As an alternative for energy intensive aerobic wastewater treatment system, development of sulfate reducing bacteria (SRB)-based anaerobic domestic wastewater treatment system was studied. This proposed energy conservative wastewater treatment system has main advantageous factors of no requirement of aeration, fast substrate utilization and less sensitiveness of SRB. Sulfate for SRB’s synthesis could be supplied from SOx in industrial gas, thus the system also can contribute to the economical treatment of hazardous sulfur compound in the gas. With the dissolved sulfate as an electron acceptor, SRB degrade organic matters in wastewater and produce sulfide in continuously operating upflow anaerobic sludge blanket reactor (UASBr). Also the produced sulfide is used as an electron donor for electricity generation in sulfide fuel cell system connected to the UASB reactor.
This study focuses on obtaining SRB dominant granule from methanogen dominant anaerobic granule in a competition between SRB and methanogens. As a strategy to increase SRB activity and to suppress methanogens, starvation was applied by not providing organic source but continuous supplying sulfate. Based on the well adopted hypothesis on the granule structure, methanogens located relatively inner core of a granule died out due to the lack of organic source, while SRB located at outer part of granule survived utilizing extracellular polymeric substance (EPS) or organic matters from died out microbes as an organic source. By eliminating the competition, the portion of organic matter degradation by SRB was increased from 19% before starvation to 93% after starvation, achieving 80% COD removal in total. In case of sulfate removal, 20% improvement was achieved compared to the control reactor. This effect of starvation on the granule characteristics were confirmed with scanning electron microscope (SEM) and transmission electron microscope (TEM). In addition, increased population of SRB and decreased methanogenic population was also observed with next-generation sequencing (NGS).
Not only wastewater treatment by SRB but also electricity generation with sulfide fuel cell system was studied. Several sets of batch tests were conducted and continuous operation was also carried out with UASBr effluent containing 80mg/L - 100mg/L biologically produced sulfide. Through the batch tests, hydraulic retention time (HRT) was optimized to 8 h, removing almost all of the sulfide from the effluent, having the highest power density of 0.033 mW/cm2. With the optimized HRT, sulfide fuel cell was connected to the UASBr and continuously operated for over 50 days with successive electricity generation with biologically produced sulfide. This newly developed wastewater treatment system with SRB integrated with electricity generating sulfide fuel cell system is believed to be a promising technology for more sustainable wastewater treatment systems.
국내 기존 하수는, 빠른 처리속도와 높은 처리효율을 기반으로 하는 호기성 시스템을 통해서 처리 되어 왔다. 그러나 폭기 과정의 과다한 전력 소모와 다량의 슬러지 발생으로 인한 슬러지 처리 문제에 따른 새로운 하수처리 공정의 개발이 요구되고 있다. 따라서 본 연구에서는 혐기성 미생물인 황 환원균이 우점된 슬러지를 이용 도시 하수를 처리함과 동시에, 생산된 황화수소에 황 연료전지를 적용하여 전기를 생산하는, 신개념 하수처리 시스템 구축을 위한 공정을 개발하였다. 본 연구에서 제안된 공정은 혐기성 하수처리 시스템으로 폭기가 요구되지 않고, 잉여슬러지의 발생량이 적다는 점과, 타 혐기성 공정과 비교하여 유기물 분해 속도가 빠르다는 장점을 이용하여 기존 하수처리 시스템의 단점을 보완 할 수 있을 것으로 사료된다.
본 연구에서는 혐기성 그래뉼 슬러지를 이용하여 황 환원균을 우점화 하기 위한 방법으로 Starvation을 적용하였다. 이는 연속 운전을 하면서 Sulfate는 지속적으로 주입해 주고 유기물의 공급은 중단하는 방법이다. Starvation은 메탄균과 황 환원균의 경쟁관계에서 황 환원균을 우세화 하기 위하여 혐기 그래뉼의 구조적 특성을 이용한 것으로, 유기물의 공급을 중단 함으로써 그래뉼 가장 내부에 위치하는 메탄균은 충분한 유기물의 공급을 받지 못해 활성을 잃게 된다. 반면 비교적 그래뉼 외곽에 위치한 황 환원균은 지속적으로 공급되는 Sulfate를 이용하고, 그래뉼에 존재하는 Extracellular polymeric substances (EPS)와 분해된 미생물을 유기물로 이용하여 생존하기 유리 할 것으로 예상된다. 이것은 황 환원균이 메탄균보다 기질 친화력이 높고 유기물 분해속도가 빠르다는 특성으로부터 기인 했다고 사료된다. 위 가정을 통해, 동일한 두 Upflow anaerobic sludge blanket reactor (UASBr)를 Glucose와 Sulfate 공급하면서 연속 운전하였으며 한 개의 반응조에만 Glucose의 공급을 중단하는 Starvation을 적용해 보았다. 그 결과 Starvation 기간을 거친 반응조에서 20% 향상된 Sulfate 제거율이 관찰 되었으며, 90% 이상의 유기물 분해가 황 환원균에 의해 이루어 졌다. Scanning electron microscope (SEM)과 Transmission electron microscope (TEM)을 이용한 그래뉼 구조 분석에서도 EPS가 소비된 것을 확인 할 수 있었고, 그래뉼 내부에 위치한 미생물 세포들이 파괴 된 것을 알 수 있었다. Next-generation sequencing (NGS)을 이용한 미생물 분석을 통해서도 황 환원균의 군집도 및 개체수 증가를 확인할 수 있었다. 본 연구를 통해서 Starvation이 혐기성 그래뉼 내의 메탄균과 황 환원균 사이의 경쟁 관계에서 황 환원균을 우점화 할 수 있는 효과적인 방법임을 확인할 수 있었으며 이를 이용하여 황 환원균을 이용한 하수 처리 시스템의 보다 빠른 Start-up이 가능 할 것으로 사료된다.
또한 황 환원균이 우점화된 반응조에서 생산되는 Sulfide를 이용하여 황 연료전지로부터 전기를 생산하는 시스템의 적용 실험이 진행되었다. 생물학적으로 생산된 Sulfide를 이용하여 연료전지를 Batch 모드와 연속운전 모드로 운전하였으며, 이에 따른 전기생산 가능성을 확인하였다. Batch 모드 실험 결과 8시간의 Hydraulic retention time (HRT) 조건에서 UASBr 유출수 내의 Sulfide가 100% 제거 되는 것을 볼 수 있었고, 연료전지의 목적이 전기생산과 함께 유출수 내의 Sulfide를 제거하는 것 임을 종합적으로 고려하여, 연속운전의 HRT를 8시간으로 하였다. 연속운전은 연료전지를 직접 UASBr에 연결하여 운전 하였고, 50일동안 진행되었다. 그 결과 운전 초기와 비교하여 운전이 진행됨에 따라 연료전지의 Sulfide 제거효율이 30% 감소하는 현상을 보였고, 이의 원인을 규명하기 위하여 SEM과 X-ray diffraction (XRD)을 이용한 전극 표면 분석을 수행하였다. 그 결과 Sulfide의 산화 과정에서 생산된 황이 Anode 전극 표면에 흡착됨에 따라 연료전지의 효율 감소가 나타난 것으로 규명되었다. 또한 이를 통하여 본 하수처리 시스템에서 Anode 표면의 순수한 황을 회수 하는 것의 가능성을 확인 할 수 있었다.
본 신개념 하수처리 시스템은 보다 지속 가능한 하수처리의 규현을 가능하게 할 수 있을 것으로 사료된다. 또한 본 연구 결과를 바탕으로 하수에 포함된 유기물 처리율의 추가적 향상과 연료전지의 전기 생산기능 최적화에 관한 연구가 필요할 것으로 사료된다.
