In searching for an efficient method to prepare graft-copolymers of proteins, the oxidation reaction by cerium(IV) ion was chosen and the graft copolymerizations of two vinyl monomers to silk fibroins were investigated. Ceric ion oxidation of an organic compound involves the initial formation of complex and the subsequent transfer of an electron to ceric ion. The intermediacy of free radicals in the oxidation reaction has been widely accepted and applied well to the radical graft copolymerizations of vinyl monomers to natural and organic polymers. A well known protein fiber, silk contains various polar amino acid residues, which could provide the reactive grafting sites upon the oxidation by ceric ion.
In a typical graft copolymerization of acrylamide(AM)to silk fibroins, woven silk fabric of certain weight was immersed at 30℃ in a stirred aqueous reaction mixture of ceric ammonium intrate (CAN) (0.0025M), AM(0.5M) and acetic acid(25%). Bath ratio of the silk sample was 1 to 200 and the reaction time was 3 hours. Degree of grafting was obtained by measuring the weight increase of the silk sample after thorough washing with hot water. Change in CAN concentration exhibited a maximum in degree of grafting at 0.01M. Also observed was that change in acetic acid content in reaction media gave a maximum in degree of grafting at 7% acetic acid in water. Degree of grafting was increased generally with increase in AM concentration, reaction time and reaction temperature.
When acrylonitrile(AN) monomer was used for grafting, different results were obtained. Addition of nitric acid enhanced the degree of grafting more than the addition of acetic acid. Generally the grafting of AN to silk fibroins was less efficient than the grafting of AM.
An AM-grafted sample was subjected to the extractions with Lowe's reagent to remove ungrafted portion of silk. IR spectra of the insoluble portion in Lowe's reagent exhibited the absorption bands characteristic to both vinyl polymers and silk fibroins, indicating the grafting of vinyl monomers to silk fibroins.
To examine the molecular weight of grafted vinyl polymer a sample of grafted silk was hydrolyzed by 10% sodium hydroxide. The grafted vinyl polymer was recovered from the hydrolyzed mixture by precipitating in methanol. Viscosity measurements indicated that the molecular weight was in the range of $10^5$.
단백질에 대한 효과적인 그라프트 공중합을 검토하기 위하여 cerium (IV) 이온에 의한 산화반응을 택하였다. Cerium (IV) 염은 일전자 산화제로서 산화성기를 가진 유기화합물을 산화시킬 때 착물을 만들고 산화중간과정으로 자유 라디칼을 생성한다고 알려져 있는 데, 생성된 자유 라디칼이 비닐 모노마에 포착되면 중합반응이 개시된다.
단백질 섬유인 견섬유는 알코올, 아민, 카르복실기등 극성 아미노산 잔기를 많이 갖고 있으므로 cerium (IV)이온에 의하여 산화되면 자유 라디칼이 생성되어 비닐 모노마의 그라프트 공중합이 가능하게 된다. 견섬유에 대한 그라프트 반응에서 cerium (IV)염으로는 ceric ammonium nitrate (CAN) 을 사용하여 중합반응을 개시하였고 비닐 모노마로는 acrylamide (AM)와 acrylonitrile (AN)을 사용하였다.
견섬유에 대한 AM 모노마의 그라프트 공중합반응에서 표준반응조건으로 CAN 0.0025M, AM 0.5M, 초산 25%, 반응온도 30℃, 반응시간 3시간으로 하여 실험을 하였다. 이때 견섬유의 반응용액에 대한 욕비는 1:200 으로 하였다. CAN 농도를 변화시켰을 때 약 0.01M 에서 그라프트율의 최대를 보이고, 또한 초산의 양을 변화시키는 경우에는 약 7% 초산 수용액에서 최대의 그라프트율이 나타났다. AM 농도, 반응온도및 반응시간에 대한 그라프트율을 살펴보면, 이런 변수들의 값이 커짐에 따라 그라프트율도 대체로 증가되었는 데, 반응시간 보다는 반응온도의 영향이 더 크게 작용했고, AM 농도의 증가에 대하여는 더 큰 그라프트율의 증가를 가져왔다. 모노마로서 AN 을 쓰면 AM 의 사용시와는 달리 질산용액에서 그라프트율이 초산수용액에서 보다 더 높았다. 그렇지만 그라프트율은 AM 사용시 보다 현저히 낮았다.
그라프트 견섬유를 10% 가성소다 용액에서 가수분해시켜 그라프트 중합체를 분리하였다. 생성된 호모 중합체와 그라프트 중합체의 분자량을 점도법으로 측정하여 계산한 결과 $10^5$ 정도 였는데, 그라프트 중합체의 분자량이 더 크게 나타났다. 그라프트된 견섬유는 Lowe 시약 (동 - 글리세린 용액) 에서 용해가 잘 안 되었다. Lowe 시약에 대한 그라프트 견섬유의 용해거동과 그라프트 견섬유의 IR 흡수 스펙트럼으로 보아 그라프트 자리수가 매우 적은 것을 알았다. 반응시킨 견섬유의 Lowe 시약에 대한 불용해성과 60℃ 온수에서의 용출실험의 결과 및 IR 흡수 스펙트럼의 비교로 그라프트 중합반응이 일어난 것을 확인하였다.
유기물 - cerium (IV)이온 산화환원계에 의한 비닐 모노마의 중합반응에 대한 반응 메카니즘과 속도론적 고찰을 하였다. Cerium (IV) 이온의 농도에 따라 개시단계와 종결단계에서 서로 다른 반응이 일어나는 것을 알 수 있다.
Cerium (IV) 이온에 의한 그라프트 공중합반응에서 호모 중합체의 생성에 대한 검토실험을 하였고 그 생성이유에 대하여 고찰해 보았다. 견섬유에 대한 AM 와 AN 의 공중합반응에서는 예상보다 많은 양의 호모중합체가 생성되었다. 특히 질산용액에서 AM 의 그라프트 공중합 반응에서는 초산 용액에서 보다 더 많은 양의 호모 중합체가 생성되었다.
