From the $\underline{Zymomonas mobilis}$, having potential value for ethanol fermentation, the structural gene encoding alcohol dehydrogenase (ADH; EC 1.1.1.1) responsible for the final step during alcoholic fermentation was cloned into $\underline{Escherichia}$ $\underline{coli}$ with plasmic pUC9 using allyl alcohol.
Two recombinant plasmids were isolated from $\underline{E}$. $\underline{coli}$ transformants showing ADH activity, which were sensitive to allyl alcohol, named pADS93 and pADL99. These plasmids was shown to share a common $\underline{Z}$. $\underline{mobilis}$ chromosomal DNA of 2.6 kb.
Electrophoretical analysis of total cell extrats on a nondenaturing polyacrylamide gel indicated that the two clones, $\underline{E}$. $\underline{coli}$ (pADS93) and $\underline{E}$. $\underline{coli}$ (pADL99), produced the identical enzyme displaying a same band of activity comigrating with one (ZADH-2) of the two $\underline{Z}$. $\underline{mobilis}$ alcohol dehydrogenase isozymes (ZADH).
In addition, the enzyme from crude extracts of $\underline{E}$. $\underline{coli}$ (pADS93) was purified to compare with the ADH produced by $\underline{Z}$. $\underline{mobilis}$. SDS-polyacrylamide gel electrophoresis of these final preparations revealed that ZADH-2 subunit purified from $\underline{E}$. $\underline{coli}$ (pADS93) had a single band identical to the enzyme subunit from $\underline{Z}$. $\underline{mobilis}$ at 40,000 dalton of molecular weight. Analytical gel filtration of Sephadex G-200 led to conclusion that this enzyme is a tetramer with molecular mass 170,000 dalton.
Southern hybridization indicated that the structural gene (zadhII) for ZADH-2 was homologous between $\underline{Zymomonas}$ strains, though it did not have homology to that for other isozyme (ZADH 1).
The complete nucleotide sequence of the zadhII was determined. The zadhII consists of an open reading frame, 1152 bp long, commencing from the ATG start codon encoding a polypeptide of 383 amino acid residues and a TAA stop codon. The predicted amino acid sequence of the ZADH-2 enzyme agreed with the previously determined amino acid compostion (C. Wills, P. Kratofil, D. Londo, and T. Martin, Arch. Biochem. Biophys. 210: 775-785, 1981) and 49 amino acid sequence of N-terminal region of the purified enzyme except for three positions (A. D. Neale, R. K. Scopes, J. M. Kelly, and E. H. Richard, Eu. J. Biochem. 154: 119-124, 1986).
The codon utilization pattern in zadhII is shown to closely resemble that in highly expressed $\underline{E}$. $\underline{coli}$ gene. In addition, zadhII has a total G/C content of 50% while at the third base of the codon G/C content is 48.7%.
Upstream from the translation initiation codon of zadhII, a putative ribosomal binding site was identified. However, there were no sequences which could be idnetified as being homologous to the generalized promoter structure for $\underline{E}$. $\underline{coli}$. The nucleotide sequence following TAA stop codon was shown to include a 7 base pairs palindrome sequence presumed to constitute transcription terminator.
The $\underline{Z}$. $\underline{mobilis}$ gene in recombinant plasmids was found to express in $\underline{E}$. $\underline{coli}$ regardless its orientation to be transcribed from the lac promoter, indicating that $\underline{Zymomonas}$ gene could be transcribed and translated from $\underline{Z}$. $\underline{mobilis}$ control system which are recognized by the $\underline{E}$. $\underline{coli}$ transcriptional and translational system. It was also supported by the fact that the DNA fragment corresponding to 5' flanking region of zadhII made a foreign gene express in $\underline{E}$. $\underline{coli}$.
For the purpose of developing a plasmid vector suitable for transforming $\underline{Zymomonas}$ cells a 1.7kb cryptic plasmid has been isolated from $\underline{Z}$. $\underline{anaerobia}$, named pZA2. It was used to construct a shuttle vector by inserting useful parts of pUC9, pBR322, and pRK2501. $\underline{E}$. $\underline{coli}$ was employed to clone the new plasmid designated pSR12. The 7.7-kb plasmid pSR12 reisolated from the host cells could transform competent cells of $\underline{Z}$. $\underline{anaerobia}$ at $2×10^{-7}$ frequency. This shuttle vector contains two antibiotic resistance markers, kanamycin and tetracycline resistance genes, as well as restriction sites such as EcoRI, PstI, and XhoI, suitable for DNA recombinations.
근래에 알코올 발효 측면에서 많은 각광을 받고 있는 $\underline{Zymomonas}$ mobilis ATCC 10988 로 부터 알코올 탈수소 효소 유전자를 분리하였다. 이때 대장균을 숙주로, pUC9 플라스미드를 유전자 운반체로 사용하였는데 알코올 탈수소 효소 유전자에 의해 형질 전환이 일어난 대장균은 allyl alcohol 에 대해 민감해졌다. 이러한 현상은 알코올 탈수소 효소에 의해 allyl alcohol 이 산화되어 세포에 유독한 acrolein 으로 변환되기 때문에 일어난다.
대장균 형질 전환체들로 부터 두종류의 재조합 플라스미드를 분리 하였으며 이들을 각각 pADS93 과 pADL99 로 명명 하였다. 여러가지 제한효소를 사용하여 재조합 플라스미드를 절단하고 agarose gel 에서 전기영동을 실시한 다음 절단된 각 DNA 조각들의 분자량을 축정한 결과 pADS93 은 약 2.6 kb insert 를, pADL99 는 약 4.0 kb insert 를 함유하고 있는 것으로 나타났으며 또한 이들 insert 는 약2.6 kb 크기의 부분이 서로 동일 하였다.
이러한 재조합 플라스미드에 의해 형질 전환이 일어난 대장균의 세포 추출물과 $\underline{Z}$. $\underline{mobilis}$ 의 세포 추출물을 polyacrylamide gel 에서 전기영동을 실시한후 알코올 탈수소 효소 활성을 보이는 위치를 비교한 결과 두종류의 대장균 형질 전환체는 모두 동일한 효소를 생산하고 있으며 이것은 $\underline{Z}$. $\underline{mobilis}$ 에 존재하는 두종류의 알코올탈수소 효소 isozymes (ZADH) 중의 한가지인 ZADH-2 임이 밝혀졌다. 이 효소를 분리 정제하여 기질 특이성을 조사한 결과 대장균 형질전환체에서 분리된 효소와 $\underline{Z}$. $\underline{mobilis}$ 에서 분리된 효소가 동일한것으로 나타났다. 그리고 이효소를 SDS-polyacrylamide gel 에서 전기영동하여 subunit 의 분자량을 축정하고 Sephadex G-200 을사용하여 gel filtration 를 행한 결과 이효소는 약 40,000 의 분자량을 갖는 4개의 동일한 subunits 로 구성되어 있음이 밝혀졌다.
한편 분리된 알코올 탈수소 효소 유전자를 이용하여 여러 종류의 $\underline{Zymomonas}$ 균주로 부터 분리한 DNA 와 Southern hybridization 을실시 함으로써 각 균주들이 거의 동일한 유전자를 지니고 있으나 두종류의 알코올 탈수소 효소 isozymes 유전자간에는 서로 유사성이없음이 밝혀졌다.
pADS93 에서 분리한 이 유전자의 크기를 줄여 최종에 1.8kb 로 만든후 Sanger 등의 dideoxy chain termination 방법에 의해 염기배열 순서를 결정하고 그것의 특성을 살펴본 결과 다음과 같았다. Z.mobilis 의 알코올 탈수소 효소 유전자는 translation 개시 codon 인 ATG 로 부터 종결 codon 인 TAA 까지 1152 염기들로 구성되어있다. 염기배열 순서로 부터 예상되는 효소는 382 아미노산 잔기들로 (처음의 Methionine 잔기제외) 구성되며 그 분자량은 39,998 로 앞에서 구한 효소 subunit 의 분자량 40,000 과 일치 하였다. 그리고 예상되는 아미노산 조성도 분리된 효소의 것과 일치 하였다. 이 유전자의 codon 사용정도는 대장균에서 고도로 발현되는 유전자의 것과 유사하였으며 Guanine 과 Cytosine 의 함량은 50% 로 나타났다.
Translation 개시 codon 인 ATG 앞쪽에는 ribosome 결합 장소인 Shine-Dalgarno sequence 로 추정되는 GTGAGGT 가 위치해 있다. 한편 tanslation 종결 codon 인 TAA 뒤쪽에는 transcription 을 종결시키는 머리핀 구조를 이루는 7 base pairs palindrome sequence 가 존재하며 그곳에서 뒤쪽으로 12 bases 떨어진 곳에 4T 가 뒤따르는 것으로 보아 이 유전자의 transcription 은 rho-independent termination 되는 것으로 추정된다. 비록 ATG 앞쪽에 대장균 유전자에서 밝혀진 promoter 의 consensus sequence 와 동일한 염기배열을 갖는 지역이 없었으나 promoter 역할을 할 수 있는 것으로 예상되는 7 종류 sequences 가 발견 되었다. 이런 sequences 를 함유한 ATG 앞쪽 부분에 해당하는 DNA 조각을 분리하여 promoter 와 Shine-Dalgarno region 이 제거된 섬유소 분해 효소 유전자의 ATG앞쪽에 도입시킨 결과 대장균내에서 섬유소 분해 효소 유전자가 약50배 정도 증폭 발현됨을 알 수 있었다. 이로 미루어보아 $\underline{Z}$. $\underline{mobilis}$ 알코올 탈수소 효소의 translation 개시 codon ATG 앞쪽부분이 대장균내에서 promoter 활성도를 지는 것으로 사료된다.
또한 재조합 플라스미드에 존재하는 $\underline{Z}$. $\underline{mobilis}$ 유전자가 lacpromoter 의 transcription 방향과 관계없이 대장균내에서 발현된다. 그러므로 $\underline{Zymomonas}$ 유전자의 조절계는 대장균의 transcription 과 translation system 에 의해 인지될 수 있는 것으로 판단된다.
$\underline{Zymomonas}$ 세포를 형질 전환시키기 위해 쓰일 유전자 운반체를 개발하기 위해서 $\underline{Z}$. $\underline{anaerobia}$ 로 부터 1.7 kb 크기의 cryptic plasmid 를 분리하여 이것을 pZA2 라 명명하고 제한 효소 지도를 작성하였다. pZA2 플라스미드에 이미 널리 알려진 대장균의 유전자운반체인 pUC9, pBR322 그리고 pRK2501 로 부터 유용한 부분을 도입함으로써 4종류의 재조합 플라스미드를 만들고 이들을 각각 pSR12, pSR22, pSR32, pSR202 로 명명 하였다. 이들은 모두 tetracycline 과 kanamycin 내성 유전자와 pBR322 의 replicon 를함유하고 있다. 그리고 replicon 을 보호하기 위해서 서로 다른부위가 절단된 pZA2 플라스미드가 각각 존재한다.
이렇게 조작된 plasmids 를 이용하여 $\underline{Z}$. $\underline{anaerobia}$ 를 형질전환시켜 tetracycline 내성을 갖는 $\underline{Z}$. $\underline{anaerobia}$ 로 부터 plasmid 를 분리하여 여러가지 제한효소로 절단하여 조사한 결과 $\underline{Z}$. $\underline{anaerobia}$ 를형질 전환시킨 것이 pSR12 임이 판명 되었다. 따라서 새로 개발된 pSR12 는 대장균과 $\underline{Z}$. $\underline{anaerobia}$ 에서 모두 유지될 수 있는 shuttle vector 이며 또한 이것은 다른 유전자를 도입하기에 유용한 제한효소 절단 부위를 지니고 있다.
