Transition-metal catalyzed C-H functionalizations with directing groups have provided straightforward methods for the construction of carbon-carbon or carbon-heteroatom bonds. While strongly coordinating substrates have been widely functionalized, the use of weakly directing groups suffers from difficulty on the formation of the key metallacyclic intermediate. Especially, the utilization of ester as the directing group is still rare and its application on C-N bond formation have not been reported. This study describes the Ir(III)-catalyzed direct amidation on aryl and olefinic sp2 C-H bonds with organic azides using ester and ketone as viable directing groups under mild conditions.
In the mechanistic aspect, a Cp*Ir(III)-acetophenone metallacycle complex was isolated and characterized by X-ray crystallography. KIE experiments suggested that C-H bond cleavage is rate-determining step. Low-temperature 13C NMR study suggested that iridium monoacetate complex is the catalytically active species. The result of this work is significant considering that those directing moieties can be converted to various functional groups under ambient conditions.
탄소-수소 결합은 유기 분자의 기본 골격을 이루고 있는 만큼 이를 직접적으로 다른 작용기로 치환하는 반응은 유기 합성 화학에 있어 효과적인 도구를 제공할 것이다. 전이금속 촉매와 기질의 배위 유도 작용기를 이용하여 탄소-수소 결합을 탄소-탄소 혹은 탄소-헤테로원자 결합으로 치환하는 반응은 입체선택적이고 직관적인 탄소-수소 결합의 변형을 가능케 해 왔다. 하지만 강하게 배위하는 작용기들을 이용하여 탄소-수소 결합을 변형하는 방법은 활발히 보고되어 온 반면 카보닐 등 약한 유도 작용기들은 전이상태 및 중간체의 높은 에너지로 인해 그 이용이 많이 보고되어 있지 않다.
본 연구에서는 약한 유도 작용기인 케톤과 에스터를 이용하여 이리듐 촉매와 아지드 화함물로 위치 선택적인 sp2탄소-수소 결합의 아미드화 반응을 개발하였다. 본 탄소-질소 결합 형성 반응은 완만한 조건 하에서 이루어졌으며 방향족성 탄소-수소와 함께 올레핀 탄소-수소 결합 역시 아미드화시킬 수 있었다. 매커니즘적인 측면으로 케톤 배위 유도 작용기를 지닌 아세토페논과 촉매 사이의 고리형 중간체가 합성 및 분리되어 결정 구조가 분석되었다. 낮은 온도에서의 13C NMR 연구는 이리듐 모노아세테이트 화합물이 활성 촉매 형태임을 암시하였다.
본 연구에서의 완만한 조건과 낮은 온도, 유도 작용기로 사용된 케톤과 에스터 자체의 유기 합성 과정에서의 유용성 등을 고려할 때 본 아미드화 반응은 합성 화학에 있어 유용한 합성 방법을 제공할 수 있을 것으로 보인다.
