The biosynthesis of cephalosporin C (CPC) and sulfur metabolism by methionine analogue (DL-seleno-methionine) resistant mutants of $\underline{Cephalosprium acremonium}$ were studied.
In the parent cell, the minimum inhibitory concentration (MIC) of DL-seleno-methionine on growth was 3 ㎍/ml. There were three main groups in the methionine resistant mutants selected after N'-nitro-N-nitrosoguanidine (NTG) treatment.
The first group was the most resistant to DL-seleno-methionine than other mutants and had decreased activity for L-methionine uptake. The second group with increased activity for sulfate uptake and decreased activity for L-methionine uptake, and utilized methionine by somewhat diffusion. The third groups of increased activities for sulfate and L-methionine uptake, accumulated excess methionine from sulfate, and had less inhibitory effects on the function and biosynthesis of sulfate permease by cysteine and methionine than the parent cell.
The mutants with increased activity for sulfate uptake produced more CPC from sulfate than the parent cell, and replaced methionine by sulfate as sulfur source in the production of CPC.
It was revealed that L-cysteine is an inhibitor and L-methionine is a repressor in the sulfate permease of $\underline{C.acremonium}$. And enzymatic studies indicated that L-cysteine might act as a metabolic regulator on the sulfate utilizing enzymes.
Methionine 유사체(DL-seleno-methionine) 에 내성을 갖는 $\underline{Cephalosporium acremonium}$ 변이주에서의 cephalosporin C (CPC) 생산성과 황화합물 대사의 상호관계에 대해서 조사해 보았다.
모균의 경우 이 methionine 유사체에 대해 성장을 저해하는 최저 농도는 3ug/ml 이었는데, 돌연변이 유도물질인 N'-nitro-N-nitrosoguanidine (NTG) 처리로 이 물질에 내성을 갖는 여러 변이주를 얻을수 있었다.
첫째, 이 methionine 유사체에 가장 내성이 컸으며, L-methionine permease 가 크게 손상된 변이주 291-1 이었고, 둘째, 모균보다 sulfate permease 의 활성도가 증가된 반면, L-methionine permease 의 활성도가 크게 감소된 변이주 MAR-80 이었고, 이 변이주는 methionine 을 어느 정도 확산에 의해 이용했다. 셋째, sulfate permease 와 L-methionine permease 의 활성도가 모균보다 증가 되었으며, sulfate 에서 과량의 methionine 을 세포내에 축적하는 변이주 MS-92 이었고, 이 변이주의 sulfate permease 는 이의 작용과 생합성을 저해하는 L-cysteine과 methionine 에 대해 모균보다 적게 영향을 받았다.
모균보다 sulfate permease 의 활성도가 높은 변이주의 경우, sulfate에 서 보다 많은 양의 CPC 를 생산했으며, 최대 CPC 생산을 위해 모균보다 작은양의 methionine 이 요구되었다.
C. $\underline{acremonium}$ 균주에서 L-cysteine 은 sulfate permease 의 합성을 저해하는 물질이었으며, methionine 에서 자란 균은 sulfate permease 를 거의 생성하지 않았다. 또한, L-cysteine 은 sulfate 를 이용하는 효소들을 조절하는 대사 산물로 추정되었다.
