Two-photon fluorescence microscopy has been used widely as powerful tool for in vivo studies. Great depth penetration and high resolution achieved by two-photon fluorescence microscopy are the main advantages. However, in three-dimensional image formation, the profound axial elongation was observed, especially in thick biological specimens. Therefore, it makes hard to figure out the real axial length and that largely affects the volumetry of proteins. The reason is that 3D image formation in the real experimental environment is different from the ideal. Ideal imaging performance of a fluorescence microscopy requires the excitation and emission light to pass through samples with optical properties identical to those of the immersion medium. Any deviation causes optical distortions, known as aberrations. Deconvolution is a computerized inversing Point Spread Function (PSF) method for restoring an image distorted during the image formation process. Therefore, the accuracy and quality of the PSF are essential to ensure the correct outcome. Although good theoretical PSF models exist, they apply only to perfect lenses and well defined optical paths similar to the ideal state, which are never found in practice. It is therefore important to characterize suitable PSF to the individual optical system from experiments. Previous experimental determinations of PSFs for fluorescence microscopy have revealed that experimental PSFs frequently differ significantly from the theoretical PSF. In this paper, I addressed this issue by the comparative study of axial elongation reduction by deconvolution with several PSF models from theoretical model which are adjusted to fit the image from our system and experimental models. I have demonstrated that, for all processed images presented, deconvolution with our experimental PSF clearly improves the image quality and axial resolution. It enhanced contrast and decreased blurring in the axial direction. Moreover, application of experimental PSF, extracted from beads in vivo, to amyloid plaque images in vivo showed significant reduction of axial elongation. The results showed that PSF should be characterized in the most similar environment to the real experimental environment.
다광자 형광 현미경은 깊은 투과성과 적은 손상성 때문에 생체실험에서 널리 이용되고 있다. 그러나 생체조직을 이용한 경우의 3차원 이미지 형성과정에서 현저한 z축 방향 길이 신장이 관찰되었다. 이러한 현상으로 인해 이미지 상에서 z축 길이를 정확히 알기 어려우며 이는 부피측정에도 영향을 미친다. 이러한 현상은 실제 이미지 형성과정이 이상적인 환경과 다르기 때문에 나타난다. 이상적인 형광 현미경에서의 이미지 형성에서는 빛이 샘플과 현미경의 매체를 통과할 때 같은 광학적 성질을 가져야 한다. 약간의 편차도 광학적 수차를 발생시킨다. 역필터링은 점퍼짐함수의 역을 취해주는 방법으로 이미지 형성 과정에서 발생하는 이미지 변형을 복구시킨다. 따라서 이미지 보정을 위해서 정확한 점퍼짐함수가 필요하다. 그러므로 이 논문에서 적합한 PSF를 구함으로써 z축 길이신장의 감소를 연구하였다. 이론적 점퍼짐함수와 실험적 점퍼짐함수로 역필터링을 한 결과, 현저한 z축 길이의 감소를 보였다. 또한 아밀로이드 플라크 길이신장의 보정을 위해서 생체 내에서 z축 해상도보다 작은 크기의 비드를 이용하여 역필터링을 한 결과, 뇌조직 절편에서 구한 PSF보다 더 나은 길이감소를 보였다. 이로부터 이미지 보정에 있어서 실제 이미징 환경과 가장 유사한 조건에서 적절한 점퍼짐함수 정의의 중요성을 알 수 있었다.
