Flap endonuclease1 (Fen1) and Dna2 are two critical endonucleases that work conjointly for processing of Okazaki fragments in eukaryotes. Mus81-Mms4 is another structure-specific endonuclease to resolve stalled replication forks as well as toxic recombination intermediates. In this study, we found that Mus81-Mms4 suppresses the defect of dna2 mutations and that it has functional and physical interactions between Fen1. Mus81-Mms4 stimulated significantly Fen1 activity, accounting for its ability to restore the growth defect caused by dna2 mutation. Surprisingly, Fen1 dramatically stimulated the rate of Mus81-Mms4 cleavage of its all known substrates including regressed replication fork substrates. The ability of Fen1 to stimulate Mus81-Mms4 was independent of the catalytic function of Fen1. The mutual stimulation between Fen1 and Mus81-Mms4 occurred via a specific protein-protein interaction. Our $\It{in vitro}$ data indicate that Mus81-Mms4 and Fen1 are likely to act together during DNA replication via direct interaction. This is supported by the findings that $\It{fen1 mus81}$ or $\It{fen1 mms4}$ double mutants could not rescue of lethality. We discuss the significance of the physical interactions between Fen1, Dna2, and Mus81-Mms4 in context of DNA replication.
Flap endonuclease I (Fen1)과 Dna2 단백질은 세포 내에서 DNA 복제 과정 중에 생기는 5’ flap 구조를 제거하는 효소이다. 5’ flap 구조는 lagging strand DNA를 복제하는 과정에서 polymerase $\delta$ 의 processivity의해 생겨나게 되는데 이 구조를 제거 하지 못하면 DNA 복제가 stalling이 일어나고 DNA break가 쉽게 유도되어 genomic instabilty가 일어난다. 특히 Dna2는 helicase domain 과 nuclease domain이 결합되어 genomic instability가 유도되기 쉬운 긴 5’-flap 구조를 제거하는 역할을 한다. 세포 내 essential한 단백질 Dna2의 backup system은 무엇인가에 대하여 연구하기 위해 yeast multi-copy system을 이용하여 새로운 factor을 찾으려는 실험을 수행하였다.
그 중 Mph1이라는 단백질은 DNA를 translocation 하는데 필요한 ATP를 binding 하는 domain을 가지고 있는 $3’\rightarrow 5’$ helicase로서 human에서 Fanconi anemia 핵심 복합체 중에 하나인 FANCM단백질과 homology가 있다는 사실이 보고되었다. 또한 이 단백질은 C-terminal 부분에 yeast homology인 Mus81 endonuclease와 homology가 있다는 사실을 바탕으로 yeast의 Mph1 과 Mus81의 세포 내 관계를 규명하기 위하여 two-hybrid을 수행하여 관계를 입증했고 Mph1과 더불어 Mus81 단백질 역시 lagging strand 합성에 관여되어 있음을 확인했다.
세포 내에서 Dna2가 없을 때 그 역할을 대신 수행할 수 있는 Fen1의 활성을 증가시켜 줌으로서 genomic stability을 유지한다는 가설 하에 Mus81 단백질이 Fen1을 활성을 증가시키는지 확인하였고 어떠한 mechanism을 통해 활성을 증가시키는지 규명하고자 하였다. 두 단백질은 세포에서 서로 binding하고 있는 것을 확인하였고 그러한 binding은 다른 단백질이나 DNA에 의한 것이 아닌 specific한 protein-protein interaction이었다.
이러한 binding을 통해서 흥미롭게도 Mus81 단백질 또한 Fen1에 의하여 그의 활성이 증가 된다는 사실을 알게 되었다. Mus81은 그 활성을 위하여 partner 단백질인 Mms4 단백질을 필요로 하고 DNA복제가 일어나는 중 DNA damage에 의해 replication fork이 stalling이 일어났을 때 그 특이한 구조를 인식하여 DNA가 복구될 수 있는 구조로 변형시켜주는 initiator 역할을 한다. 이때 DNA합성에 관여하고 있는 단백질 Fen1이 Mus81-Mms4의 활성을 증가시켜 빠른 시간 내에 DNA 복구가 일어날 수 있게 해준다고 설명할 수 있다.
Mus81 단백질은 DNA recombination에 의한 복구 과정 중에 생겨나는 Holliday Junction을 제거하는 역할 또한 수행한다. DNA 복구 과정 중에 생겨나는 중간산물이 제거되지 못한다면 세포는 죽게 된다. 따라서 세포는 이러한 중간산물을 제거하는 여러 pathway를 구축하고 있고 그 중의 한가지 backup system이라고 생각되는 게 Mus81 단백질에 의한 pathway다. Mus81 단백질이 중간 산물을 제거 하는 과정에서 생겨나는 구조 중에서 5’-flap의 구조 역시 생겨나게 되는데 이러한 구조를 빠르게 제거 할 수 있도록 하기 위해 protein-protein binding 에 의해 그 구조의 접근성을 유리하게 해 줄 수 있다.
이 논문에서 DNA metabolism 즉 DNA replication, repair, recombination에 작용하는 단백질들은 서로 긴밀하게 연관되어 있어 여러 가지 형태로 서로의 활성에 영향을 미친다는 결론을 내리고자 한다.
