A study on purification of rifamycin B oxidase by affinity chromatography and its biochemical properties.
Purification of rifamycin B oxidase from a microbial strain, $\underline{Humicola}$ spp. ATCC 20620, which is known to catalyze the oxidative reaction of rifamycin B to rifamycin O, was achieved using affinity chromatography as a single purification step.
The preparation technique of affinity gels in affinity chromatography involves
(a) Covalent attachment of ligands to matrix gels through amino, carboxyl, phenolic, alcoholic hydroxyl, or keto groups.
(b) Changing the length of ligands attached to the gel matrix backbone.
An optimal condition for the preparation of affinity chromatography column was investigated in this work. This step showed excellent purification yield and resulted in a preparation.
The purified enzyme showed a max. activity at pH 7.5 and 50℃ and had $K_m$and $V_{max}$ of 1.876mM and 21.28 mM/h.
Rifamycin B oxidase는 $\underline{Mocillium}$ spp. ATCC 20621과 $\underline{Humicola}$ spp. ATCC 20620으로 부터 추출되는 효소로 rifamycin B를 산화시켜 rifamycin S로 가수분해 되는 반응을 촉매한다.
본 연구에서는 이 효소의 기질인 rifamycin B와 이 화합물과 구조가 유사한 방향족 화합물을 affinity ligand로 하여 rifamycin B oxidase 를 정제 했으며 matrix와 ligand사이에 도입되는 spacer arm의 길이와 1igand 의 작용기가 결합 친화력에 어떤 영향을 미치는 가를 조사하고 또 대략적인 효소 특이성을 조사했다.
이 화합물들을 ligand로 한 affinity adsorbent에서 정제된 효소는 효소 반응성, 최적PH, 최적온도 금속온도 금속이온효과 등에서 알려진 rif amycin B oxidase 의 성질과 일치했다.
