Roles of geo-biochemical interactions on reductive degradation of chlorinated organics were investigated in this Ph. D. dissertation to provide better understanding of contaminant fate in sub-surface environment. Especially, effect of interactions on the degradation of chlorinated organics was investigated using nano-sized zero-valent iron (nZVI), reactive iron-bearing soil minerals (IBSMs), iron reducing bacteria (IRB), and electron transfer mediators (ETMs).
The reductive degradation of 1,1,1-trichloethane (1,1,1-TCA) by interaction of nZVI and magnetite (FeII1III2O4) was investigated as the nZVI/IBSMs/contaminant interaction. A remarkable reductive degradation of 1,1,1-TCA (0.289 hr-1) was observed in nZVI (0.01 g/24 mL) suspension in 5 h, while the degradation was not significant in nZVI (0.01 g)/magnetite (0.5 g) suspension in 80 h. This indicated that magnetite can inhibit the reactivity of nZVI for 1,1,1-TCA degradation. No significant difference in the inhibition of nZVI reactivity by magnetite was observed in the pH range of 6 to 9. The result of X-ray photoelectron spectroscopy revealed that magnetite surface was reduced by an electron transfer from nZVI. The results showed that electrons from nZVI were used for the reduction of magnetite surface rather than the reduction of 1,1,1-TCA, leading to the decrease of 1,1,1-TCA degradation in this study.
The effect of ETMs on biotransformation of lepidocrocite (γ-FeIIIOOH) by Shewanella putrefaciens CN32 (CN32) was investigated as the IRB/IBSMs/ETMs interaction. In PIPES buffer without phosphate, green rust ([Fe4IIFe2III(OH)12][A2- o 6H2O]) formation was observed with fast Fe(II) production rate (0.44-0.56 mM d-1) during the bioreduction of lepidocrocite with exogenous ETMs (quinones). In contrast, goethite (α-FeIIIOOH) formed at slow Fe(II) production rate (0.24-0.29 mM d-1) with endogenous ETMs (flavins). In the presence of phosphate, formation of green rust and vivianite (FeII3(PO4)2o8H2O) was observed with fast Fe(II) production rate (0.54-0.74 mM d-1) during the bioreduction of lepidocrocite with exogenous ETMs, while vivianite formed at moderate Fe(II) production rate (0.36-0.40 mM d-1) with endogenous ETMs. Vivianite formed in all experimental cases with phosphate in a broad range of Fe(II) production rates (0.23-0.74 mM d-1). The results clearly showed that both types of ETMs and presence of phosphate can significantly affect bioreduction of lepidocrocite and its mineral transformation.
The reductive degradation of carbon tetrachloride (CT) by interaction of IBSMs (i.e., magnetite and lepidocrocite) and CN32 was investigated as the IRB/IBSMs/contaminant interaction. Chloroform (16.1-29.4%), carbon monoxide (2.4-23.8%), and formate (0-58.0%) were measured as main products during the degradation of CT by magnetite and lepidocrocite with CN32. X-ray diffraction and electron microscope analyses revealed that the biogenic vivianite can form during the CT degradation in magnetite and lepidocrocite suspensions with CN32. This indicated that enhanced CT degradation was caused by formation of biogenic vivianite. The dechlorination kinetics of CT by chemogenic vivianite was much faster than that by magnetite and lepidocrocite with CN32. The highest formate production (84.2%) was observed during a full degradation of CT by the chemogenic vivianite. The experimental results showed that biogenic vivianite formed during the bioreduction of IBSMs played a pivotal role for the reductive dechlorination of CT.
Lastly, the reductive degradation of CT by interaction of nZVI and riboflavin was investigated as the nZVI/ETMs/contaminant interaction. The degradation kinetic of CT by nZVI without riboflavin (0.556 ± 0.044 h-1) was 1.5 times higher than that with riboflavin (0.370 ± 0.012 h-1) at first cycle. Significant amount of riboflavin (94%) was adsorbed on reactive nZVI surface in 25 h and competed with CT for the adsorption sites on the nZVI surface, which results in the decrease of the degradation kinetic of CT at first cycle. On the other hand, the degradation kinetic of CT by nZVI without riboflavin declined to 0.122 ± 0.033 h-1 at third cycle, while that with riboflavin (0.663 ± 0.005 h-1) significantly increased. Riboflavin was reduced by nZVI at third cycle and played a role of electron transfer shuttle to (1) reduce passive iron oxides on nZVI surface leading to excavation of reactive Fe (0) core and to (2) directly reduce CT to chloroform by one-electron transfer causing the enhanced reductive dechlorination of CT at third cycle. The experimental results showed that the riboflavin reduced by nZVI played a pivotal role for prolongation of nZVI reactivity during the continuous dechlorination of CT.
본 박사학위 논문에서는 토양 및 지하수 환경에서 실제 발생할 수 있는 여러가지 지질-생화학 상호작용들이 염소계 유기화합물의 환원적 탈염화에 끼치는 영향에 대해서 연구하였다. 특히 최근 많은 연구가 진행 되고 있는 나노 영가철과 철 함유 토양광물 같은 반응성 있는 환원제들에 의해 염소계 유기화합물이 분해 될때 철 환원 미생물 또는 토양내 유기물들이 각 상호작용에 끼치는 영향에 대해서 알아보았다. 각 상호작용은 몇몇 가지 대표 물질들을 선택하여 실제 자연환경을 보다 간단한 모델로 해석하여 연구되었다.
먼저 나노 영가철과 마그네타이트 (FeII1III2O4)의 상호작용이 1,1,1-trichloethane (1,1,1-TCA) 분해에 끼치는 영향을 영가철/산화철/오염물질 상호작용의 일환으로 연구되었다. 나노 영가철에 (0.01 g/ 20 mL) 의해 약 80%의 1,1,1-TCA가 5시간안에 분해 되었지만, 0.5 g의 마그네타이트를 넣은 결과 약 20 %의 1,1,1-TCA가 80 시간안에 분해 되었다. 이는 마그네타이트가 나노 영가철의 1,1,1-TCA분해능을 저해한 것을 보여주는 결과이다. 이러한 나노 영가철의 분해능 저해 현상은 6에서 9의 pH범위에서 전반적으로 나타났다. X-선 광전자 분광법을 이용한 결과, Fe(III)로 충만했던 마그네타이트의 반응 표면이 나노 영가철의 주입으로 인하여 Fe(II)로 환원된 것이 관찰 되었다. 결과적으로 나노 영가철의 전자가 1,1,1-TCA의 분해를 위해 쓰인 것보다, 마그네타이트의 표면을 환원 시키는데 사용되었고, 이 때문에 1,1,1-TCA의 분해능이 현저히 떨어진 것임을 보여준다.
다른 종류의 전자전달체들이 레피도크로사이트 (γ-FeIIIOOH)와 Shewanella putrefaciens CN32 (CN32)의 상호작용에 끼치는 영향을 미생물/수산화철/전자전달체 상호작용의 일환으로 연구되었다. 인산염이 없으며 외인성 전자전달체 (퀴논)가 존재하는 경우에는 빠른 Fe(II) 생산속도 (0.44-0.56 mM d-1)를 보이면서 레피도크로사이트가 그린러스트 ([Fe4IIFe2III(OH)12][A2- o 6H2O])로 변하였으나, 내인성 전자전달체 (플라빈)이 존재할 때는 느린 Fe(II) 생산속도 (0.24-0.29 mM d-1)를 보이며 고에타이트 (α-FeIIIOOH)로 변하였다. 인산염이 존재하며 퀴논이 존재할때는 빠른 Fe(II) 생산속도 (0.54-0.74 mM d-1)를 보이며 그린러스트와 비비안나이트 (FeII3(PO4)2o8H2O)로 변하였지만, 플라빈이 존재할때는 느린 Fe(II) 생산속도 (0.36-0.40 mM d-1)를 보이며 비비안나이트만 생성이 되었다. 본 결과는 다른유형의 전자전달체들과 인산염의 존재유무가 레피도크로사이트의 미생물적 환원과 그에 따른 광물 변형에 중대한 영향을 끼치는 것을 보여준다.
철 함유 토양광물 (마그네타이트, 레피도크로사이트)과 CN32의 상호작용이 carbon tetrachloride (CT) 분해에 끼치는 영향을 미생물/산화철,수산화철/오염물질 상호작용의 일환으로 연구되었다. 마그네타이트와 레피도크로사이트가 CN32에 의해서 환원되면서 CT가 chloroform (16.1-29.4%), carbon monoxide (2.4-23.8%), formate (0-58.0%)으로 분해되었다. X-선 회절과 전자현미경 데이터를 분석한 결과, 마그네타이트와 레피도크로사이트가 CN32에 의해서 비비안나이트로 변형된 것이 관찰되었다. 이는 생성된 비비안나이트에 의해서 CT의 분해가 촉진되었음을 보여주는 간접적인 증거이다. 화학적으로 합성된 비비안나이트를 이용하여 CT 분해를 수행한 결과, 기존의 마그네타이트, 레피도크로사이트와 CN32의 상호작용에 의한 분해속도보다 현저히 빠름이 증명되었으며 대부분이 formate로 분해됨을 밝혔다. 본 결과들은 비비안나이트가 CT분해에 탁월한 분해능을 가지고 있다는 것을 보여준다.
마지막으로 나노 영가철과 리보플라빈의 상호작용이 CT 분해에 끼치는 영향을 나노 영가철/전자전달체/오염물질 상호작용의 일환으로 연구하였다. 첫 번째 사이클에서는 나노 영가철에 의한 CT를 분해 키네틱 (0.556 h-1) 이 리보플라빈을 넣었을 때 (0.370 h-1) 보다 약 1.5배 빠른것으로 나타났다. 리보플라빈의 나노 영가철 흡착실험을 통해서 리보플라빈과 오염물질인 CT와의 경쟁적인 흡착이 CT분해 저해현상을 야기시킨것으로 확인되었다. 세번째 사이클에서는 CT분해에 대한 나노 영가철의 반응성이 현저히 떨어지는 것을 관찰 (0.122 h-1) 하였으나, 리보플라빈을 넣은 샘플에서는 오히려 증가하는 것 (0.663 h-1) 으로 나타났다. 이는 나노 영가철에 의해서 환원된 리보플라빈이 부동태화된 나노 영가철의 표면을 Fe(III)에서 Fe(II)로 환원시키는 기작과 직접 전자를 CT에 빠르게 전달하는 기작을 통해서 나타나는 결과임을 실험을 통해 증명하였다. 본 결과는 리보플라빈이 연속된 CT 분해 중에 나노 영가철의 반응성을 유지 또는 증가시킬 수 있음을 보여준다.
