Although the etiology of complex diseases includes both genetic and environmental factors, many studies focus singularly on either type of factor. Consideration of both factors and the interactions between them will advance the current understanding of pathogenetic mechanisms. However, because disease inci-dence is often influenced by long exposure to environmental factors (e.g., the 30-year latency period between smoking and the onset of lung cancer), cumulative observations of environmental exposure are necessary in order to accurately determine gene-environment interactions. Previous work, including epidemiological stud-ies, has been limited to measurement of current exposure to environments, primarily by observing concentra-tions of compounds in serum or urine. To address this challenge, we monitored the accumulated effect of environmental factors on the epigenomic profile. By integrating this information with data regarding chemical alterations to the epigenome, we developed a method for identifying gene-environment interactions related to pathogenesis that takes into account past exposure to environmental factors. We applied this method to breast cancer and found two candidate environmental compounds (arsenic and furan) that interact with a BRCA1 to contribute to breast-cancer incidence. Furthermore, we show that arsenic and BRCA1 interact through estrogen receptor alpha, a protein that has been implicated in breast cancer.
질병은 유전적 소인과 환경적 소인 모두에 의해 발생함에도 불구하고, 많은 기존 연구들은 유전 혹은 환경적인 소인, 둘 중 하나에만 관심을 기울이고 있다. 이 두 가지 요소가 모두 고려되어 질병 연구가 진행되어야 하며, 또한 두 요소 사이의 상호작용이 고려되어 질병 연구가 진행되어야 한다. 특별히 유방암과 같은 복잡성 질환(complex disease)는 유전적 소인만이 아니라 환경적 소인이 매우 큰 것으로 나타나고 있다. 본 논문은 유전-환경 상호작용 연구를 위한 첫 단계로, 유전-환경 상호작용의 결과물로 볼 수 있는 대사체(metabolome) 데이터에 대한 연구를 수행하였고, 두 번째로 유전-환경 상호작용 추론에서의 난제인 장기간 환경노출의 측정 방법을 제안하는 연구를 수행하였다.
대사체 연구를 통해서 본 논문은 유방암 환자의 소변으로부터 정량된 대사체 데이터 정보로부터 효과적으로 유방암 환자군과 정상인을 구분해 내는 다변수 유방암 분류기를 구축하였으며, 다변수 방법의 적용을 통하여 기존 단변수 방법에 비해 보다 정확한 분류가 가능해짐을 확인하였다.
또한, 장기간 환경노출의 측정 방법의 개발을 통해 정확한 유전-환경 상호작용 추론을 가능케 하는 연구를 수행하였다. 질병의 발생은 단기간의 유해환경 노출에 의해서가 아닌, 장기간의 노출에 의해서 발생한다. (담배와 폐암 발병의 평균기간: 약 30년) 이를 해결하기 위해 본 연구는, 환경에 의해 그 상태가 변화되는 후성유전체(epigenome) 정보를 사용하여 과거로부터의 환경노출을 측정하는 방법을 제안하였다. 이를 통해 비소와 BRCA1유전자의 변이가 서로 상호작용하여 유방암을 일으킬 수 있는 후보 상호작용 관계임을 밝혀내었다.
