Pullulan is an α-glucan wherein maltotriose units are repeatedly polymerized via α-1,6 linkages on the terminal glucose and have many good properties to be applicable.
For the efficient production of pullulan from $\underline{Aureobasidium pullulans}$, the study on optimization of culture medium was carried out and the composition of optimized culture medium were as follows; sucrose 5%, yeast extract 0.1%, $K_2HPO_4$, 0.5%, NaCl, 0.1%, $(NH_4)_2 SO_4$ 0.06%, $MgSO_4.7H_2O$ 0.02%. The optimal culture temperature of $\underline{Aureobasidium pullulans}$ IFO 4464 was 27℃. The initial pH of culture medium appeared important factor for the production of pullulan. The lower the initial pH became, the more cells were obtained but the less pullulan was obtained.
The effects of the rate of aeration and agitation on the production of pullulan were investigated. In highly aerated and agitated systems(600 rpm, 2.0 vvm) the maximum production yield and productivity of pullulan were obtained, 60~66% and 0.290g/l.hr, respectively, which were higher values than those of poorly aerated and agitated system (200rpm, 1vvm), having 40% and 0.090g/l.hr, respectively. The rheological properties of culture broth of pullulan fermentation were investigated. The culture broth obeyed the power-law, $\tau = K(\gamma)^n$ and showed pseudoplastic behavior. As time passed, it became more pseudoplastic in proportional to the accumulation of pullulan. When the maximum production of pullulan was obtained during the fermentation process, the minimum non-Newtonian index, n, was obtained, around 0.2. Consistency index, K, increased with the increase fermentation time up to 7 days.
In order to investigate the relationship between concentration of pullulan and kinetic parameters of power-law, two equations were introduced. $ln K(t) = ln \tau_0-n(t) ln \gamma_0, K = AP^B$, where $\tau_0, \gamma_0$ A and B are constant. Experimenta data fitted well to these equations and showed good correlations. Thus under the constant culture conditions, the prediction of rheological properties of culture broth was possible by the measurement of pullulan content. To increase the production of pullulan in a batch, fed-batch fermentations were carried out using a mutant strain, $underline{Aureobasidium pullulans}$ JK, which exhibited more sucrose endurability than wild type of $underline{Aureobasidium pullulans}$ About 60 g/l of pullulan production was achieved by fed-batch fermentation with continuous or exponential feeding system.
불완전 곰팡이의 일종인 $\underline{Aureobasidium pullulans}$가 생산하는 천연다당류인 pullulan은 물성이 뛰어나 산업적 용도가 광범위하게 개발되고 있으며, 이에따라서 pullulan 생산을 증대시키기위한 연구가 집중되고 있다. 본 실험에서는 pullulan 발효에 영향을 미치는 인자, 즉 온도, pH, 교반, 통기, 탄소원과 질소원을 비롯한 배지 조성등을 기초적으로 연구한 결과는 다음과 같다. Sucrose 5%, yeast extract 0.1%, NaCl 0.1%, $K_2HPO_4$ 0.5%, $(NH_4)_2 SO_4$ 0.06%, $MgSO_4 .7H_2O$ 0.02%, 초기pH7.5, 배양온도 27℃. 유기 질소원인 yeast extract 와 무기 질소원인 황산암모늄이 모두 필요하며, 발효중에 질소원이 제한이 되어야 pullulan 생산이 증가된다는 사실을 확인했다. 배양액의 초기 pH도 pullulan 생산에 중요한 역활을 한다. 초기 pH가 낮으면 세포는 많이 자라는 반면, pullulan 은 거의 생산되지 않았다. 초기 pH를 7.5로 했을때 pullulan이 가장 많이 생산되었다. Pullulan발효는 호기적 조건에서 잘 일어나므로 교반과 통기의 속도가 증가할수록 pullulan생산이 증가되는것을 확인했으며, 최고 30 - 35 g/1의 pullulan을 생산할 수 있었다. Pullulan발효는 발효액의 점도가 높아 효과적인 산소전달과 발효액의 충분한 혼합이 이루어지지 않는 특성이 있기 때문에 발효액의 물성학적 성질을 연구해서 물질전달과 혼합에 관한 기본 자료로 삼고 또한 scale-up과 혼합이 잘이루어지는 새로운 발효조 설계를 위한 자료로 삼고자 하였다. Pullulan발효액은 power-law, $\tau = K (\dot{\gamma)^n$ 에 잘 따르며 발효의 시간이 경과하면서 pullulan 생성이 증가할수록 더욱 pseudoplsatic한 성질을 띄며 n값이 0.2정도까지 낮아졌다. Pullulan함량과 이들 power-law상수간의 상호관계를 고찰하기 위해 다음 두식을 도입했다. lnK(t)=$ln$\tau_o-n(t)ln\gamma_o$, $K=AP^B$. Pullulan발효액은 이들 식에의해 잘 표현되며 pullulan함량 측정에 의해 발효액의 물성을 예측할수있다. 발효조당 pullulan생산량을 늘리기위해 연가식 (fed-batch) 발효도 수행했다. 이 실험에서는 고농도의 sucrose에 대해 좀더 강한 내성을 보이는 변이균주 $\underline{Aureobasidium pullulans}$ JK를 이용해서 실험한 결과 60 g/l의 pullulan 을 생산할수 있었으며 이는 약 50%의 탄소원의 전환수율이다.
