A new method to convert the primary amines of heterocyclic compounds to the corresponding halides using tertiary butyl thionitrate(t-BuS-N=O) or tertiary butyl thionitrate ($t-BuSNO_2$) was developed. The reaction of heterocyclic primary amines with t-butyl thionitrite or t-butyl thionitrate in the presence of Copper(Ⅱ) halides in aprotic media such as acetonitrile, tetrahydrofuran afforded the corresponding halides under mild conditions (ca. 25℃). Since the sulfur-nitrogen bond of thionitrite or thionitrate is relatively weaker than the oxygen-nitrogen bond of nitrites and also the stronger complexation of thio-derivatives with copper salt than the oxygen and chloro analogs, thionitrite or thionitrate is considered to be better reagents for deaminative halogenation of heterocyclic primary amines.
t-Butyl thionitrate afforded the halo compounds in better yields (70-95%) than the t-butyl thionitrite (30-42%). In case of fluorination, heterocyclic primary amines such as (2',3',5'-triacetyl)-9-D-ribofuranosyl 6-aminopurine, 2-amino-5-nitrothiazole were reacted with $NaBF_4$ instead of Copper (Ⅱ) fluoride to afford the corresponding fluoro compounds in good yields (62-100%).
시 산화 질소와 메르？탄을 사용하여 티오나이트라이트와 티오나이트 레이트를 만든 후 아민기를 가진 헤테르 고리 화합물 과 아세로나이트 아일 용매하에서 카파 합라이드 존재에서 반응시킨 결과 좋은 수율로써 할로겐으로 치환된 헤테로 고리 화합물을 얻을 수 있었다.
헤테로 고리 화합물을 티오나이트 라이트를 사용하는 것보다 티오나이트레이트와 반응 시키면 좋은 수율로 할로겐으로 치환된 헤테로 고리 화합물을 얻었다.
플루오린으로 치환할 경우에는 플루오르화 구리 대신 플루오르화 나트륨을 사용함으로써 종래 어려운 반응으로 알려진 플루오르화 반응에서 비교적 좋은 수율로 플루오르 고리 화합물을 얻었다.
