigated the in vivo significance of the interac-tions observed between the two enzymes and showed that the N-terminal 120 amino-acid re-gion of Mus81 was required for functional interactions with Rad27. Mus81 $\Delta$ 120N lacking the N-terminal 120 amino-acid region displayed catalytic activity comparable to that wild-type when it formed a complex with Mms4, but failed to be stimulated by Rad27. Interestingly, the mus81 $\Delta$ 120N allele showed synthetic lethality when combined with sgs1$\Delta$. Considering that Sgs1 constitutes an alternative pathway in parallel to Mus81-Mms4, our results suggest that the physical and functional interactions observed between Mus81 and Rad27 are physiologically important in resolving toxic recombination intermediates. In support of this, the lethality of sgs1$\Delta$ mus81 $\Delta$ 120N was efficiently rescued by further deletion of RAD52, a key recombination mediator essential to generate the toxic intermediates. Mutagenic analyses of the N-terminal region identified two distinct motifs, named N21-26 (aa from 21-26) and N108-114 (aa from 108-114) important for the in vitro and in vivo functions of Mus81. Our findings indicate that the N-terminal region of Mus81 acts as a landing pad to interact with Rad27 and that Mus81 and Rad27 work conjointly for efficient removal of various aberrant DNA structures.
Mus81-Mms4 과 Fen1 은 DNA 복제 그리고 재조합 등의 DNA 처리 과정들 동안에 형성될 수 있는 다양한 DNA 구조적 중간 생성물들의 처리 과정에 관여한다. Mus81이 효모 Fen1 인 Rad27 과 물리적 상호작용할 뿐만 아니라 이들이 특수한 단백질 상호작용에 바탕을 둔 방식으로 상호적으로 서로를 활성화시킨다는 것이 최근에 보고 되었다. 본 논문에서 우리는 이 효소들 사이에서 관찰된 상호작용의 in vivo상에서의 중요성을 조사했고, Mus81 N-말단의 120개 아미노산 영역이 Rad27 과의 기능적 상호작용에 요구된다는 점을 보였다. N-말단의 120개 아미노산 영역이 없는 Mus81 $\Delta$ 120N 는 Mms4와 복합체를 이룰 시 wild type에 견줄만한 촉매 활동을 보여주지만 Rad27 에 의해 활성화되는데 실패했다. 흥미롭게도, 돌연변이 유전자 mus81 $\Delta$ 120N 는 sgs1$\Delta$ 와 배합되었을 경우 합성적 치사성을 보인다. Sgs1이 Mus81-Mms4와 병행된 대체 경로를 구성한다는 점을 고려해보면, 우리의 결과는 Mus81과 Rad27 간에 관찰된 물리적/기능적 상호작용이 해로운 재조합 중간생성물들을 제거하는데 있어서 생화학적으로 중요하다는 걸 제시한다. 이를 뒷받침하는 증거로써, sgs1 $\Delta$ mus81 $\Delta$ 120N 의 치사성은 해로운 중간생성물들을 생성하는데 있어서 필수적인 핵심 재조합 매개 단백질의 유전자인 RAD52의 삭제로 효과적으로 막아질 수 있다. 돌연변이 도입을 통한 Mus81 N-말단 영역의 분석은 두 뚜렷한 모티프들을 밝혔는데, N21-26 (aa from 21-26) 과 N108-114 (aa from 108-114)는 Mus81의 in vitro와 in vivo 상의 기능에서 있어서 중요하다. 우리의 연구 결과는 Mus81의 N-말단 영역이 Rad27과 상호작용하기 위한 착륙 받침으로 작용하고 Mus81과 Rad27이 다양한 DNA 구조들의 효과적인 제거를 위해 연대하여 함께 작용한다는 걸 나타낸다.
