Functional inorganic nanostructures have been much attractive for various applications to biosen-sors, catalytic systems, electronic and optical devices. Various porous polymer nanostructures serve as a tem-plate for the formation of inorganic nanostructures. Electrospinning is widely used to produce highly porous nanofiber structures with large surface area-to-volume ratio. Several techniques has been employed to homo-geneously generate inorganic nanostructure on electrospun nanofibers, however, there has a need to a new approach for surface modification of electrospun nanofibers for homogeneously and spontaneously generating inorganic nanostructures under ambient conditions.
Recently there has been increasing attention on a mussel-inspired surface modification because of their capability for the coatings of various organic and inorganic materials. It is known that the co-existence of the catechol moiety of 3,4-dihydroxy-L-phenylalanine (DOPA) and the amine group of lysine is crucial for strong mussel adhesion in aqueous environment. Catechol moiety has enough reducing capability for generating metal nanoparticles with a strong adhesive property.
Here we introduce a mussel-inspired surface modification for inorganic coatings of electrospun pol-ymer nanofibers. Firstly, catechol-grafted polymer nanofibers are used as a functional template for the for-mation of noble metal nanostructures. The synthesis of metal nanostructures is driven by the oxidation of the catechol moiety having an enough reducing potential for metal ions. 3,4-dihydroxyhydrocinnamic acid is chemically grafted to poly(vinylalcohol) (PVA) using ethylenediamine as a linker, producing catechol-grafted PVA (PVA-g-ct). Noble metal ions (e.g., Ag+, Au3+ and Pt4+) are spontaneously reduced to metal nanoparticles on electrospun PVA-g-ct nanofibers. The highly open porous, three-dimensional metal nanostructures are generated when the catechol-grafted nanofibers are simply immersed in metal ion precursor solution under ambient conditions.
Secondly, polydopamine coatings are employed for simultaneous reduction of metal ions to prepare electrospun polymer nanofibers functionalized with metal nanoparticles. The oxidative polymerization of dopamines mediates the reduction of silver ions to metallic silver nanoparticles and their deposition on polymer nanofibers. PVA is electrospun to nanofibers, and then incubated in a mixture of dopamine hydrochloride and silver nitrate (AgNO3) in methanolic solution. The formation of silver-pD-coated PVA nanofibers stabilizes the porous structure of nanofibers in aqueous solution even with vigorous stirring. Although polydopamine coatings usually generate irregular aggregates on the nanofibers, one-pot synthesis of a silver-pD hybrid layer on electrospun nanofibers shows a relatively uniform surface morphology.
These mussel-inspired reductive processes on polymer nanofibers can provide a facile method to homogeneously generate metal nanoparticles and unique metal nanostructures within three dimensional po-rous structures, providing functional metal nanostructures that can be used for a wide range of practical applications.
다공성의 기능성 무기 나노 구조체는 부피비 넓은 표면적에 기인하여 바이오센서, 촉매 시스템, 조직공학, 전자 또는 광학 디바이스 등의 다양한 분야에 응용되고 있다. 다양한 다공성의 고분자 나노 구조체는 이러한 무기 나노 구조체의 합성을 위한 지지체로 이용되고 있다. 고분자 나노 구조체를 제조하기 위한 다양한 기술 중에서도, 특히, 전기방사 기술은 대면적의 다공성 나노섬유 구조체를 적은 비용으로 손쉽게 제작할 수 있고, 다양한 고분자를 이용할 수 있기 때문에 다공성의 고분자 나노 구조체를 제조하기 위한 효과적인 방법으로써 널리 이용되고 있다.
전기방사로 제작된 고분자 나노섬유의 물리적, 화학적 성질을 개선하기 위하여 다양한 접근방법으로 나노섬유 표면에 무기 나노 구조체를 도입하기 위한 연구가 진행되고 있다. 그러나 대부분의 경우에서는 무기 나노 구조체를 형성하기 위하여 무기물 나노입자의 개별적인 합성을 필요로 하거나, 금속 이온을 이용하여 직접 나노섬유 표면에 금속 나노입자를 합성할 경우 금속 이온의 환원을 위한 산화제를 첨가하거나 고온의 합성 조건을 필요로 한다. 따라서 상온, 상압의 조건에서 나노섬유에 무기물 나노 구조체를 균일하고 자발적으로 도입하기 위한 새로운 표면 개질 방법의 필요성이 대두되고 있다.
최근 홍합 접착 특성을 모방한 표면 개질 방법의 경우 금속, 산화물, 고분자, 생체 분자 등 광범위한 무기 또는 유기 물질의 표면에 코팅될 수 있기 때문에 이에 대한 관심이 증가하고 있다. 도파로 알려져 있는 3,4-dihydroxy-L-phenylalanine (DOPA)의 카테콜 성분과 라이신 (lysine)의 아민 기의 존재가 이러한 홍합 접착 특성에 결정적인 역할을 하는 것으로 보고되고 있다. 특히, 카테콜의 경우 강한 접착 특성뿐만 아니라 다양한 금속 이온을 금속 나노입자로 환원시킬 수 있는 충분한 환원 능력을 가지고 있다.
본 연구에서는 홍합 접착 특성을 모방한 표면 개질 방법을 이용하여 무기 나노 구조체를 합성하기 위한 두 가지 접근 방법을 제시한다. 첫째로, 카테콜 기가 결합된 나노섬유를 금속 나노 구조체의 형성을 위한 기능성 지지체로 이용한다. 나노섬유 표면의 카테콜 기는 자발적인 산화를 통하여 나노섬유 표면에 금속 이온을 환원시킴으로써 금속 나노 구조체의 형성을 유도한다. 이러한 접근방법을 이용하기 위하여, 에틸렌디아민을 연결기로 이용하여 카테콜 기를 포함하고 있는 3,4-dihydroxy-hydrocinnamic acid를 PVA 고분자에 합성한다. 합성된 고분자를 이용하여 제조된 나노섬유는 상온, 상압에서 금, 은, 백금 등의 금속 이온 용액에 단순히 담그는 것만으로도 금속 이온을 나노섬유 표면에 환원시킬 수 있고, 이를 통하여 금속 나노 구조체를 쉽게 합성할 수 있게 된다.
둘째로, 전기방사로 제조된 PVA 나노섬유에 금속 나노 구조체를 합성하기 위하여 폴리도파민 코팅을 이용한다. 도파민의 구성요소인 카테콜 기의 산화에 기인하는 폴리도파민 코팅은 은 이온의 환원을 유도함과 동시에, 폴리도파민 특유의 접착 능력을 통하여 환원된 은 나노입자를 나노섬유 표면에 접착할 수 있도록 중재하는 역할을 한다. 전기방사로 제조된 PVA 나노섬유를 도파민과 질산은의 혼합 용액에 배양함으로써 PVA 나노섬유 표면에 은 나노입자를 쉽게 합성할 수 있게 된다. 은과 폴리도파민이 코팅된 PVA 나노섬유는 수용액 상에서도 안정적으로 다공성의 나노섬유 형태를 유지한다.
본 연구에서 제시하고 있는 홍합에서 모방한 표면 개질 방법은 전기방사로 제조된 다공성의 나노섬유 표면에 금속 나노입자와 금속 나노 구조체를 도입하기 위한 새로운 접근방법을 제시한다. 이러한 방법으로 합성된 다공성의 기능성 금속 나노 구조체는 바이오센서, 촉매 시스템, 조직공학, 전자 또는 광학 디바이스 등의 다양한 응용 분야에 적용할 수 있을 것으로 기대한다.
