Type 1 diabetes (T1D) is the result of an autoimmune destruction of pancreatic β cells. The cellular and molecular defects that cause the disease remain unknown. Human pluripotent stem cells, especially pluripotent cells generated from patients with T1D represent a potentially unlimited source of functional pancreatic endocrine lineage cells and would be useful for disease modeling. Here I report a highly efficient approach to induce human embryonic stem (ES) cells and induced pluripotent stem (iPS) cells to differentiate into endocrine cells and mature insulin-producing cells in a chemical-defined culture system. In addition, I show that induced pluripotent stem (iPS) cells can be generated from patients with T1D by reprogramming their adult fibroblasts with four transcription factors (OCT4, SOX2, KLF4 and CMYC). T1D-specific iPS cells, termed DiPS cells, have the hallmarks of pluripotency and can be differentiated into pancreaticendocrine and insulin-producing cells. These results are a step toward using DiPS cells in T1D disease modeling, as well as for cell replacement therapy.
제1형 당뇨병은 자가면역으로 인한 췌장 베타세포의 파괴로 인해 일어나며 정확한 세포 분자생물학적인 병인은 아직 알려지지 않고 있다. 제1형 당뇨병 환자로부터 역분화 줄기세포를 만든다면 췌장의 내분비세포를 무한히 얻을 수 있기 때문에 질병의 모델링에 큰 역할을 하게 될 것이다. 본 연구에서를 통해 배아줄기세포와 역분화줄기세포로부터 기능을 하는 인슐린 생산세포를 효율적으로 만들수 있는 배양 시스템을 확립하였다. 또한 제1형 당뇨병 환자의 체세포로부터 4가지 인자(OCT4, SOX2, KLF4, cMYC)를 발현시켜 역분화줄기세포를 만들었으며 인슐린을 생산하는 세포로 분화유도하였다. 이 결과는 제1형 당뇨병 연구뿐만 아니라 세포치료의 가능성을 높이는 중요한 연구가 될 것이다.
