The abnormal regulation of serotonin (5-HT) and dopamine (DA) in the brain has been implicated in neuropsychiatric disorders associated with impairment in empathy. The anterior cingulate cortex (ACC) is known to be centrally associated with empathy. However, the relationship between 5-HT&DA systems in the ACC and empathic behavior has been unknown. We investigated the role of 5-HT&DA in empathy-like behav-ior and in the regulation of ACC neural activity. Observational fear conditioning (OFC) was conducted following microinjection of 5-HT, DA, 5-HT&DA together, methysergide (5-HT receptor antagonist), SCH-23390 (DA D1 receptor antagonist), or haloperidol (DA D2 receptor antagonist) into the mouse ACC. The ACC neural activity influenced by 5-HT and DA was characterized by in vitro and in vivo electrophysiology. The microinjection of haloperidol decreased the fear response of observing mice, demonstrating that DA D2 receptors in the ACC are required for OFC. The administration of 5-HT, but not DA, reduced the freezing response of observing mice, suggesting that increased 5-HT levels could disrupt OFC. 5-HT also enhanced delta-band activity and reduced alpha and gamma-band activities in the ACC, whereas DA reduced only alpha-band activity. In addition, only 5-HT reduced the complexity of ACC neural activity in vivo, suggesting that increased 5-HT levels could alter the regularity of ACC neural oscillations.
뇌 내 세로토닌과 도파민의 비정상적 조절은 공감능력의 문제와 관련된 각종 정신신경 질환들과 연루되어있으며, 전대상피질 (Anterior cingulate cortex)은 공감능력에 매우 중요한 역할을 하는 것으로 알려져 있다. 하지만 전대상피질 내의 세로토닌과 도파민 시스템들이 공감관련 행동에 어떠한 역할을 하는지는 연구되지 않았다. 이 연구에서 우리는 전대상피질의 세로토닌과 도파민이 공감관련 행동과 전대상피질의 신경활동에 어떠한 영향을 미치는지 알아보았다. 그러기 위해 세로토닌, 도파민, methysergide (세로토닌 수용체 길항약), SCH-23390 (도파민 D1 수용체 길항약), haloperidol (도파민 D2 수용체 길항약)을 생쥐의 전대상피질에 주입한 후 관찰적 공포학습을 행했다. 또한 체내 및 체외 전기생리학으로 세로토닌과 도파민이 전대상피질의 신경활동에 어떠한 영향을 미치는지 관찰하였다. Haloperidol을 주입할 경우 공포 반응이 감소하였으며, 이는 전대상피질의 도파민 D2 수용체가 관찰적 공포에 필요하다는 것을 암시한다. 또한 세로토닌을 주입할 경우 공포 반응이 감소하였는데, 이는 전대상피질에서 증가된 세로토닌이 관찰적 공포학습을 방해한다는 것을 시사한다. 하지만 도파민 주입은 관찰적 공포학습에 영향을 주지 않았다. 또한 도파민이 체내에서 전대상피질의 알파 파워만을 감소시킨 데에 반해, 세로토닌은 델타 파워를 증가시켰으며 알파와 감마 파워를 감소시켰다. 그리고 세로토닌만이 전대상피질 신경활동의 복잡성을 감소시켰다. 이는 증가된 세로토닌이 전대상피질의 신경활동을 변화시킬 수 있다는 것을 암시한다.
