The binding of cytochrome c to phospholipid vesicles was studied by the ultracentrifugation method as a function of pH and ionic strength in order to correlate the binding properties with membrane fusogenic properties of this protein. The pH dependent binding profile of the cytochrome c is complicated showing binding maxima around pH3 and pH9. Appreciable binding persistes even at high ionic strength and the binding reaction is partially irreversible indicating the involvement of hydrophobic interaction. The cytochrome c induced fusion of phospholipid vesicles, as observed by the intermixing of vesicles contents, also shows a complicated pattern. Above pH7, there is a fusion maximum at around 9. Below pH7, however, the extent of fusion increases steadily with decreasing pH.
The relationship between the binding of cytochrome c to vesicles and the cytochrome c induced fusion of vesicles is discussed in terms of hydrophobic interaction between this protein and vesicles.
인 지질 막에 대한 Cytochrome C 의 결합을 Cytochrome C의 막 융합 성질과 비교해 보았다. Cytochrome C 의 pH에 따른 결합 양상은 복잡하였는데 pH 3 과 pH 9 근처에서 최고를 나타내었다. 이러한 결합은 높은 이온 강도하 에서도 일부 유지 되었으며 이는 이 결합에 소수성 작용이 관여함을 나타낸다. 또한 인지질 막 내부의 수용액 성분의 혼합으로 Cytochrome C에 의해 유도되는 융합을 살펴본 결과 역시 상당히 복잡한 양상을 보였는데, pH 7 이상의 알카리성 수용액 에서는 pH 9근처에서 최고치를 나타내었으며, pH 7이하의 산성 수용액 에서는 pH 가 낮아 질수록 융합 정도가 높아졌다. 이러한 Cytochrome C 에 의한 인지질 막에 대한 결합 양상과 융합 양상에는 소수성 결합 작용이 작용함을 알수 있으며, 이는 Cytochrome C 의 C-말단 부분의 소수성 부분이 작용하는 것으로 생각되어 진다.
