Mammalian T-type $Ca^{2+}$ channels are encoded by three separate genes ($Ca_v$ 3.1, 3.2, 3.3). In mammals, T-type channels are reported to be sleep stabilizers that are important in the generation of the delta rhythms of deep sleep, but controversy remains. Progress in identifying the precise physiological functions of the T-type channels has been hindered by many factors, including possible compensation between the products of these three genes and a lack of specific pharmacological inhibitors. Invertebrates have only one T-type channel gene and its physiological functions are less well-studied. In this study $Ca-\alpha 1T$, the only $Ca_v3$ channel gene in the \emph{Drosophila melanogaster} genome, was cloned and expressed in \emph{Xenopus} oocytes or HEK-293 cells, and verified whether it is capable of passing typical T-type currents. Voltage-clamp analysis revealed that the biophysical properties of $Ca-\alpha 1T$ show mixed similarity to three mammalian T-type channels. It was also found that $Ca-\alpha 1T$ is broadly expressed across the adult fly brain in a pattern vaguely reminiscent of mammalian T-type channels. In addition, flies lacking $Ca-\alpha 1T$ show an abnormal increase in sleep duration that is most pronounced during subjective day under continuous dark conditions despite normal oscillations of the circadian clock. This study suggests invertebrate T-type $Ca^{2+}$
channels promote wakefulness rather than stabilizing sleep.
전압 의존성 칼슘 채널은 세포막에서 막전위 변화를 인식하여 칼슘을 통과시키는 단백질이다. T형 칼슘 채널은 전압의존성 칼슘 채널의 한 종류로서 비교적 빠른 불활성 속도와 작은 전도도를 특징으로 한다. 또한 T형 칼슘 채널은 다른 전압 의존성 칼슘 채널보다 낮은 막전위에서 활성이 시작되어 신경 세포에서 저역치 칼슘 전도 (low-threshold spike)를 생성하며 다발적 활성화 (burst firing)를 일으키는데 중요한 역할을 한다. 뇌의 시상 세포는 비렘 (NREM, non-rapid eye movement) 수면 상태에서 규칙적인 다발적 활성화 (burst firing) 현상을 보이며 이를 통해 델타 ($\delta$)형 수면 뇌파 (electroencephalogram)를 형성하여 다발적 활성화에 기여하는 T형 칼슘 채널 또한 수면 뇌파 형성에 중요하다고 생각되어져 왔다. 그러나 포유 동물에서는 세 종류의 T형 칼슘 채널 단백질을 모두 조절하기 힘들어 T형 채널이 수면에 미치는 정확한 역할을 밝히는데 어려움이 있었다. 이 연구에서는 한 종류의 T형 칼슘 채널을 가지고 있는 노랑초파리 (\emph{Drosophila melanogaster})를 이용하여 T형 칼슘 채널의 역할을 밝히고자 하였다. 먼저 초파리 T형 칼슘 채널 유전자를 복제하여 개구리 난세포 (\emph{Xenopus} oocyte)에 발현시켜 초파리 T형 채널이 전형적인 T형 칼슘 채널의 전기 생리학적 특징을 가지고 있음을 확인하였다. 또한 녹색 형광 단백질(GFP, green fluorescent protein)과 결합된 초파리 T형 채널 단백질이 초파리 성체 뇌에서 전반적으로 발현하고 있음을 밝혔다. 마지막으로 유전자 조작을 통해 초파리 T형 칼슘 채널이 수면을 억제시키는 역할을 하고 있음을 밝혔으며 이러한 역할은 신경 세포에 발현하는 T형 칼슘 채널을 통해 이루어지고 있음을 확인하였다.
