The objective of the present study is to propose a method of deconvolution in highly overlapped chromatograms and to show the applicability of the technique to real chromatograms.
The method is an extension of the previously reported work by Jung et al. which involves the solution of the cubic or quartic equation of τ. The deconvolution procedure is performed adopting Marquardt formalism. The study was carried out on the various effects of the degree of resolution, the different peak sizes, the random noise level, and constant peak shape on the deconvolution of overlapped peaks. In all cases the area factors, A's, which were linearly proportional to concentrations were reproducible within 2.0% in present technique at the noise value up to 1.0%. And the applicability of the technique to quantitative analysis has been discussed in detail.
본 연구 목적은 고중첩화된 크로마토그람을 비중첩화하는 한 방법을 제안하고 실제 크로마토그람들에 본 방법의 응용성을 보이려는데 있다.
본 방법은 본 실험실에서 보고한 방법의 연장이며 그 방법은 τ에 대하여 3차 혹은 4차 방정식을 풀어야한다. 비중첩화 과정은 Marquardt formalism 으로 수행된다.
본 연구에서는 피크 크기 차이, resolution 의 정도, random noise 준위 및 일정한 피크 모양이 중첩화된 피크들의 비중첩화에 미치는 영향을 연구하였다. 이런 모든 경우들에서 농도에 선형 비례하는 면적 인자 A는 1.0% noise value 에서 2% 이내의 재현성을 보였다. 본 방법에 대한 정량적 비중첩화에 대하여 자세히 검토하였다.
