In eukaryotes, the condensin complex, which contains two structural maintenance of chromosomes (SMC) subunits, SMC2 and SMC4, plays a vital role in the formation of higher order chromosome structures. The SMC2 and SMC4 subunits are known to form a heterodimer through their hinge domains, like other SMC proteins. In addition to mediating dimerization, the hinge domain has been suggested to be involved in other functions such as DNA binding. However, what is still unclear is how the two SMC proteins specifically recognize each other and how they bind to DNA. To address these questions, I purified and crystallized the complex between the yeast SMC2 and SMC4 hinge domains. The crystals of the SMC2/4 hinge belonged to the monoclinic space group C2 with unit cell dimensions of a = 186.25, b = 50.16, c = 153.82 A , β = 93.21º, and were likely to contain two molecules of the hinge domain complexes in the asymmetric unit.
진핵 생물의 염색체는 체세포 분열 전기에 응축되어 특유의 고차원적인 구조를 형성하는데, 이 때 핵심적인 역할을 한다고 알려진 단백질이 Condensin 복합체이다. 진핵 생물의 Condensin 복합체는 SMC 계열 단백질인 SMC2와 SMC4 단백질을 포함하며, 이 SMC2와 SMC4 단백질은 그들의 hinge domain 간의 결합을 통하여 2량체를 형성한다고 알려져 있다. 최근에는 hinge domain이 단순히 2량체를 만드는 데 필요할 뿐만 아니라, DNA와의 결합을 비롯한 다른 기능을 가지고 있다는 보고가 나오고 있다. 하지만 어떠한 작용 기작에 의해 SMC2 와 SMC4 단백질이 서로를 선택적으로 인식하고, DNA와 결합하는지에 대해서는 아직 분명하지 않은 상황이다. 이러한 문제들을 해결하기 위해, 나는 진핵 생물의 일종인 효모의SMC2/SMC4 hinge domain복합체를 정제하고 결정화하여 3.0 A 의 해상도까지 data를 얻는데 성공하였다. SMC2/4 hinge domain의 결정은 C2 space group으로 a = 186.25, b = 50.16, c = 153.82 A, β = 93.21º 의 단위 격자 안에 2개의 hinge domain 복합체 분자를 포함하고 있는 것으로 보였다.
