A series of amphiphilic graft copolymers, poly(aspartic acid) grafted with dodecyl chains with different degree of substitution (DS) were synthesized, and their self-assembled nanoparticles in aqueous solution were prepared by a direct dissolution method. In this study, the morphological changes of self-assembled nanoparticles were induced as a function of DS of dodecyl chains, which are related to hydrophilic/hydrophobic ratio.
At the same polymer concentration condition, the morphologies of self-aggregates are changed from spherical micelles to wormlike micelles, disklike micelles, and finally to tubular networks and vesicles in orders of DS of dodecyl chains. Various morphologies were characterized by TEM, DLS and SANS measurement, and their structural transitions could be explained in terms of packing parameter and interfacial energy.
Charge interaction is also an important factor affecting self-aggregates behaviors. The most important aspect of this study is that a novel complex morphology was observed just by mixing two different charged polymers. The complexes of negatively charged $PAsp-g-C_{12}$ and positively charged PLL showed the structural transition of morphology from spherical micelles to vesicles, and it would be due to the coassembly of molecules induced by the electrostatic charge interaction.
On the other hand, in case of tubular network (A60) and wormlike micelle (A35) aqueous solutions, there was phenomenal increase of viscosity according to the increase of polymer concentration. Their rheological behaviors were confirmed by both steady and dynamic shear measurement using ARES rheometer. And their rheological mechanisms could be explained in terms of the formation of three dimensional tubular networks and the entanglement of wormlike micelles.
Consequently, self-aggregates behaviors and their structures are affected profoundly by the hydrophilic/hydrophobic ratio and the electrostatic charge interaction at molecular level. And rheological behaviors of self-assembled nanostructures are related to the formation of transient networks by the increase of polymer concentration.
생분해성 친수성 고분자인 poly(aspartic acid) (PAsp)에 다음과 같은 합성과정을 통해 소수성인 dodecyl 사슬을 접목한 양친성 고분자를 합성하였다. 우선 아미노산의 한 종류인 $_L$-aspartic acid로부터 PSI를 합성한 후 적절한 양의 dodecylamine과의 aminolysis를 통해 원하는 정도의 dodecyl 사슬을 접목시킨다. Dodecylamine과 반응하지 않은 PSI 단위는 NaOH와 반응시킴으로써 친수성의 PAsp의 고분자 백본을 갖는 양친성 그래프트 공중합체가 합성된다. 이렇게 하여 4가지의 서로 다른 dodecyl 사슬 접목도를 갖는 양친성 고분자 $PAsp-g-C_{12}$를 합성하였고 물에 직접 용해하여 구조체를 유도하였다. 이 연구에서 고분자가 수용액상에서 형성하는 자기조립체의 구조는 고분자의 조성, 즉 dodecyl 사슬의 접목도(DS) 조절에 의한 친수기와 소수기의 비율의 변화를 변수로 하여 관찰되었다. 나노구조체의 구조변화는 주로 TEM을 통하여 관찰하였고 그 밖에 DLS, Small angle neutron scattering을 이용하여 분석하였다.
TEM 사진을 통해 관찰한 결과 dodecyl 사슬의 접목도가 증가함에 따라 자기조립체의 구조가 spherical micelle에서 wormlike micelle, disklike micelle 그리고 tubular network와 vesicle로 변화하였다. 이러한 구조체의 구조변이는 packing parameter와 계면에너지의 관점에서 설명할 수 있다. 먼저 packing parameter의 관점에서 볼 때 소수기의 접목도가 증가함에 따라 고분자에서 dodecyl 사슬 한 개가 갖는 친수기의 면적이 줄어들게 되고 이로 인해 packing parameter가 감소하게 되어 결국 sphere에서 cylinder형, 그리고 마지막으로 bilyer로 구조가 변화하게 되는 것이다. 특히 이 연구에서는 disk, tubule, vesicle과 같은 다양한 형태의 bilayer가 발견되었으며 이러한 bilayer 내의 구조변이는 계면에너지의 관점으로 잘 설명될 수 있다. 양친성 물질은 에너지적으로 평행의 배열을 먼저 선호하며 이는 disk 구조체가 나오는 원인이 된다. 하지만 이러한 disk가 점점 성장하여 그 크기가 커지면서 물과의 접촉면적이 넓어지는 상황이 될 때 표면장력효과로 인한 에너지 손실을 감소하기 위하여 vesicle과 같은 구조체를 형성한다고 할 수 있다.
또한 고분자가 수용액상에서 형성하는 자기조립체의 구조가 charge interaction에 의하여 변화될 수 있는지 알아보기 위하여 음전하를 띠는 양친성 고분자 $PAsp-g-C_{12}$ 와 양전하를 띠는 PLL 사이의 정전기적 charge interaction을 유도하였다. Spherical micelle을 보이던 $PAsp-g-C_{12}$과 PLL을 단순히 섞어주어 $PAsp-g-C_{12}/PLL$ complex를 제조한 결과 구조체가 vesicle형태로 변화하는 것을 TEM 사진을 통해 확인할 수 있었다. 이러한 구조변화는 서로 다른 두 전하를 갖는 분자들간의 상호작용으로 인하여 coassembly가 가능하게 되며 그로 인하여 새로운 구조로 변화할 수 있게 된 것이다.
한편, tubular network와 wormlike micelle을 형성하는 고분자의 농도를 증가시킬 때 용액이 점점 겔화되는 현상이 발견되었다. 이는 고분자 농도 증가로 인하여 구조체 형성이 활발히 진행되며 이로 인해 용액 내에 넓은 network가 형성되었기 때문이다. 각 용액이 가지는 유변학적 물성은 ARES rheometer를 이용한 다양한 실험을 통하여 관찰하였다.
