Screening of novel bacterial strains metabolizing ginsenosides was performed in the ginseng field soil. Through four round screenings, 20 novel strains were found to be able to transform major ginsenosides into minor ginsenosides. The phylogenetic analysis based on the 16S rRNA gene sequences revealed that, three of these isolates, designated as strains Gsoil 202, Gsoil 3054 and Gsoil 3082, were novel species of the genus Arthrobacter in the Actinobacteria, Rhodanobacter in the Gammaproteobacteria and Terrabacter in the Actinobacteria, respectively, and they were characterized by a polyphasic approach to clarify their taxonomic positions. On the basis of the results obtained in this study, we propose that these three strains represent novel species of genus Arthrobacter, Rhodanobacter and Terrabacter, and the names Arthrobacter ginsenosidimutans sp. nov., Rhodanobacter ginsenosidimutans sp. nov. and Terrabacter ginsenosidimutans sp. nov are proposed.
The investigation on the biotransformation pathway of ginsenoside Rb1 revealed that strain Gsoil 202 could transform ginsenoside Rb1 to Rg3, via ginsenoside Rd as an intermediate, by hydrolyzing terminal glucose and consecutively inner glucose at the C-20 position. Strain Gsoil 3054 could hydrolyze not only major ginsenoside Rb1, but also other major ginsenosides Rb2 and Rc, to ginsenoside Rd, by cleaving the β-(1→6)-glucosidic linkage at C-20 of Rb1, the -O-β-D-glucose-(6→1)-α-L-arabinopyranoside linkage attached to the hydroxyl group at the C-20 site of Rb2 and the -O-β-D-glucose-(6→1)-α-L-arabinofuranoside linkage attached to the hydroxyl group at the C-20 site of Rc, respectively. Strain Gsoil 3082 could mainly transform Rb1 to Rg3 via ginsenoside Rd by hydrolyzing terminal glucose and consecutively inner glucose at the C-20 position. Moreover, minor of ginsenoside F2 was produced by the strain Gsoil 3082 via gypenoside XVII by being hydrolyzed one molecule of terminal glucose at the C-20 and the other terminal glucose at the C-3 position of ginsenoside Rb1, which means the strain may contain two kinds of ginsenosidases involved in the hydrolysis of ginsenoside Rb1. The optimum conditions for the ginsenosidase production by these three isolates were investigated.
The purification and characterization of the ginsenoside-hydrolyzing β-D-glucosidases from these three isolates were also performed. The results indicated that the molecular weights of these β-D-glucosidases were about 47, 62 and 68-81 kDa for the strains Gsoil 202, Gsoil 3054 and Gsoil 3082, respectively. In the case of the strain Gsoil 3082, it was found that there were two types of β-D-glucosidases supposed to be involved in the hydrolysis of ginsenoside Rb1. One is able to hydrolyze ginsenoside Rb1 to Rd by cleavaging terminal glucose at the C-20 positon and the other is able to hydrolyze gisenoside Rb1 to gypenoside XVII by cleavaging terminal glucose at the C-3 position. In order to clarify the hypothesis that the strain Gsoil 3082 contains two types of β-D-glucosidases with different pathways for ginsenoside Rb1, cloning and expression of the genes encoding these β-D-glucosidases were performed. As a result, we successfully cloned one of these β-D-glucosidases gene (designated as bgl-gyp 17) involved in the transformation pathway of ginsenoside Rb1 to gypenoside XVII and succeeded in construction of high-copy-number plasmid carrying the bgl-gyp 17 gene and expression in the E. coli C41 (DE3). This is the first report on cloning and overexpression of ginsenoside-hydrolyzing β-D-glucosidase from bacterial strain in the world. It will pave the way for the pharmaceutical research on the minor ginsenosides thus will promote the inductrialization of minor ginsenosides.
인삼밭토양으로부터 4가지의 스크리닝 단계를 거쳐 총 20주의 인삼사포닌전환 신종세균을 분리하였다. 16S rRNA 유전자의 부분 염기서열에 기초한 계통학적 연구결과 그 중에서 Gsoil 202, Gsoil 3054 그리고 Gsoil 3082 균주가 각각 Actinobacteria의 Arthrobacter, Gammaproteobacteria의 Rhodanobacter과 Actinobacteria의 Terrabacter에 속하는 신종세균으로 밝혀졌고 유전형질 및 표현형질의 특성에 대한 연구결과에 기초하여 이들 세 균주를 각각 Arthrobacter ginsenosidimutans sp. nov., Rhodanobacter ginsenosidimutans sp. nov. 와 Terrabacter ginsenosidimutans sp. nov로 제안하였다.
TLC와 HPLC를 이용하여 이들 세가지 균주들이 인삼사포닌 Rb1을 생물전환하는 경로를 연구하였다. Gsoil 202은 일단 Rb1의 20번째 탄소에 결합된 바깥쪽 포도당 하나를 떼어내면서 Rd로 전환시키고, 다음 안쪽의 포도당도 떼어내서 최종적으로 Rg3로 전환하는 양상을 보여주었다. Gsoil 3054는 Rb1의 20번째 탄소에 결합한 바깥쪽 포도당 하나를 가수분해시켜서 Rd로 전환시킬 수 있을 뿐만 아니라 Rb2와 Rc의 20번째 탄소위치에 결합한 바깥쪽 arabipyranoside와 arabionofuranoside를 각각 떼어냄으로써 두 가지 물질 모두 Rd로 전환시킬 수 있다. Gsoil3082는 인삼사포닌 Rb1을 동시에 두 가지 물질로 전환시킬 수 있었다. 한가지는 Rd를 거쳐서 Rg3로 전환시킬 수 있고, 다른 한가지는 gypenoside XVII를 거쳐서 F2로 전환시킬 수 있었다. 이는 Gsoil3082가 두 가지 전환경로에 관여하는 각기 다른 효소를 갖고 있을 수 있다는 것을 암시해 주었다. 따라서 이들 균주들로부터 인삼사포닌전환에 관여하는 효소들을 분리 및 정제하여 특성연구를 진행하였다. 연구결과 Gsoil 202의 경우는 분자량이 47 kDa인 베타글루코시데이즈가 인삼사포닌전환에 관여하였고 pH 4.5와 온도 25 ℃에서 가장 활성이 높았다. Gsoil 3054로부터는 62 kDa인 베타글루코시데이즈를 분리 및 정제하였고 이 효소는 pH 4.8과 온도 30 ℃에서 가장 활성이 높았다. Gsoil 3082는 인삼사포닌 Rb1을 서로 다른 산물로 전환 시킬 수 있는 두 가지 효소가 존재하였고 DEAE-cellulose 칼럼을 통해서는 분리가 어려웠으며 크기는 각각 68과 81 kDa 사이였다.
Gsoil 3082로부터 Rb1의 두 가지 전환경로에 관여하는 글루코시데이즈를 연구하기 위하여 이들 효소들을 발현하는 유전자 클로닝을 시도하였다. 결과적으로 Rb1을 gypenoside XVII로 전환하는 효소를 세계 처음으로 클로닝 하였고 이 유전자를 bgl-gyp 17로 명명하였으며 크기가 2,016 bp였다. 정제를 수월하게 하기 위하여 이 유전자를 pHIS-parallel 벡타에 삽입하여 N-terminal에 $His_6$-tag를 붙힌 뒤에 E. coli C41(DE3)에서 과다발현 시켰다. 발현된 베타글루코시데이즈를 니켈칼럼을 이용하여 분리 및 정제하였고, 분리된 효소는 분자량이 81kDa 이였으며 인삼사포닌 Rb1을 gypenoside XVII로 전환하는 특성을 보여주었다.
본 연구는 세계서 처음으로 인삼사포닌전환에 관여하는 미생물유래 베타글루코시데이즈를 클로닝 및 과다발현에 성공한 사례로서 상당한 학문적 및 산업적 가치가 있다. 금후 마이너 인삼사포닌의 의학적 연구를 위한 토대를 마련하였고 마이너 사포닌의 산업화에 커다란 이바지를 할 것으로 사료된다.
