The yeast Pso2 is known as an interstrand crosslink (ICL) repair protein which has a 5’ exonuclease activity and hairpin structure specific endonuclease activity. In this report, Pso2 is identified as a multicopy suppressor of dna2Δ405N mutant. Both nuclease deficient Pso2 and N-terminal region of Pso2 devoid of nuclease domain could suppress dna2Δ405N mutant though the nuclease activity of Pso2 in ICL repair is essential. We investigated the interaction between Pso2 and Okazaki fragment processing proteins. We showed that Pso2 stimulated the flap endonu-clease activity of Fen1 and this is most likely through their physical interaction. Pso2 also stimulated the endonuclease activity of Dna2. These findings indicate that Pso2 is involved in Okazaki fragment processing.
Pso2는 가닥간 공유결합 DNA 손상에 작용하는 단백질로 5’ exonuclease이자 hairpin 구조에 특이성을 보이는 endonuclease로 알려져 있다. Pso2가 dna2Δ405N돌연변이의 multicopy suppressor임을 밝혀냄으로 이 연구가 시작되었다. 이 억제작용에는 Pso2의 nuclease 활성이 필요하지도 않으며, nuclease domain도 필요 없이 Pso2의 N-terminal domain만으로도 억제가 가능하였다. 가닥간 공유결합 DNA 손상 처리 과정에서 Pso2의 효소활성이 필수점임을 감안할 때에 이는 지금까지 알려지지 않은 Pso2의 활성이 존재함을 시사한다. dna2돌연변이의 억제 기작에는 Pso2와 Fen1간의 관계가 중요함을 밝혀내었다. Pso2는 Fen1의 endonuclease 활성을 증폭시키며 그 과정에는 Pso2와 Fen1의 물리적인 상호작용이 관여할 것이라 생각된다. 뿐만 아니라 Pso2는 Dna2의 endonuclease활성도 증폭시킴을 또한 밝혀내었다. 이와 같은 실험 결과를 토대로 Pso2는 오카자키 단편 처리과정에서 Fen1과 Dna2의 활성을 조절하는 인자로 작용함을 밝혀내었다.
