Choline acetyl transferase (EC 2.3.1.6) (ChAT) present in cholinergic nerve endings catalyzes the synthesis of acetyl choline (Ach) from choline and acetyl-CoA.
The membrane-bound form (integral membrane protein) of the choline acetyl transferase was prepared from squid head ganglia. The preparation procedure includes repeated hypoosmotic disruption and washing by ultracentrifugation in a high ionic strength buffer. It appears that approximately 20% of the synaptosomal choline acetyl transferase is present as a integral protein, non-ionically bound to membrane.
It was found that at high concentrations, the acetylcholine(Ach) inhibits soluble ChAT to a greater degree than the membrane-bound enzyme form. The activities of soluble and membrane-bound ChAT also decrease with increasing calcium concentration but the activity of soluble ChAT is inhibited less than that of membrane-bound ChAT. The activity of soluble ChAT is completely lost at 2.0 mM SDS concentration whereas the membrane-bound ChAT still exhibits about 74% of the control value at 3.0 mM SDS concentration. Soluble ChAT exhibits a sharp pH optimum near 6.6 in potassium phosphat buffer wheas membrane-bound ChAT shows a broader pH-activity profile. Optimum temperature of both soluble and membrane-bound forms of ChAT is 32-35℃ but over the range of 35-56℃, the membrane-bound ChAT is more stable than the soluble enzyme. Soluble and membrane-bound forms of ChAT exhibit identical Km values of approximately 0.12 mM for the substrate of acetyl-CoA.
These results strongly indicate that two forms of ChAT, one soluble and one membrane-bound, are present in squid head ganglia.
회수된 synaptosome ($P_2$) 을 반복된 저삼투에 의한 파열과 100 mM $KH_2PO_4$ 완충용액 (pH 6.8) 에서 초원심 분리 하여 synaptoplasm에 용해된 soluble ChAT 와 막에 비 이온적으로 결합된 membrane-bound ChAT 를 분리하였다.
$WP_4$ 부분 (membrane-bound ChAT) 의 효소 활성은 $P_2$ (synaptosome) 로 부터 회수된 전체 ChAT 활성의 약 20% 를 차지 하였다.
높은 acetyl choline 농도에서 soluble form 의 효소 활성이 membrane-bound form 보다 훨씬 높은 정도로 저해되었다.
Calcium 농도를 10 mM 에서 100 mM 로 증가시킴에 따라 soluble form과 membrane-bound form의 효소 활성은 유사한 형태로 감소 하였으나 soluble form 의 효소 활성이 전체적으로 membrane-bound form보다 덜 저해되는 것으로 나타났다.
2.0 mM SDS 농도에서 soluble form 의 효소활성은 완전히 소실 되었으나 membrane-bound form은 3.0 mM SDS 농도에서 기준점 효소활성의 약 74% 를 가지고 있었다.
Soluble form의 최적 pH 는 6.6 근처에서 sharp 한 반면 membrane-bound form 은 훨씬 더 broad 한 pH-활성 프로필을 보였다.
Soluble form 과 membrane bound form 의 최적 온도는 공히 32℃-35℃ 이었으나 35℃-56℃ 영역에서 membrane bound form 이 soluble form 보다 더욱 안정하였다.
Acetyl-CoA 에 대한 $K_m$ 값은 soluble form 과 membrane-bound form 공히 0.12 mM 이었다. 이러한 결과들은 squid head ganglia 에서 ChAT 의 membrane-bound form 이 존재한다는 것을 시사한다.
