Batch experimental investigation was performed on enzyme immobilization using emulsion liquid membrane. Hydrolysis reaction of L-phenylalanine methyl ester to L-phenylalanine was selected as a model system.
The effect of experimental variables, such as internal phase enzyme concentration, carrier concentration and type, phase volume ratio, stirring speed, surfactant concentration, external and internal phase pH, and external phase substrate concentration was analyzed.
Enzyme reaction rate in aqueous solution was fast at pH 7, whereas enzyme immobilized in emulsion liquid membrane was fast at internal phase pH 6.
Optimum conditions obtained from the experiment, are as follows: Internal phase enzyme concentration is 0.4g/l. Stirring speed is 300rpm. pH in the internal and external phase are 6 and 8. Surfactant concentration is 10wt%.
