The hydrolysis of triacylglyceride by reversed micelle of the lipase was studied. The enzyme was solubilized in reversed micelle of bis (2-ethylhexyl) sodium sulfosuccinate (25 mM, AOT) in isooctane and water (0.31%). The enzyme activity depended on the molar ratio of water to AOT (R value), and maximum activity was observed when the 'R value' was 7.
It is remarkable that under the conditions investigated, the reaction rate in the micelle was ca. 6 times faster than that in aqueous emulsion system. The pH-activity profile showed that optimum pH was 7.3 in isooctane-AOT micellar solution.
Optimum temperature was at 30-35℃. Also, Ea, activation energy was 7.9 Kcal/mole. When 'R' value was seven, apparent Km and Vmax for olive oil were 0.80% and 51 uM/mg/hr, respectively. In the enzyme stability against isooctane, the relative activity of the lipase entrapped in AOT-isooctane rapidly dropped to ca. 50% after 2 hours.
The order of the reaction complied with 2nd order reaction.
Isooctane-AOT reversed micellar solution 에서 C. rugosa 로 부터얻은 Lipase 에 의한 olive oil 의 가수 분해에 대해 실험을 하였다.
실험결과에 의하면 본 system 에서는 lipase 의 최대활성도가 R=7에서 관찰되었고 최적 $P^H$ 는 7.3 으로 확인 되었다. 한편 최적 온도는 30℃ - 35℃ 범위 였으며, Ea 는 7.9 Cal/mole, 그리고 Olive Oil에대한 Km(app) 와 Vmax(app)는 각각 0.80\%, 51 uM/mg/hr 였다.
Isooctane 에대한 "효소활성도 - 시간" 결과에서는 활성도가 2 시간 이내에 처음활성도의 약 50% 가 파괴되고, 14시간 까지는 약 45% 가량이 남는 것으로 나타났다.
본 system 과 emulsion system 에서의 반응속도 비교결과 전지에서 약 6 배 가량 속도가 빨랐으며 본 system 에서의 반응은 2차 반응을 따랐다.
