The purification of angiotensin I-converting enzyme which is known to act as a key enzyme in the control of blood pressure through the Renin-Angiotensin-Aldosterone system, was achieved using affinity chromatography as the principal purification step.
The angiotensin I-converting enzyme was partially purified from hog lung homogenates by acidification, DEAE-cellulose chromatography and ultrafiltration. The kinetic properties and the effect of sodium chloride have showed the known characteristics of angiotensin I-converting enzyme. Therefore, it was confirmed that the enzyme was angiotensin I-converting enzyme.
An effective affinity gel was prepared on Sepharose 6B matrix using succinyl-L-proline competitive inhibitor as a ligand and spacer arm was introduced by 1,6-hexanediamine between the matrix and a ligand. With this affinity column, the enzyme preparation showed high degree of purity and good purification yield.
Angiotensin I 전환 효소는 혈압 조절과 관계하는 Renin-Angiotensin-Aldosterone 계의 중요한 효소로 알려져 있다. 본 연구에서는 affinity chromatography 를 주요한 단계로 하는 angiotensin I 전환 효소의 정제를 시도하였다.
전 단계에서 acidification 과 DEAE-cellulose 이온 교환 chromatography, 그리고 ultrafiltration 을 사용하여 돼지 허파의 TritonX-100 추출액을 부분 정제하였다. 효소의 반응성, 최적pH, NaCl 효과 등은 알려진 angiotensin I 전환 효소의 성질과 일치되므로 분리 되어진 효소가 원하는 angiotensin I 전환 효소임을 확인하였다.
상경적 억제물질인 succinyl-L-proline 을 affinity ligand 로 하여 Sepharose gel 에 1,6-Hexanediamine spacer 를 사이에 두고 결합시킴으로써 효과적인 affinity adsorbent 를 준비하였다. 이렇게 준비된 affinity adsorbent 를 사용하여 정제된 angiotensin I 전환 효소는 높은 순도와 좋은 수득률을 보여 주었다.
