Primary cilia are microtubule-based cellular organelle which functions as sensory antennae detecting chemical and mechanical stimuli from the environment. The presence or absence of primary cilia exerts a profound impact on cell signaling and cell cycle progression. Thus, ciliogenic activity is an important determinant of cell behavior. Recently, actin destabilization has been recognized as a dominant inducer of ciliogenesis. However, exact cellular mechanisms that regulate ciliogenesis remain poorly understood. Here, we show that actin-dependent regulation of ciliogenesis involves two related transcriptional coactivators YAP and TAZ, which respond to mechanical stresses on cells, the architecture of actin cytoskeleton, as well as Hippo signaling. Active ciliogenesis, either in spatially confined cells or in cells treated with an actin destabilizer, coincides with the cytoplasmic sequestration of YAP/TAZ. Moreover, the knockdown of YAP/TAZ is sufficient to induce ciliogenesis; whereas lysophosphatidic acid, which promotes both actin polymerization and YAP/TAZ nuclear accumulation, suppresses ciliogenesis. YAP/TAZ suppress ciliogenesis by promoting the expression of negative ciliogenesis regulators, AURKA and PLK1. We identified actin remodeling factors LIMK2 and TESK1 as key players in the actin-dependent control network wherein YAP/TAZ are integral components. These findings suggest that the actin cytoskeleton provides a critical cue for the regulation of ciliogenesis through nuclear factors.
원발 섬모는 세포 외부의 물리적 그리고 화학적 자극을 세포 내로 전달하는 안테나 역할을 하는 세포 소기관이다. 이외에도 원발 섬모의 존재 여부는 세포 내 신호전달 경로 및 세포 주기에 큰 영향을 미치기 때문에 세포의 증식과 분화 등 세포 운명의 결정에 중요한 기관이라고 볼 수 있다. 최근 들어 액틴 동역학의 변화가 섬모 형성을 촉진하는 주된 요소로 밝혀지고 있으나 섬모 형성을 조절하는 명확한 기전은 아직까지 밝혀지고 있지 않다. 본 연구에서는 액틴 동역학에 의해 유발되는 섬모 형성이 Hippo 신호전달 경로뿐만 아니라 액틴 구조 등 물리적 자극에 의해 조절 되는 YAP과 TAZ라는 전사조절인자의 활성도와 밀접한 관련이 있음을 밝혔다. 세포 밀도가 높거나 액틴 중합 억제제를 처리할 경우 섬모 형성이 유도되는데 이때 YAP/TAZ의 활성 감소 (YAP/TAZ의 세포질 위치)가 동반되어 있음을 발견하였다. 또한 YAP/TAZ를 발현 억제한 경우 무혈청 상태가 아님에도 섬모 형성이 유발되었고, YAP/TAZ를 활성화 시키는 lysophosphatidic acid를 처리할 경우 섬모 형성이 억제되는 것을 관찰하였다. 또한 이러한 현상의 기저에는 YAP/TAZ의 표적 유전자이자 섬모 형성 억제 인자인 AURKA와 PLK1의 발현 정도 변화가 있다는 것을 알아냈다. 더 나아가 본 연구에서는 액틴 조절 인자 인 LIMK2와 TESK1이 이러한 액틴 골격의 변화 및 YAP/TAZ 활성도 변화에 조절되는 섬모 형성에 중요한 역할을 한다고 밝혔다. 이러한 결과는 섬모 형성에서 액틴 동역학이 핵 인자를 통한 유전자 발현 조절을 통해 중요한 역할을 한다는 것을 시사한다.
