The condensin machinery plays a vital role in the partitioning and proper segregation of
chromosomes during cell division in all branches of life. The SMC subunits of the condensin contains
two globular domains, the hinge domain which is responsible for dimerization of two SMC`s and the
head domain which is a ABC ATPase, separated by a long coiled coil domain. Here I present the
crystal structures of the hinge domains of the yeast condensin and the Pyrococcus furiosus condensin
with long coiled coils that resemble rods. I also present solution SAXS envelopes, ALEX-FRET,
TIRF-FRET, and BMOE induced cross-linking data showing that the prokaryotic Smc-ScpAB and the
eukaryotic condensin have coiled coils juxtaposed closely to each other forming rod-like structures. I
also present SAXS data suggessting the same rod-like structure in cohesin. Interestingly, when the
hinge binds DNA, the coiled coils of the Smc-ScpAB takes on a more open form. My data suggest
that a structural change in the head domain induced by ATP binding regulates the association with
DNA. These results reveal a conserved theme in SMC complexes, provide a mechanism for Smc`s
association with chromosomes, and also offer a molecular basis for Cornelia de Lange syndrome.
세포분열 동안 chromosome의 올바른 partitioning과 segregation은 모든생물체에게 있
어서 중요한 일이다. Condensin 복합체는 chromosome condensation을통해 DNA를
partition하며 세포분열시 각 각의 딸세포에게 DNA가 정확히segregate 되도록 한다. 지
금까지 밝혀진 condensin 복합체가 여럿 있다: ㅇ원핵생물에서 Smc-ScpAB, MukBEF,
MksBEF와 진핵생물의 condensin I 과condensin II가 있다. Condensin 복합체들은 다 동
일하게 SMC family의 ~50 nm 정도의 매우 긴 단백질들을 포함하고 있다. gproteobacteria에서는
MukB 또는 MksB를 다른 다수의 bacteria들은 Smc 그리고 진핵생
물에서는 Smc2와 Smc4 가 dimer를 형성한다. Smc 단백질들은 N-terminal 과 Cterminal이
engage하여 ABC ATPase인 head를 형성하며 반대편에는 또 다른 globular
domain인 hinge가 있고 bacteria에서는 homodimer 그리고 진핵생물에서는 heterodimer
를 형성 한다. 이 두 globular domain들은 많은 연구가 지금까지 되어 왔지만 이 둘의
중간에 위치한 기다란 coiled coil domain은 무슨 역할을 하는지 지금까지연구과 미흡
했다.
여기서 나는 150 A 정도의 coiled coil을 포함한 효모 Smc2-Smc4 hinge
구조와 70 A 정도의 coiled coil을 포한한 P.furiosus Smc hinge 구조를 규명하였다.
두 구조에서는 이전에 규명된 Smc hinge 구조들과는 달리 coiled coil이
juxtaposed되어 닫혀있는 것으로 나온다. 두 구조에서는 EM에서 볼수 있는
holocomplex들의 rod shape과도 matching이 잘된다. 그리고 SAXS와 FRET을 통해
이 닫힌 coiled coil 구조가 실험적 artifact가 아니라는 것을 볼수 있었다.
이러한 juxtapositioning은 Smc coiled coils들의 permanent한 구조는 아닐
것이라고 DNA와 ATP를 포함한 BMOE cross-linking 실험들은 말한다. 실험들의결과는 마치 coiled coil의 closed form (juxtapositioning)은 resting state이며 ATP를
통한 head domain의 engagement과 hinge domain의 DNA binding이 open form (ring
formation)이 되게 하는 것으로 보인다. ATP hydrolysis cycle 동안에 head에 가까운
coiled coil이 conformation 변화가 일어나 여기에 있는 coiled coil이 open form을
갖게 될수 있다. 이 변화는 BMOE cross-linking 실험들의 결과처럼 coiled coil
전체가 더 open form을 취하도록 촉진 할 수 있다. 그래서 hinge domain에 DNA가
binding하는 것을 촉진 할것으로 hypothesize 할 수 있다.
요약하면 본 연구는 어떻게 DNA가 condensin의 hinge domain에 binding하는
것이 open conformation에서만 가능하며 hinge domain이 마치 DNA sensor로
작용하여 DNA binding을 통해 멀리 떨어져 있는 head domain에서의 ATP
hydrolysis cycle을 유도 할수 있다는 것을 보여준다. 본 연구에서는 cohesin
hinge도 juxtaposed된 coiled coils들을 형성 한다는 것을 SAXS로 볼수 있었다. 본
연구를 바탕으로 cohesin hinge를 연구하는 것도 매우 흥미로운 작업이 되겠다.
