For the kinetic analses of plasmid instability and gene expression, batch and continuous cultures, employing a recombinant Escherichia coli W3110 ΔtrpLD $trpR^{ts}$ $tna^-$/pCRT185 as a model plasmid-host cell system, were carried out. In the batch culture, the indirect estimation of cell substrate and product concentrations, which were obtained in on-line b mass spectrometric analysis of exhaust gas from fermentor provided the excellent agreements with the experimental data at both temperature of 37℃ and 42℃. From these indirect estimates various fermentation parameters were evaluated at the repressed and derepressed conditions. Under derepressed condition of 42℃ maintenance coefficient $k_1$ and yield coefficient $k_2$ were found to increase by 70-80% and 10%, respectively, than the values under repressed condition of 37℃. In addition it was also found that specific rates of oxygen uptake and carbon dioxide evolution were also increased by ca. 10% as compared with those determined from host cell culture at 42℃. Substrate consumption rate, however, showed a little difference between 37℃ and 42℃, as with maintenance energy(m) and cell yield (Y). These results suggested that the expression of foreign gene burdened the cell with extra metabolic and energy stresses. In the single-stage continuous culture at derepressed condition, 42℃, plasmid-free cells appeared more rapidly in culture broth as the dilution rate decreases. At the lowest dilution rate D=0.075 $hr^{-1}$, the productivit of cloned gene product and plasmid content showed the highest values of ca. 10 mg-trp/g-cell.hr and 0.5 mg-DNA/g-cell, respectively. Plasmid DNA content per g-cell was decreased from 0.5 mg/DNA/g-cell to .02 mg-DNA/gcell as the dilution rate increased. In order to assess quantitatively the plasmid instabilit, two parameters, i.e., the probability of plasmid loss (P) and the difference in the specific growth rate δ between the plasmid-harboring and plasmid-free cells, were estimated from a set of experimental results conducted in single-stage continuous culture under derepressed condition of 42℃. Simulation results, using the estimated parameters and model equations for heterogeneous cell population containing the plasmid-free and plasmid-harboring cells agreed ver well with the experimental data. By the analsis of sensitivit for plasmid instability, the change of zero time as the starting point for steady state considerably affected the plasmid instability. In addition, it was also found that the plasmid instability was more significantl influenced by the difference in specific growth rate (δ) than the proability of plasmid loss (P). In the two-stage continuous culture, the plasmid stability and specific production rate of cloned gene product could be stabl maintained for more than 500 hrs in second-stage, which is impossible in single-stage continuous culture without selective pressure. It was also found that specific production rate of tryptophan, plasmid content and trptophan snthetase enzyme activity showed the highest values at the lowest growth rate examined. But the specific snthesis rate of TSase enyme showed an maximal value nearby the specific growth rate of 0.12 $hr^{-1}$. Plasmid gene expression efficienc showed a linear relationship with the specific growth rate.
유전자 재조합 균주의 배양에서 plasmid 의 불안정성과 유전자 발현에 대한 속도론적 분석을 수행하였다. 이를 위해 37℃ 이하에서는 plasmid 상의 trp-operon의 발현이 억제되고 42℃ 에서는 발현되는 변이주 Escherichia coli W3110 ΔtrpLD $trpR^{ts}$ $tna^-$/pCRT185 를 사용하여 회분식과 일단계 연속 및 이단계 연속배양을 수행하였다.
(1) 회분배양에서 mass spectrometer 를 이용한 발효배기 가스의 분석을 통해 산소 소비속도 (OUR) 과 이산화탄소 발생속도 (CER) 을 real-time 으로 계산하였고, 이를 세포, 기질 및 산물의 농도를 간접 추정하는데 이용하였다. 세포, 기질 및 산물의 간접 추정치들은 37℃와 42℃ 회분배양에서의 실험결과치와 매우 잘 일치하였다. 한편, 산소와 이산화탄소에 대한 속도상수인 $k_1$과 $k_2$ 값은 37℃에 비해 plasmid 유전자가 발현되는 42℃에서 각각 약 70-80% 와 10% 정도로 증가하였다. 또한, 산소 비소비속도 ($qo_2$) 와 이산화탄소 비발생속도 ($qco_2$)는 plasmid 유전자 발현에 의해 약 20-30% 증가하였으며, 42℃ 숙주세포의 회분배양에서의 $qo_2$와 $qco_2$와 비교할때 약 10% 의 증가치를 보였다. 그러나, 기질 소모속도는 숙주세포와 유전자 재조합 균주에서 큰 차이를 보이지 않았다.
(2) 42℃ 일단계 연속배양에서 plasmid 잃어버린 세포는 희석율이 낮을수록 더 빨리 나타났으며 이와같은 plasmid 불안정성 때문에 산물의 생산성은 안정하게 유지되지 않았다. 낮은 희석율 (D = 0.075 $hr^{-1}$) 에서 산물인 tryptophan 의 비생산속도 ($q_p$) 와 plasmid 함량은 각각 약 10 mg-trp/g-cell.hr 와 0.5 mg-DNA/g-cell로 최고값을 보였으며, plasmid 함량은 희석율이 증가할수록 감소하는 양상을 보였다.
(3) 일단계 연속배양에서의 plasmid 불안정성을 정량적으로 규명하기 위해, 실험결과로 부터 두 상수 δ 와 p 값을 구하였다. 희석율이 증가할수록 growth advantage δ 값은 0.02 에서 0.07 로 증가하였으며 plasmid 소실 확률 p 는 $10^{-2}$ 에서 $10^{-5}$ order 로 감소하였다. 이들 상수값과 model 식을 이용하여 plasmid 불안정성을 예측한 결과 여러 희석율에서도 실험치와 매우 잘 일치하였다. 또한, plasmid 불안정성에 미치는 상수들의 sensitivity 분석에서 plasmid 소실 확률 p 보다 growth advantage δ에 의해 plasmid 불안정성은 더 크게 영향을 받으며 특히, 정상상태의 시발시간에 의해서도 plasmid 불안정성은 크게 좌우됨을 알았다.
(4) 세포성장과 plasmid 유전자 발현을 온도에 의해 분리시키는 이단계 연속배양에서 plasmid 는 500 시간 이상 거의 100% 로 안정하게 유지되었으며, 산물인 tryptophan 의 생산성도 일단계 연속배양에서와는 달리 일정한 수준으로 안정화 되었다. 산물의 비생산속도 ($q_p$) 와 plasmid 함량 및 tryptophan synthetase (TSase) activity 는 낮은 비증식속도 ($μ_2$ = 0.011 $hr^{-1}$) 에서 가장 높았으며, 비증식속도가 증가할수록 감소하는 양상을 보였다. 또한, plasmid 유전자가 시간당 생성하는 산물의 생산성인 유전자의 발현효율성 (gene expression efficiency), $q_p/C_p$ (mg-trp/㎍-DNA.hr) 와 $E_p/C_p$ (U-TSase/㎍-DNA.hr), 은 비증식속도와 비례하여 증가하였다.
