In this thesis, studies of Clostridium botulinum type B toxin were approached by the three sequential but methodologically different steps. For the purification of Clostridium botulinum type B toxin of strain Lamanna, four column chromatography works including DEAE-Sephadex, Sephadex G-200 and Sephacryl S-300 were performed. The finally obtained toxin was the mixtures of nicked and unnicked form of toxin. The molecular weight of whole toxin, heavy chain and light chain of the toxin were 142 kDa, 98 kDa and 50 kDa, respectively on the SDS-PAGE.
Four monoclonal antibodies were prepared and characterized using the purified toxin as an immunogen. The isotypes of the monoclonal antibodies were Ig M, Ig Gl, Ig A, Ig Gl, respectively. One of them showed high neutralizing activity and all of them were bound to the heavy chain of toxin. 1 : 1000 dilution of ascites fluid which was made from the one of the hybridoma cell lines was not bound to the type A toxin and could detect 0.1 ng of type B toxin on the dot immuno-binding assay.
Using a commercial $\lambda$gt 11 packaging kit, a genomic library of the whole cellular DNA of C. botulinum was made. Two $\lambda$gt 11 clones of the toxin gene were identified from the library using the monoclonal antibody specific to the heavy chain of type B toxin. Neither of the expressed fusion protein from the lysogenic E. coli Y1089 showed any botulinal toxic activity. One of the clones hybridized to the oligonucleotide probe which was synthesized according to the amino acid sequence of N-terminus of heavy chain of C. botulinum type B. A DNA fragment (HindIII - XbaI) hybridized with the oligonucleotide probe was isolated and subcloned for the sequencing. The sequence analysis revealed that highly homologous regions exist in N-terminus of heavy chain among botulinum neurotoxins(type A, B) and tetanus toxin on the amino acid sequence level. This means that the three toxin genes are might derived from a common ancestral gene whose function in prokaryotic cell is still unknown.
$\underline{Clostridium}$ $\underline{botulinum}$ type B Lamanna 균주에서 분비되는 신경독소를 세 종류, 네번의 크로마토그라피 방법으로 정제하였다. 마지막 정제 후에 SDS - polyacrylamide gel 에서 나타난 분자량은 전체 독소가 142 kDa, 큰 하위 단위체가 98 kDa, 그리고 작은 하위 단위체가 50 kDa 인 것으로 나타났다. 한편, 정제된 독소는 끊어진 형태와 끊어지지 않은 형태가 함께 공존하였다.
분리된 독소를 톡소이드로 만든다음 항원으로 사용하였을때, 면역된 생쥐(Balb/c)의 비장세포와 myeloma 세포를 세포융합 시킴으로써 독소에 대한 단일클론 항체를 분비하는 세포주를 모두 네 종류 얻었다. 이들이 분비하는 단일클론 항체를 분석한 결과, 이들 모두는 type B 독소의 큰 하위 단위체에 붙는 특성을 가지고 있음을 알았고, 그 중 하나는 (BTBM 3) 독소 중화 능력을 갖고있는 것으로 보였다. 또, 다른 한 종류의 단일클론항체 (BTBM 4)를 type A 독소와 결합시켜 보았을 때 전혀 붙지 않았고, 이 단일클론 항체는 $10^{-1}$ ng 수준의 독소를 검출할 수 있는 것으로 나타났다.
한편, $\underline{Clostridium}$ $\underline{botulinum}$ 전체 DNA를 분리하여 λgt 11 을 이용한 genomic library를 만든 다음, 앞서 만들어진 한 종류의 단일클론 항체 (BTBM 4)를 이용해서 immunoscreening을 수행했을 때 두개의 DNA 클론을 찾을 수 있었다. 그 중 한 개의 클론이 $\underline{C}$. $\underline{botulinum}$ type B 독소의 큰 하위 단위체의 아미노 말단으로부터 만들어진 oligonucleotide와 결합함을 Southern blotting 실험을 통해 알아냈다. 한편, 이들 두 λgt 11 클론을 대장균 Y1089 균주에 lysogenization 시켰을 때 만들어지는 β-galactosidase 연결 단백질들은 어떤 독성도 나타내지 않음을 생쥐를 이용한 실험으로부터 확인 하였다.
$\underline{C}$. $\underline{botulinum}$ 독소의 큰 하위 단위체의 아미노 말단에 해당되는 부위의 염기 서열을 분석해 보았을 때, 적어도 이 부분에서는 $\underline{C}$. $\underline{botulinum}$ type A 및 $\underline{C}$. $\underline{tetani}$의 큰 하위 단위체의 아미노 말단과 상당히 유사한 부위가 있는 것으로 판명되었다. 이 결과는 이들 세 종류의 유전자가 하나의 유전자로부터 진화되어 왔음을 강력하게 시사 해준다.
