In this study, Kluyveromyces. fragilis. was cultured and fixed on the ceramic raschig ring packed in the pyrex culumn (dia 4.3 cm length 60 cm) under the anaerobic condition.
Range of the initial glucose concentration was expanded up to the twice of the limiting substrate concentration i.e., 5- 50 g/l, and the dilution rate were varied from 0.1 to 1.0/hr.
For a glucose limited continuous system using Kluyveromyces. fragilis. maximum productivity 1.0 g/l is obtained at the initial grucose concentration of 20 g/l and dilution rate of 0.33/hr. It was also found that the consumption rate of glucose, $(S_o-S_e)/S_o$, was not a function of the initial concentration or dilution rate but a function of D·$S_o$.
And the fact that the area constant in this experiment was linearly increased with increasing the dilution rate was derived.
ceramic raschig ring 을 충진한 pyrex colum ( dia 4.3 cm, Lengh 60 cm ) 을 사용하여 혐기성 조건하에서 Kluyveromyces fragilis 를 배양시켜 균을 충진물 주위에 골고루 입혀서 Glucose 농도는 Limiting Substrate 구간의 2배까지 즉, 5 - 50 g/l 로 Dilution rate 는 0.1 - 1.0 까지로 하여 실험을 하였다.
연속 system 에서 실험을 한 결과 초기농도가 20 g/l, Dilution rate 가 0.33 이상에서 productivity의 최대값인 1.0을 얻었다. 또한 Glucose 가 소모되는 율 $(S_o - S_e) / S_o$ 는 초기농도 Dilution rate 만의 함수가 아니라 $D S_o$의 함수임이 실험을 통하여 밝혀졌으며, 본 실험장치인 Reactor 의 Area constant 는 Dilution rate 에 linear 하게 증가한다는 결론을 얻었다.
