Previously we have shown that the chromatophore preparation of $\underline{Rs}. \underline{rubrum}$ is able to reduce $NAD^+$ photochemically through the direct electron transfer to the ferredoxin coupled $NAD^+$ -oxidoreductase system. On the while, it has also been reported that there exists a nitrogenase system in the crude cell extract of the same bacteria $\underline{Rs}. \underline{rubrum}$, but its reductive activity for nitrogen fixation is found to be light-independent rather it requires other electron donors as in the case of the known nitrogenase system.
In this paper, we wish to report our new finding that by mixing the bacterial chromatophore preparation with the crude extract of nitrogenase, the light induced reductive activity of nitrogenase could be demonstrated in the presence of exogenous electron donors such as succinate or DCPIP. This light dependent generation of reductant could be occurred by noncyclic electron transfer, and was electron donor dependent. We also observed that pyruvate served as electron donor for nitrogenase in the dark. From a series of kinetic experiments it was suggested that the bacterial chromatophore was able to couple with a high potential reductive enzyme system so that it performs photochemical reductive processes like a hydrogenation reaction.
최근에 광합성 세균에서 분리한 Chromatophore 가 Ferredoxin 을 통한 직접적인 전자 전달에 의해 광화학적으로 $NAD^+$ 을 환원시킬수 있다는 보고가 있었다. 또한 같은 세균에서 Ferredoxin과 같이 강력한 환원제를 요구하는 질소 고정 효소가 발견 되어 있다. 그러나 질소 고정에 필요한 환원력은 빛에 의해 형성되지 않고, 다른 질소 고정 세균 처럼 화학적인 전자 공여체를 요구하고 있다고 알려져 왔다.
우리는 Chromatophore 와 세균의 추출 용액을 서로 연계시켜, 외부의 전자공여체가 관여된 일련의 실험을 통해 질소 고정 효소의 환원력이 빛에 의해 형성 된다는 사실을 관찰하였다. 이러한 환원력은 Noncyclic 전자 전달계에 의하여 빛이 존재할 때 형성되며, 외부의 전자 공여체의 농도에 의존적이다. Noncyclic 전자 전달에 따른 광환원 작용은 광인산화 작용과 서로 길항적으로 이루어진다. 또한, Pyruvate 가 빛이 없을 때에도 질소 고정 효소의 환원제로 작용하는 것을 관찰하였다.
이러한 일련의 실험을 통해 볼 때, 이 세균은 질소 고정 효소처럼 강력한 환원력을 요구하는 System 에 대하여 광의존적으로 환원력을 제공 할 수 있다고 여겨진다.
