Genetic aspects of autism spectrum disorders (ASDs) have recently been extensive-ly explored, but environmental influences that affect ASDs have received consider-ably less attention. Zinc (Zn) is a nutritional factor implicated in ASDs, but evidence for a strong association and linking mechanism is largely lacking. Here, I demon-strate that post-synaptic Zn elevation rapidly rescues social interaction in two inde-pendent mouse models of ASD. In mice lacking Shank2, an excitatory postsynaptic scaffolding protein, postsynaptic Zn elevation induced by clioquinol (a Zn chelator and ionophore) improves social interaction. Postsynaptic Zn is mainly derived from presynaptic pools, and activates NMDA receptors (NMDARs) through postsynaptic activation of the tyrosine kinase Src. Clioquinol also improves social interaction in mice haploinsufficient for the transcription factor Tbr1, which accompanies NMDAR activation in the amygdala. These results suggest that post-synaptic Zn elevation induced by clioquinol rescues social deficits in mouse models of ASD through postsynaptic Src and NMDAR activation.
자폐 스펙트럼 질환의 유전적 요소에 대해서는 활발한 연구가 이루어 지고 있으나, 환경적 요소에 대해서는 훨씬 적은 관심만 주어지고 있다. 아연은 자폐 스펙트럼 질환과 연관되어 있다고 알려진 영양 요소인데, 그 연결 고리의 기작에 대해서는 잘 알려져 있지 않다. 나는 본 연구를 통해서, 시냅스후 아연 증가가 두 종류의 서로 다른 자폐 스펙트럼 질환 쥐 모델의 사회적 교감을 빠른 속도로 회복시킨다는 것을 보고하고자 한다. 흥분성 시냅스후 얼개 단백질인 Shank2가 없는 쥐들에서, 클리오퀴놀(아연킬레이터이자 아연운반체)에 의한 시냅스후 아연의 증가는 쥐들의 사회적 교감 정도를 증진시킨다. 시냅스후 아연은 주로 시냅스전 저장소에서 기인하고, 시냅스후 부분으로 들어가 티로신 인산효소인 Src의 활성화를 유발하여 NMDA 수용체의 기능을 증가시킨다. 또, 클리오퀴놀은 전사인자인 Tbr1이 반 밖에 없는 쥐들의 사회적 교감도 증진시키는데, 이는 편도체의 NMDA 수용체의 기능 항진을 수반한다. 이 결과들은 클리오퀴놀에 의한 시냅스후 아연의 증가가, 시냅스후 Src 효소와 NMDA 수용체의 활성화를 통해, 자폐 스펙트럼 질환 쥐 모델들의 사회적 교감 정도를 회복시킬 수 있다는 것을 제시한다.
