Xylitol is a five-carbon sugar alcohol that has global markets. It has beneficial health properties and represents an alternative to current conventional sweeteners. It has low calory, but its sweetness level is equal to that of sucrose. Additionally, it has anticariogenic effect that inhibits the growth of the tooth-decaying bacterium. Xylitol can also be used as a chemical building block for the production of high-value chemicals such as xylaric acid and glycols.
The asporogenic diploid yeast, Candida tropicalis, is of academic and industrial interest. Especially, C.tropicalis has been extensively studied in the production of xylitol in terms of their distinguished xylose-assimilating ability. The synthesis of xylose reductase (XR) in C. tropicalis is considerably repressed in cells grown on glucose medium because of glucose repression. For this reason, glucose is not an appropriate cosubstrate for supporting cell growth and xylitol production. However, for economic feasibility, inexpensive hemicellulose should be used as a raw material and the hemicellulose is hydrolyzed by mineral acid to produce hydrolysate that is mainly composed of glucose and xylose.
A gene encoding glyceraldehyde-3-phosphate dehydrogenase (GAPDH) was isolated from the genomic DNA of C. tropicalis ATCC 20913 to construct a constitutive expression system without glucose repression. The endogenous XR gene (xyl1) was placed under the control of GAPDH promoter and integrated into the genome of C. tropicalis N43 which is xyl2-disrupted and xyl1-partially disrupted mutant. Expression of XR was confirmed by determining enzyme activity in cells grown on a medium containing glucose as a carbon source. The resulting recombinant yeast, C. tropicalis JA4, showed higher XR activity (465±77 mU/mg of proteins) whereas that of BSXDH-3, the control strain, was not detectable.
Glucose uptake inhibits xylose utilization in xylose-assimilating yeasts such as engineered Saccharomyces cerevisiae and Pichia stipitis. Our strain, C.tropicalis have also same problem during the fermentation in glucose and xylose mixture. Xylose uptake was delayed by glucose during the initial culture phase.
Heterologous expression of xylose transporter in C.tropicalis was performed to enhance the xylose uptake rate and improve xylitol production. For additional expression of xylose transporters in C. tropicalis, we constructed expression system that can remove the selection marker. The codon optimized Neurospora crassa XR, NXRG was inserted into expression vector harboring GAPDH promoter for its potent activity toward xylose and then the expression cassette was inserted into the genomic DNA of C. tropicalis L10. For integration of heterologous gene expression cassette, URA3 gene was deleted from NXRG-expressing strain, LXU1 through pop out. The resulting strain, LX was ready to introduce a heterologous xylose transporter for improving xylose uptake.
Heterologous four xylose transporter genes, At5g59250, At5g17010 from Arabidopsis thaliana, Sut1 from Pichia stipitis and Gxf1 from Candida intermedia were codon optimized for functional expression in Candida tropicalis. The codon-optimized xylose transporter genes, CoXT1, CoXT2, CoSut1, and CoGxf1 were placed under the control of GAPDH promoter and integrated into the genome of C. tropicalis LX which is xyl2- disrupted and NXRG introduced mutant. The resulting strain C. tropicalis LXT2 (a At5g17010 gene introduced mutant) showed highest xylose uptake ability and volumetric productivity. Xylose uptake rate of C. tropicalis LXT2 was increased by 29% compared to that of LXU1 during incubation time, from 12 to 42h. LXT2 produced xylitol with a volumetric productivity of 1.14g L-1 h-1, and this value was 25% higher than those of LXU1, the parent strain.
Heterologous expression and localization of the A. thaliana xylose transporter encoded by CoXT2 were confirmed by membrane protein extraction and western blot analysis. The data indicated that the A. thaliana transporter encoded by CoXT2 were expressed successfully and localized to the plasma membrane in C. tropicalis.
자일리톨 (xylitol)은 설탕의 대체 감미료로서 세계적인 시장을 가지고 있는 오탄당 알코올이다. 자일리톨은 설탕에 비해 낮은 열량을 가지면서도 비슷한 수준의 당도를 가지고 있으며 충치를 예방하는 효과를 가지고 있다. 또한 자일라릭 산 (xylaric acid) 이나 글라이콜 (glycol)과 같은 높은 가치를 지니는 화학물질들의 전구체로 쓰인 다는 점에서 식품산업 에서는 물론 화학산업 에서도 중요한 물질이다. 실제 산업적으로 자일리톨 생산은 바이오매스 산가수분해물을 원료로 사용하여 이온교환수지를 통해 자일로스를 분리하고 정제한 뒤 화학적 수소첨가반응을 이용해서 생산하고 있다. 하지만 이러한 화학적인 생산방법은 많은 비용과 환경오염을 유발하기 때문에 최근의 친환경적인 산업으로의 전환을 요구하는 세계적인 추세와는 거리가 있다. 따라서 미생물의 발효에 의한 생물학적 생산방법이 새로운 대안으로 떠오르고 있으며 무포자성 이배체 효모인 캔디다 트로피칼리스 (Candida tropicalis)는 지금껏 알려진 여러 자일로스 이용 균주들 가운데서 뛰어난 자일리톨 생산능력으로 학문적으로 널리 연구 되어왔다.
실제 산업에서 생물학적 자일리톨 생산방법이 경제성을 가지기 위해서는 값이 싼 바이오매스 산가수분해물을 이용하게 되는데 이 때 산가수분해물은 자일로스 뿐만 아니라 포도당도 포함하고 있다. 따라서 자일리톨 생산시 포도당을 보조기질로 사용한다면 효율적인 생산이 가능하다. 하지만 배지에 포도당이 존재하는 조건에서는 자일로스를 자일리톨로 전환시켜주는 중요효소인 자일로스 환원효소 (xylose reductase; XR)의 합성이 억제되는, 즉 이화대사산물억제가 생긴다는 것이 문제점이었다.
포도당 존재 하에서도 XR을 발현시키기 위해 캔디다 트로피칼리스 균주의 글리세르알데히드-3-인산 탈수소효소 (glyceraldehydes-3-phosphate dehydrogenase; GAPDH)의 프로모터 (promoter)를 이용하여 항구적 발현 시스템을 구축하였다. 캔디다 트로피칼리스 균주의 XR을 포함하는 항구적 발현카세트를 캔디다 트로피칼리스 균주에 도입한 결과, 구축된 균주 JA4의 XR은 포도당을 배지로 하는 조건에서 높은 활성 (465±77 mU/mg of proteins)을 보이는 반면 대조 균주 (control strain)에서는 활성이 나타나지 않았다.
포도당에 의한 XR의 합성을 저해하는 문제를 극복했음에도 불구하고 포도당과 자일로스의 혼합배지에서 자일리톨을 생산 시 세포 내로의 포도당의 흡수가 자일로스의 흡수보다 선호되어 자일리톨의 생산이 저해되는 문제 또한 남아있었다. 이를 해결하기 위해서 이종의 자일로스 트랜스포터 (xylose transporter)를 캔디다 트로피칼리스 균주에서 발현시켜 자일로스의 흡수와 자일리톨의 생산을 향상시키고자 하였다. 효율적인 자일리톨의 생산을 위하여 자일로스에 대한 높은 활성을 가지는 뉴로스포라 크라사 (Neurospora crassa)의 자일로스 환원효소 (NcXR)를 항구적 발현 카세트를 이용하여 캔디다 트로피칼리스 균주에 도입하여 LXU1 균주를 구축하고 나서 추가적인 이종의 트랜스포터를 포함하는 발현카세트를 도입하기 위해 표지자 (selection marker)인 URA3 유전자를 제거하여 LX 균주를 구축하였다.
애기장대 (Arabidopsis thaliana)의 자일로스 트랜스포터 유전자인 At5g59250 과 At5g17010, 피키아 스티피티스 (Pichia stipitis)의 Sut1 그리고 캔디다 인터미디어(Candida intermedia)의 Gxf1 유전자를 캔디다 트로피칼리스에서 성공적으로 발현시키기 위해 이들 유전자들에 대해 코돈 최적화(codon optimization)을 수행하였다. 코돈 최적화가 수행된 자일로스 트랜스포터 유전자 CoXT1, CoXT2, CoSut1, 그리고 CoGxf1들을 각각 항구적 발현 카세트에 넣어 캔디다 트로피칼리스 균주에 도입하였다. 그 결과 코돈 최적화 된 애기장대의 자일로스 트랜스포터 유전자 At5g17010가 도입된 LXT2 균주가 대조 균주인 LXU1과 다른 트랜스포터 발현 균주들에 비해 뛰어난 자일로스 흡수능력과 자일리톨 생산능력을 보였다. LXT2 균주의 자일로스 흡수속도 (1.40g L-1 h-1)는 대조 균주에 비해서 29%, 생산성 (1.14g L-1 h-1)은 25% 향상되었다. 실제로 애기장대의 자일로스 트랜스포터가 캔디다 트로피칼리스에서 발현되고 세포막으로 이동하였는지를 확인하기 위해 세포막 단백질을 분리정제 하여 western blot 분석을 시행하였다. 특이적 항원 항체 반응을 이용하여 분석한 결과 이종의 트랜스포터가 성공적으로 발현되고 세포막으로 이동한 것을 확인하였다.
