Separation of particles and biological particulates has many potential applications in the bio-chemical research field such as purification of sample, blood cell fractionation, separation of mammalian cells from normal cells and others. Due to its non-invasive nature, the optical force has been successfully used for bio-logical cell manipulation in various applications. Although several types of optical force platform has been developed and many studies for single and multiple particulates manipulation, purification and sorting pur-pose, there is some limit of throughput and non-continuous matter. This thesis presents theoretical modeling and experimental approaches of optical cell separation and practical applications of cross-type optical particle separator (COPS) for separation and manipulation of cells.
First, in order to apply an cross-type optical particle separation method, a method for improving the microfluidic size separation technique known as pinched flow fractionation (PFF) has been demonstrated experimentally and analyzed by performing numerical calculations. This enhancement of separation is achieved by imposing a cross-type optical scattering force on the original PFF design. Three different polystyrene latexes (PSLs) with diameters of 2, 5, and 10 um were separated with PFF and optically enhanced PFF (OEPFF) devices. The separations achieved with the two devices were compared and enhancements in the separation distance by factors of up to approximately 15 were achieved. Theoretical calculations were also performed to interpret these results.
Next developed and experimentally verified the double emulsion behavior in the cross-type optical particle separation as the simplest model of cell in the COPS. The behavior of double emulsions in a cross-type optical particle separation system was studied for different combinations of the refractive indices and different inner and outer layer radii. The radii and refractive indices of the double emulsions were easily ad-justed by taking advantage of the co-flowing geometry of a cross-type optical particle separation device. An analytic expression of the optical forces on a pair of concentric spheres was derived using the photon stream method in the ray optics regime. The predicted optical force distributions and trajectories of the double emul-sions were calculated and found to agree well with the experimental data.
For applying COPS device in real blood cell separation, we measure the optical mobility of each cell components. The optical mobility of blood cell components depend on their size and shape. Without any staining, blood cell components have been isolated with FACS machine. Because of their irregular scattering characteristics, RBCs have the largest retention distance. And the theoretical and experimental comparison of resolution has been carried.
The final section of this thesis deals with the use of cross-type optical particle separation for the practical application. A cross-type optical particle separation method and optically enhanced flow fractional method are used for separating platelet from whole blood in a continuous flow manner. A device was designed to separate small particulate from relatively larger particle. Blood platelet can be separated from blood since platelet has relatively small size to other blood cell components. We assumed that the platelet size is smaller than 5um in diameter and other cell components (WBC and RBC) are larger than 5 um. By means of the flow cytometric analysis, the platelet was successfully separated.
본 학위논문에서는 광력에 의한 세포의 분리를 모사하기 위한 이론적인 모델링과 실험적인 측정이 수행 되었다. 또한 교차형 광학 분리기기 기법의 실제적인 적용을 통하여 실제 세포를 제어하고 분리하였다.
먼저 교차형 광학 분리기를 실제로 적용하기 위하여 미세입자의 유체 내부에서의 분리기법인PFF 기법이 실험적으로 적용되었으며 이를 수치적으로 해석하였다. 일반적으로 알려진 PFF와 교차형 광학 분리기를 통합함을 통하여 분리 성능을 향상시킬 수 있었다. 세가지의 서로 다른 크기의 PSL 입자(2, 5, 10 um) 를 사용하였으며 PFF 와 OEPFF에서 분리를 하여 보았다. 15배 이상의 분리성능이 향상 됨을 확인 할 수 있었으며 이의 이론적인 해석도 병행 되었다. 또한, 실험적/ 이론적 분리 정밀도를 통하여 장비의 성능을 평가 하였다.
세포의 가장 간단한 교차형 광학 분리기 내에서의 이론적인 모델링을 위하여, 이중 액적의 교차형 광학 분리기 내에서의 거동이 이론적으로 개발되었으며 실험적으로 검증되었다. 내부와 외부 액적의 굴절률을 조절함을 통하여 이중 액적의 거동 특성을 확인하였다. 이중 액적의 크기와 굴절률은 동심형 구조를 갖는 디바이스를 통하여 쉽게 조절 할 수 있으며, 이를 교차형 광학 분리기에 적용하였다. 이중 구형입자에 대한 광력의 이론해는 광자 흐름 방법을 통하여 계산 되었으며, 실험적으로 이를 검증하였다.
실제 혈액 세포에 교차형 광학 분리기를 적용하기 위하여, 이론적인 해석방법과 함께, 세포의 불균일성을 보정하기 위하여 각 혈액 세포의 광 이동도(optical mobility)를 측정하였다. 염색등을 거치지 않고 FACS 장비를 통하여 세포를 분리하였다. 광 이동도는 혈액 세포의 크기와 모양에 따라 결정 됨을 확인하였으며, RBC의 경우에는 디스크 모양으로 인하여 가장 큰 분리 거리를 가짐을 확인 할 수 있었다. 또한 이론적/실험적 분리 정빌도에 대한 비교가 수행 되었다.
이와 함께 실제 분리를 위하여 기존의 교차형 광학 분리기 방법과 OEPFF 기법을 이용한 혈소판 분리를 수행하였다. 크기에 따라서 입자들이 분리됨을 이용하여 디자인 되었으며, 혈액의 혈소판은 다른 혈액의 세포 성분에 비하여 상대적으로 작은 크기를 가짐을 이용하여 분리를 할 수 있었다. 혈소판은 보통 3um 보다 작을 것으로 예상 했으며 다른 세포 성분은 5um 보다 클 것으로 예상하였다. FACS 분석을 통하여 혈소판의 분리가 성공적으로 이루어 짐을 확인 할 수 있었다.
