Plant senescence constitutes an essential developmental process of life cycle that relocates nutrients from old or less efficient parts to newly developing or storage organs. Senescence-associated genes are activated by several transcription factors including ORE1. Phytochromes have been implicated to perceive environmental light conditions to inhibit senescence; however, a precise signaling mechanism remains elusive. Here I show that in Arabidopsis, two basic helix-loop-helix TFs, PHYTOCHROME-INTERACTING FACTOR 4 (PIF4) and PIF5, promote senescence and that phytochrome B delays senescence by repressing PIFs activities. PIFs activate ORE1 expression by forming multiple coherent feed-forward loops involving EIN3 and Group A bZIP TFs. PIFs activate the expression of not only ORE1 but also EIN3 and bZIP genes including ABI5 and EEL through binding their promoters. The bZIPs and EIN3 also bind to ORE1 promoter and activate its expression. Such coherent feed-forward loops have been suggested to have the signaling process more robust, making PIF4 and PIF5 act as key regulators of senescence in response to the changing light conditions.
As an independent study, I also investigate the crosstalk of light and ethylene signaling pathways in young seedling. During the first several days after seed germination, Arabidopsis seedlings are highly sensitive to the environmental cues, and show a surprising flexibility in their developmental adaptation. I found that dark-grown seedling mutants of PIFs and EIN3 have a number of common phenotypes such as open apical hook/cotyledon and photobleaching. Based on the central transcription factor positions of PIFs and EIN3 in light and ethylene signaling, I hypothesized that the two different signaling pathways would share certain downstream target genes to regulate the similar phenotypes. Bioinformatic and genetic analysis revealed that HOOKLESS1 (HLS1) is a representative shared downstream gene which functions for the phenotypes. The study revealed a representative model showing that independently triggered signaling pathways converge into a common effector gene.
장기간의 광부족은 빛을 받지 못하는 식물의 부위에서 노화를 유도하는데 이는 광합성을 제대로 하지 못해 기능성이 떨어지는 잎을 사멸시켜 질소와 같은 중요한 영양성분을 재활용하여 효율적으로 기능하는 다른 잎에 공급하기 위함이다. 이 과정에서 식물의 광수용체인 파이토크롬이 노화를 억제한다고 알려져 있다. 하지만 파이토크롬이 어떻게 노화를 억제하는 지에 대한 기작은 알지 못한다. 본 연구에서는 파이토크롬의 하위 신호전달자인 애기장대 PIF4와 PIF5가 암흑 상태에서의 노화를 촉진시키는 중요한 역할을 함을 밝혀내었다. 암흑 상태에서 파이토크롬 B는 비활성화되는데, 이는 곧 PIF4와 PIF5 단백질의 축적을 야기한다. 생물정보학적 분자생물학적 분석을 통해, PIF가 두 종류의 호르몬 신호전달 매개 전사인자의 발현을 직접 증가시키는 것과 증가된 해당 전사인자들이 PIF와 함께 다시 또 다른 목표인 ORESARA1 유전자의 프로모터에 직접 결합하여 해당 유전자 발현을 촉진시킨다는 것을 발견하였다. 또한, pif4와 pif5 돌연변이체의 자연적 노화 속도 또한 감소되어있는 것을 관찰하여, 두 PIF가 광학적, 발생학적 배경에서 노화를 조절하는 중요 조절 인자임을 밝혔다.
독립적인 연구로서, 암흑상태의 애기장대 묘종 발달에 있어서 빛 신호전달 경로와 에틸렌 신호전달 경로 사이의 교섭을 탐구하였다. 종자가 발아한 이 후 몇 일 동안, 애기장대 묘종은 환경 신호들에 매우 민감한 반응을 보이는데 이를 통하여 놀랍도록 유연한 발달 적응을 한다. 이러한 묘종 발달 과정 중에 pifq와 에틸렌 돌연변이체들 사이의 강한 유사성을 발견함으로써, 빛 신호 전달계와 에틸렌 신호 전달계 사이의 공유된 신호 전달 요소들이 존재할 것이라는 가설을 세웠다. 생물정보학적, 유전학적 분석을 통하여서 HLS1이라는 유전자가 대표적으로 두 신호 전달계에 의하여 조절 받는다는 것과 해당 유전자가 암흑상태의 묘종 발달에 중요한 역할을 한다는 것을 밝혔다. 본 연구는 서로 독립적인 신호전달 체계가 공통된 하위 유전자들로 귀결되어 식물 생리를 조절하는 신호전달모델을 제시하고 있다.
