The interaction between phosphatidylserine multilamellar vesicle (PS MLV) and synthetic luteinizing hormone releasing hormone (LHRH) was investigated by intrinsic fluorescence and centrifugal separation of PS MLV-LHRH complexes. PS MLV induced a blue-shift in the wavelength of maximum emission of fluorescence of LHRH by about 7nm㎚.
The influence of PS MLV on the quenching of the tryptophan in LHRH by sodium iodide (NaI) was studied. PS MLV was found to protect LHRH from the iodide quenching.
The conformation of LHRH was investigated by circular dichroic measurements. The Greenfield-Fasman and the Chang-Wu-Yang methods were used for the analysis of CD spectra. It was found that LHRH has appreciable β-structure even in aqueous solution. The β-structure content increased significantly upon binding to amphihpilic surface.
LHRH의 고유형광 발산의 관측과 원심분리법을 이용하여 LHRH와 PS MLV의 상호작용을 연구하였다.
PS MLV는 LHRH의 최대형광 발산파장을 약 7 nm 단파장 이동시켰고, LHR-H와 PS MLV 간에는 소수성 작용과 정전기적 작용이 존재하였다.
막에 결합된 LHRH 가 Sodium iodide (NaI)에 의한 Trp-quenching으로부터 보호되었는데, 이는 막에서의 구체적인 topology 연구에 기초가 되었다.
마지막으로 CD의 연구를 통하여 LHRH가 세포막에 접근하면서 어떤 구조로 변하는가를 예측하였다. 친수성 용액에서 LHRH는 약간 변형된 β-turn을 갖는 유동적인 β-structure를 형성하는 것 같다. SDS 용액에서는 β-turn이 풀리면서 β-pleated sheet 구조를 형성한 채, micelle 표면에 평행하게 결합하는 것 같다.
