Penicillin G acylase (EC 3.5.1.11, penicillin amidohydrolase), which is responsible for the hydrolysis of benzylpenicillin to 6-aminopenicillanic acid (6-APA) and phenylacetic and (PAA), was produced by $\mbox{\underline{Bacillus}}$ $\mbox{\underline{megaterium}}$ ATCC 14945. The optimal conditions of enzyme reaction were set as: at pH 8.5 to 9; at 40℃. The Km value for benzylpenicillin was 4.55 mM. Hydroxylamine assay for the determination of 6-APA was modified by using "HCl addition technique" instead of n-butylacetate extraction step.
Using above microorganism, the effects of fermentation conditions on enzyme production were studied. When the following medium composition was used, the enzyme productivity obtained was about 14.1 units per ml broth. The medium composition ranges were: casitone, 2.5 to 5%; yeast extract, 0.5 to 1.0%; glucose, 1.0 to 2.0%; benzoic acid, 0.1 to 0.2% or phenylacetic acid 0.15 to 0.3%; and silicone antifoam, 0.01 to 0.02%. Proper physicochemical conditions of fermentation or enzyme production were; initial pH of medium, 7.0; temperature of cultivation 28 to 30℃ ; and fermentation period, 2 to 3 days under highly aerobic condition.
Penicillin 으로 부터 6-aminopenicillanic acid (6-APA) 를 제조하는데 관여하는 penicillin G acylase 를 Bacillus megaterium ATCC 14945로 부터 얻었다.
이 효소의 최적 반응 조건은 40℃, pH8.5 - 9.0 으로 판명되었으며 Km 값은 4.55 mM 이었다. 한편 효소 반응 생성물인 6-APA 를 정량하기 위하여 n-butylacetate 추출단계가 필수적인 기존의 hydroxylamine 방법을 개량하였다. 용매 추출 대신에 염산을 반응 혼합물에 적당량 첨가(0.8N HCl 과 반응 혼합물을 동량 섞어줌) 함으로써 소기의 목적을 이룰 수가 있었다.
이 미생물과 상기의 개량된 정량법을 이용하여 여러 발효 조건들이 효소생산에 미치는 영향을 조사하였다. 배지성분으로 casitone 2.5 - 5.0%, yeast extract 0.5 - 1.0%, glucose 1.0 - 2.0%, benzoic acid 0.1 - 0.2% (또는 phenylacetic acid 0.15 - 0.3%)를 사용하여 배지의 초기 pH 를 7.0 으로 맞춘 후 29℃ 에서 높은 통기조건으로 약 2, 3 일 배양한 경우 효소 생성 능력은 뚜렷한 증가를 보여 처음의 효소 역가의 약 3배에 달하는 14.1 units/ml broth 를 나타내었다.
