The recent advancement of microfluidics has demonstrated its superior performance in fields of genomic, proteomic and cellular analysis. In particular, the fine controllability of microfluidics in the microenvironments enables us to investigate cellular responses and physiology, which cannot be approached by conventional methods. In this thesis, novel isotropic shaped microstructures were developed to be used for in vivo like cellular alignment for tissue engineering and single cell array such as single cell capture, cultivation and retrieval to replace the tedious and time-consuming agar plate based colony selection process.
Vascular vessels play an important role in maintaining the homeostasis of the circulatory system, and they are involved in a number of vascular diseases such as atherosclerosis and thrombosis. Microfluidic-based biomimetic devices have demonstrated more reliable and high-throughput drug screening capability compared with the conventional static 2-D cell culture system. Research on the construction of an artificial microvascular system on a chip is one of the extensively focused areas. A study on the cardiovascular disease-related functions, and assessment of vascular toxicity in drug screening requires realistic in vitro vascular models. To this end, in chapter 2, a simple and efficient fabrication method for generating isotropic microfluidic channels with a circular-cross sectional geometry was developed by exploiting the reflow phenomenon to construct three dimensional endothelial cell layer. To form a three dimensional endothelial cell layer inside circular PDMS microchannels, we cultured primary human umbilical vein endothelial cells (HUVECs), and demonstrated successful cell adhesion, proliferation, and alignment along the channel.
In chapter 3, the nanowrinkle as well as hierarchical nano/microwrinkle patterns on a PDMS substrate were developed by the strain responsive wrinkling method. The effect of the UV/O exposure time and the stretching rate of the PDMS substrate on the wavelength and the amplitude of the wrinkle patterns were investigated. Furthermore, we examined the human aortic smooth muscle cell (HASMCs) alignment on the topographical patterned surface, and found that more than 80% of the HASMCs were cultured and aligned well along with the hierarchical nano/microwrinkle rather than the nanowrinkle pattern.
In chapter 4, isotropic circular microfluidic channels were combined with the microwrinkle patterns to form an in vivo-like circumferential smooth muscle cell alignment. To construct a biomimetic smooth muscle cell layer which is aligned perpendicular to the axis of blood vessel, the orthogonally microwrinkle patterns are generated inside the half-circular microchannel by a strain responsive wrinkling method and the HASMCs are cultured circumferentially inside the channels with high alignment. A circumferential alignment of HASMC means that the creation of an in vivo-like 3D smooth muscle cell layer in the circular microfluidic channel which can provide a bioassay platforms for in-depth study of HASMC biology and vascular function.
Bioenergy which uses biological sources such as biomass for producing fuel has been considered promising. C. reinhardtii is the most widely used strain as a source of bioenergy such as hydrogen and biodiesel. To obtain such a superlative microalgae, many biologists prepared genetically engineered C. reinhardtii libraries, and screened them to isolate a promising strain. However, the conventional process for genetic modification including electroporation or a bead beating method suffers from low efficiency, and the strain selection based on the solid phase agar plating needs a prolonged time. In particular, the agar plated colony formation requires more than 2 months depending on the microalgal strain, which could be a bottleneck for development of biomass based energy production. Since the whole process is carried out on the solid phase agar plate, the diffusion rate of nutrient and antibiotics to the cells was very limited, resulting in low efficiency for isolating genetically modified C. reinhardtii. To substitute the conventional process of agar plating colony formation and retrieval, the microdevice which can handle single cells should be required. To address this issue, I, first, developed a large-area 3D hemispherical perforated isotropic microwell structure for a single bead based bioassay in chapter 5. Such a unique microstructure enables us to perform the rapid and stable localization of the beads at the single bead level and the facile manipulation of the bead capture and retrieval with high speed and efficiency. The streptavidin-coated microbead capture was achieved with almost 100 % yield within 1 min, and all the beads were retrieved in 10 sec. To demonstrate the proof-of-concept of the aptamer screening on the proposed bead microarray, lysozyme or thrombin binding aptamer labelled microbeads were trapped, the in situ fluorescence signal of the bead array was monitored after aptamer-target protein interaction, the protein-aptamer conjugated microbeads were recovered, and the aptamer was isolated for matrix assisted laser desorption/ionization time-of-flight mass spectrometry analysis to confirm the identity of the aptamer.
In chapter 6, a large-area 3D hemispherical perforated isotropic microwell structure was developed for efficient capture of C. reinhartii at single cell level to isolate hygromycin B-resistance mutant. The rapidly moving microalgal cells (wild type and hygromycin B-resistance mutant) loaded in the top layer moved into the microwell holes and captured at the single cell level with high efficiency. Then, subsequently the single cell based cultivation in a hygromycin B containing medium was performed to generate a colony in the 3D hemispherical microwell structure. While the wild type suffered from cell death, hygromycin B-resistance mutant were tolerated and well grown to form a colony in 2 days. The produced colonies in the microwells were recovered by applying a release mode in the bottom layer, so that the hydrodynamic force was exerted vertically to push out the colonies to the outlet in 10 sec. The recovered cells were cultured in a large scale medium in a flask. The recovered C. reinhartii was confirmed as a hygromycin B-resistance mutant by identifying the hygromycin gene in the colony polymerase chain reaction. Thus, Our proposed microdevice could perform solution phase based single cell capture, colony formation, and retrieval of colonies for further large scale cultivation, which could replace tedious and time-consuming solid phase agar plate processes with 7-fold time reduction.
1. 방향성 세포 배양을 위한 등방형 미세 유체 채널 소자 연구
분자 진단 또는 약물 스크리닝을 위해서는 생체내의 환경을 그대로 보존한 생체 외 분석 시스템이 필요하다. 본 연구에서는 나노/마이크로 기술을 이용한 고효율 생체 모방 혈관 시스템을 제작하고 혈관과 동일 조건에서의 3D cell based assay를 구현함으로써 HTS 스크리닝의 새로운 패러다임 분석 시스템을 제공하고자 한다. 미세유체 채널 내 혈관 세포를 배양하기 위해서는 미세유체 채널을 혈관과 유사하게 둥글게 만드는 과정이 필수적이다. 즉 수십~수백 um 구경의 마이크로 채널을 template로 하여 내부에 extracellular matrix (ECM) 또는 생체 모사 나노/마이크로 구조체를 구현하고 이를 이용하여 혈관을 구성하는 근육세포와 내피세포를 채널 벽면에 전체적으로 배양한 후 실제로 생체 분석을 통해 생체 내에서 일어나는 현상과 동일한지를 비교하는 것을 목표로 하였다.
Si 웨이퍼 기판위에 photolithography, computer numerical control (CNC) 가공, 웨이퍼 wet 에칭 기술(질산 + 불산)을 이용하여 각종 기판 위에 원하는 직경을 가진 반원의 둥근 유체 채널을 형성한 후, PDMS 폴리머를 Si 웨이퍼에 붓고 경화시킨 후 떼어내면 PDMS 폴리머 위에 양각 형태의 구조체를 지니고 있는 주형을 만들 수 있다. 이 주형을 위에 다시 PDMS를 붓고 경화를 시킨 후 떼어내면 PDMS 기판위에 초기에 가공된 기판에 형성된 음각 반원 미세채널이 형성된다. 채널 내부 표면에 선택적으로 일정한 시간의 UV/O 조사를 통하여 폴리머 표면에 채널의 수직방향으로 나노 또는 마이크로 단위의 주름형태의 패턴을 형성할 수 있다. 앞서 만들어진 두개의 PDMS 채널을 결합하여 하나의 실린더 형태의 마이크로 플루이딕 채널을 형성할 수 있고, 둥근 채널이 만들어진 이후 실제 혈관 (정맥 또는 동맥) 내부에 들어 있는 근육세포와 내피세포 및 각종 세포 외 기질들을 형성하기 위해 우선 세포 외 기질 중 하나인 단백질 (fibronection 또는 collagen) 을 채널 내부에 균일하게 코팅하여 세포가 채널 벽면에 잘 붙을 수 있도록 유도한 후, 마이크로 채널에 세포 외 기질이 코팅이 되면 근육세포를 넣고 배양하여 채널 벽면에서 성장 및 배열이 될 수 있게 유도한다. 실제로 혈관 속의 근육세포는 혈관의 수직방향으로 배향이 되어 있기 때문에 생체 외에서도 같은 형태로 배향을 유도하는 것이 중요하며, 앞서 본 연구에서는 UV/O 조사를 통하여 나노/마이크로 단위의 주름을 채널의 수직방향으로 벽면에 만들었으며, 미세유체 채널 내부에 근육세포가 배양됨에 따라 주름의 방향으로 즉 채널의 수직방향으로 배향이 되는 것을 확인 할 수 있었다. 또한 근육세포가 둥근 채널 전체를 둘러 쌓아 3D 구조 형태의 혈관과 비슷한 형태를 제작할 수 있었다. 실제로 세포의 필라멘트 엑틴, 핵 등의 종횡비를 측정하였고 세포 내에서 발현되는 단백질을 분석하여 실제 혈관과 유사함을 확인 할 수 있었다. 3D cell monolayer는 일반적인 2D system과는 달리 생체 내와 유사한 세포 환경을 제공해 줌으로써 그 세포 반응 데이터의 신뢰성을 높일 수 있다. 이후 내피세포를 근육세포 위에서 배양이 되도록 조건을 잡아 실험을 진행하였다. 내피세포는 혈액과 직접 맞닿는 부분이기 때문에 혈액의 흐름에 의해 내피세포는 근육세포와는 달리 혈관방향으로의 성장을 하며 배향이 된다. 본 연구에서는 미세유체 채널의 유체의 흐름을 조절을 통해 내피세포를 채널 방향으로 배향이 될 수 있도록 유도할 수 있었다. 둥근 단면을 지닌 미세유체 채널을 만든 후에 각종 혈관에 필요한 세포 및 세포 외 기질 등을 마이크로 채널 벽면에 부착시켜 최대한 생체 내와 같은 환경의 3D 배양환경을 제작하여 현재 2D 환경기반의 플라스크에서 수행되는 세포 분석기술의 단점을 최대한 극복하고 생채 내부에서 일어나는 반응과 최대한 근접한 데이터를 확보할 수 있는 연구를 진행하였다.
2. 단일 세포 또는 비드 포획 및 분석을 위한 마이크로 디바이스 제작
최근 미세조류와 같은 바이오매스를 이용한 에너지로의 전환은 향후 미래에너지의 대체 기술로 각광을 받고 있으며, 고효율의 바이오디젤과 같은 에너지원 생산을 위해서 최적의 미세조류 종 및 형질전환에 많은 연구가 집중되고 있다. 현재 형질전환 미세조류 분리 방법에 있어, 화학적 처리에 의한 random mutation 또는 전기화학적 방법에 의한 유전자 전달이 시도 되어 오고 있으며, 유전자 조작이 이뤄진 미세조류는 아가플레이트 상에서 콜로니 생성과정을 거친 후, 배양 최적화 단계 통해 우량종 미세조류를 발굴하고, 우량 미세조류 특성에 기인한 유전자 분석을 수행하고 있다. 본 연구에서는 Photolithography 기술을 이용하여 Flexible한 폴리머 기판에서 단일 미세 조류를 포획할 수 있는 마이크로 우물 어레이 구조체 층을 제작하고 어레이 위 아래로 미세유체 채널을 연동하여 단일 미세조류가 유체 흐름에 따라 마이크로 우물에 포획되고 장시간 실기간으로 단일 세포를 분석 할 수 있는 디바이스를 제작하였다. 단일 세포 포획 우물 구조는 위단은 60 μm 직경 홀을, 아랫단은 5 μm 직경 홀 구조를 가짐으로써, 단일세포가 갇힐 수 있도록 디자인되어 있으며, 아래 홀의 유체 흐름 방향 제어를 통해 단일 미세조류가 안정적으로 포획 되는 것을 확인 할 수 있었다. 실제로 전기화학적 방법에 의해 항생 내성 유전자 삽입된 형질 전환종을 이용하여 단일세포 포획 디바이스에 주입하고 단일 세포 단위로 포획하여 장시간 실시간으로 항생제가 포함된 배지를 동시에 주입하여 단일 야생 미세조류와 비교 분석하였다. 야생종의 경우 12 시간 이내 모든 세포가 세포사멸을 겪는 것을 확인 할 수 있었으나 항생 내성 형질 전환종의 경우 48 시간 이후 까지도 단일 세포가 콜로니 형태로 안정적으로 성장하는 것을 확인 할 수 있었다. 실제 아가플레이트에서의 콜로니 성장 후 콜로니를 걸러내는 과정은 짧게는 2주에서 길게는 2달이 걸리나 본 마이크로 디바이스에서는 액상에서 콜로니 성장이 가능하여 콜로니를 빠르게 성장시키고 걸러내는 데까지 2일 이내에 진행 할 수 있었다. 이런 단일세포 포획 분석 디바이스는 미세조류뿐만 아니라 혈중 종암세포 포획 및 분석에도 유용하게 사용될 수 있을 것으로 판단된다. 또한 위 아래 관통되어 있는 마이크로 디바이스를 이용하여 위와 같은 방법에 의해 마이크로 비드를 단일 단위로 포획을 시도하였다. 유체의 흐름에 따라 비드는 고효율로 관통형 우물에 포획이 가능하였다. 비드표면에 스트렙타비딘-바이오틴-압타머의 화학적 결합된 비드를 포획하고 미세유체 채널로 단백질 용액을 흘려주어 목적 단백질을 압타머에 의해 검출하는 실험을 실시하였으며, 형광으로 염색된 단백질과 비드 내 결합된 압타머의 상호작용에 의해 단백질이 우물 내 포획된 비드 표면에 부착하는 것을 형광 분석을 통해 확인 할 수 있었다. 포획된 비드는 아래쪽으로부터의 유체 역흐름에 의해 분리 될 수 있으며, 분리된 비드를 얻은 후에는 MALDI-TOF MS 분석을 통해 압타머를 역으로 추적/분석하는 연구도 진행하였다. 본 연구에서 제안된 관통형 우물을 지니는 마이크로 디바이스는 세포뿐만 아니라 비드 분석도 가능하여 실시간/고효율로 빠른 단백질, DNA, 세포 스크리닝에 널리 활용될 것으로 판단된다.
