The most fundamental division within the nervous system is between sensory and motor. By definition, the activity of neurons in sensory systems is correlated primarily with external sensory stimuli, and the activity of neurons in motor systems is correlated more strongly with motor outputs. However, there is no clear and well established distinction between sensory and motor neurons at the cellular or molecular levels even it is known that neurons in different parts of the brain have different electrophysiological properties. We tested a hypothesis there are specific differences at the cellular level between neurons in sensory and motor system. Many literatures have reported sensory neurons maintain homeostasis in their membrane states with depolarization-activated potassium channels. A sensory neuron should keep a homeostatic level of activity so that it can preserve its sensitivity to deviations in its stimulus-related synaptic inputs. A general theory recently published suggests that the sensory neurons use the subsets of channels for predicting and canceling their inputs, like potassium channels, included in some specific channel types which change the membrane voltage by positive-feedback mechanism. Besides, it has been known that some neurons in motor system show the property of pattern generation such as bistability, stabilized in two states of UP and DOWN with depolarization-activated calcium channels. This bistability means motor neurons represent their outputs as the form of patterns, since the bistability means not only two voltage states but also two of membrane excitability which is the probability that one synaptic input will elicit an action potential. It is proposed that the motor neurons express sets of channels that function to destabilize membrane voltage, using negative feedback mechanisms, for generating patterns. These types of channels can function predict and amplify their input signals to make patterned output in the motor neurons.
To experimentally test this hypothesis of differences, it is important to avoid a lot of cellular diversity. We used the thalamus regarded as a homogenous region and whose nuclei spans to various sensory and motor sites. Thalamocortical (TC) neurons which are the only major cell type of thalamus, of sensory and motor nucleus were recorded by patch clamp method. In our experiment, we observed all TC neurons activated T-type calcium currents that make a neuron to burst at low membrane voltages. However, in sensory nucleus (lateral geniculate nucleus; LGN), the T-type burst threshold voltages of neurons were significantly lower than those of motor nucleus (ventrolateral; VL). LGN neurons were prone to burst, because they activated T-type current at lower voltages than VL. The numbers of spikes during burst in two nuclei were also different; at low voltages, VL neurons showed more action potentials than LGN when shifting to the higher enough voltage state from holding voltages, that most of T-type channels were open.
The results were expected by our hypothesis on sensory and motor neurons. It predicted that T-type current of TC neurons helps to maintain membrane excitability in homeostatic state in the sensory nucleus, regardless of membrane voltages. Whereas, in motor nucleus it helps to have two different membrane excitabilities (as bistability) in order to generate patterns. In practice, the sensory TC neurons in LGN would respond to one synaptic input with the same membrane excitability even in the low voltage state since T-type current evoked readily at that voltage. Otherwise, VL neurons would not respond to most inputs from outside at the low voltages, since they need larger voltage changes to occur. The small membrane excitability might be maintained during most of time at that state in VL. And sometimes when T-type current was activated and the membrane reached the threshold voltage that could make spikes, the neurons would have obviously high membrane excitability. As a result, VL neurons showed two excitabilities.
흔히 신경과학에서 신경계는 감각과 운동 체계로 분류된다. 보통 감각 뉴런은 개체를 둘러싼 환경에 대한 자극 정보를, 운동 뉴런은 행동에 대한 정보를 주로 가진 것으로 정의된다. 그러나 두 체계에 속하는 뉴런들 사이에 뚜렷한 세포 및 분자 수준의 구분 기준이 마련된 적은 없다. 이는 서로 다른 뇌 부위의 세포들이 서로 다른 전기 생리학적 성질을 가지고 있다는 사실이 잘 알려진 것과 대조적이다. 본 연구에서는 감각과 운동 체계 사이에서 세포 수준의 차이가 있다는 가정 하에 이를 검증하기 위한 실험을 수행하였다.
많은 논문들에서 보고된 바에 따르면, 다양한 종류의 감각 뉴런들은 세포막의 항상성을 유지하기 위해 탈분극(depolarization)에 의해 활성화 되는 칼륨 채널을 사용한다. 이들 뉴런들은 감각 신호에 효과적으로 반응할 수 있는 민감성을 유지하기 위해 세포 활성을 일정하게 유지해야 한다. 최근에 발표된 일반 이론(general theory)은, 감각 뉴런들이 포지티브 피드백 원리를 이용하는 특정한 종류의 채널들을 이용하여 인풋 신호를 예측하고 상쇄할 것이라고 제안하였다. 앞서 언급한 칼륨 채널을 예로 들 수 있다.
반면, 척수 운동 뉴런이 두 가지 세포막 전위에서 안정되는 것에서 볼 수 있듯, 운동 체계의 뉴런들은 쌍방 안정(bistability) 등 패턴을 형성하는 특성을 빈번히 나타낸다는 보고가 있었다. 이러한 쌍방 안정 성질은 모터 뉴런들이 아웃풋 신호를 패턴 형태로 내보낸다는 것을 의미한다. 왜냐하면, 쌍방 안정은 단지 세포막의 전위 상태가 두 가지로 유지되는 것을 의미한다기 보다, 뉴런이 하나의 전기적 인풋을 받았을 때 활동 전위(action potential)을 발생시킬 확률인 세포막 활성화도(membrane excitability) 역시 두 가지로 나눠 안정화된다는 사실을 의미하기 때문이다. 이에 따라 우리는 운동 뉴런이 탈분극에 의해 활성화 되는 칼슘 채널 등, 막을 불안정하게 만드는 특정 종류의 채널들을 사용하여 패턴을 만든다는 가설을 제안한다. 이러한 종류의 채널들은 뉴런의 인풋 신호를 예측하고 증폭하여 운동 뉴런이 패턴화된 아웃풋을 만들어내는 데에 도움을 줄 것이다.
위의 가설을 실험을 통해 알아보려면, 세포 종류의 근본적인 다양성을 줄일 필요가 있다. 시상(thalamus)은 다양한 감각 및 운동 부위와 연결되어 있음에도 균일한 세포들로 구성되어 있으므로 본 가설을 시험하기에 이상적인 부위이다. 우리는 감각과 운동 시상핵에 각각 존재하는 시상의 주요 구성 뉴런인 TC 뉴런(thalamocortical neuron; 시상-피질 뉴런)들의 전기적 신호를 패치 클램프 기법으로 측정하였다. 실험 결과, 모든 TC 뉴런들은 낮은 전위 상태에서 버스트(burst)를 일으키는 T-타입 칼슘 채널들을 활성화 시키는 것으로 나타났다. 그러나 주요 감각핵인 LGN에서 T-타입 버스트의 역치 전위(threshold voltage)는 운동핵인 VL에서보다 낮았다. LGN 뉴런이 VL보다 낮은 전위에서도 쉽게 t-타입 전류를 활성화시키고, 따라서 쉽게 버스트를 일으킨다는 것이다. 또한 두 핵은 버스트 동안 활동 전위의 개수에서도 차이를 보였다. 낮은 휴지 전위에 머무르다가 대부분의 T-타입 채널이 활성화 될 수 있는 충분히 높은 전위 상태로 이동하였을 때, VL 뉴런은 LGN보다 많은 수의 활동 전위를 생산해 냈다. 이러한 실험 결과는 감각과 운동 뉴런에 대한 초기 가설과 잘 맞아 떨어진다. 가설은 T-타입 전류가 감각핵에서는 세포막 전위에 관계 없이 TC 뉴런의 세포막 활성화도를 일정하게 유지시키지만, 반대로 운동핵에서는 두 종류의 세포막 활성화도를 유지하여 패턴 형성에 기여할 것으로 예측한다. 실제로, 낮은 전위에서 T-타입 전류가 쉽게 활성화 되므로 LGN의 감각 뉴런은 낮은 인풋에 대해 같은 세포막 활성화도로 반응할 수 있었을 것이다. 반면, 운동핵은 낮은 전위 상태에서 바깥의 신호에 대해 거의 반응하지 않기 때문에, 주로 작은 세포막 활성화도를 가질 것이다. 그러나 종종 T-타입 전류가 활성화 되어 높은 전위에 도달하고, 활동 전위를 발생시키면 뉴런의 세포막 활성화도는 커진다. 따라서 VL 뉴런은 두 가지 종류의 활성화도를 가진다.
