Glyoxalase 1 (GLO1) is a ubiquitous cellular enzyme in the Glyoxalase system, a metabolic pathway that detoxifies methylglyoxal (MG). Methylglyoxal, a cytotoxic byproduct of glycolysis, is formed by degradation of the glycolytic intermediates dihydroxyacetone, phosphate, and glyceraldehyde-3-phosphate. This byproduct induces oxidative stress, apoptosis, nucleotide modification, and protein modification, forming advanced glycation end products (AGEs). To alleviate MG’s cytotoxic effects, GLO1 uses glutathione as a cofactor to enzymatically convert MG into d-lactate, which is a less reactive substance. In addition, recent findings have linked GLO1 to psychiatric diseases, such as, depression, schizophrenia, and autism. In the research for this paper, knockout GLO1 mice were generated and used to perform various characterization tests. The GLO1 knockout mice showed reduced levels of MG compared to wild-type, heterozygous mice. Moreover, GLO1 knockout mice showed an absence of GLO1 activity, decreased level of lactates, and higher levels of AGE formation. Finally, we showed that there is a positive correlation between GLO1 and anxiety-like behavior via two behavior tests: Open field and Light-dark box test.
Glyoxalase 1 (GLO1)는 Glyoxalase 시스템에 관여하는 효소로서 methylglyoxal (MG)의 세포 효소이다. Glycolysis 과정중 생성되는 부산물인 Methylglyoxal은 degradation of the glycolytic intermediates dihydroxyacetone, phosphate, 그리고 glyceraldehyde-3-phosphate 에 의해 형성된다. 이러한 부산물은 산화 스트레스, 세포 사멸, 뉴클레오티드 수정 및 단백질 변형을 유도한다. MG의 세포 독성 효과를 완화하기 위해, GLO1는 glutathione을 cofactor로 사용하여 효소 반응성이 적은 물질인 D-lactate로 변환 시켜 준다. 또한, 최근의 연구 결과는 우울증, 정신 분열증, 자폐증과 같은 정신 질환에 GLO1이 연관 되고 있다고 설명하고 있다. 이 논문의 연구에서는 GLO1 결손의 분석 연구를 하였다. 먼저 PCR, western blot 그리고 activity assay를 통하여 GLO1의 결손이 되었는지를 확인 하였다. 이어서 GLO1이 결손 됨으로써 변화하는 lactate와 MG의 양을 측정한 결과 lactate는 감소, MG는 증가하는 것을 확인 하였다. 또한 MG의 증가로 인해 MG에 의한 단백질이 변형된 AGEs를 MG-H1 antibody로 본 결과 AGEs또한 증가한 것을 확인 하였다. GLO1과 연관 되고 있다는 anxiety-like behavior를 open field 와 light-dark box test, 두 가지 행동 실험을 바탕으로 분석한 결과 GLO1과 anxiety-like behavior는 positive correlation이다 라는 결과를 얻을 수 있었다.
