Monoclonal Antibody specific for $\underline{Cl}. \underline{botulinum}$ type F toxin was prepared using the so-called hybridoma technique. Toxin was purified through two ion-exchange chromatographic steps. It was made toxoid and then injected into BALB/C mice according to immunization schedule. After boosting injection their splenocytes were isolated and fused with NS1, one of myeloma cell lines. Hybrids were selected by HAT medium, screened by ELISA technique and cloned by limiting dilution method. Again, clones were screened by ELISA one clone was chequed as powerfully positive. We named it JY101 and cultured in 50ml T-flask. Its culture supernatant showed neutralizing activity against type F toxin. And it permits no crossreaction with botulinum toxins of other types tested.
조직 배양기술을 응용하여 botulinum F 형 독소를 중화시키는 단일항체를 개발하였다.
(1) 단일 항체를 만드는 JY 101의 생장 곡선은 NS1 과 유사하였다.
(2) 단일 항체는 면역학적 효소 반응 검출법으로 정성할때 강한 양성 반응을 보였다.
(3) 단일 항체는 F 형 독소를 중화시켜 그의 항원 결정 부위가 작용부위 근처임을 보였다.
(4) 단일 항체는 F type 에만 작용한다. 따라서 F 형에만 있는 항원 결정 부위에 대한 항체이다.
