Gold nanorods, the anisotropic nanostructured plasmonic materials, are attractive for a number of applications in various fields due to their unique optical properties based on the localized surface plasmon resonance (LSPR). In this study, a modified seed-mediated synthesis of gold nanorods (GNRs) with various aspect ratios, functionalization, and their application for plasmonic bionsensors are described. In the synthesis of gold nanorods, variations in reaction conditions have led to the changes of the aspect ratio of gold nanorods. In the presence of silver ions, high yield of gold nanorods in low aspect ratio (3~4) was synthesized while high aspect ratio nanorods were grown in high yield by adding nitric acid. The effect of pH in the growth solution on the aspect ratio was also investigated. In addition, fine-tuning of the aspect ratio was achieved by additional growth of gold nanorods in the presence of gold ions at high temperature.
To utilize short gold nanorods as molecular probes, a new strategy that functionalizes gold nanorods through molecular biomimetics was developed using gold-binding polypeptide (GBP)-fused protein A. The fusion protein with a high affinity to the surface of gold successfully functionalized gold nanorods via the in situ extraction of the unbound cetyltrimethylammonium bromide (CTAB) molecules in the gold nanorod solution. GBP-fused protein A molecules were adsorbed on the surface of gold, partially replacing the CTAB bilayer on the gold nanorods. In order to fabiricate gold nanorod molecular probes (GMPs), anti-C-reactive protein (anti-CRP) was conjugated with gold nanorods through the binding interaction between the protein A domain and the non-antigenic site of antibodies.
The longitudinal plasmon absorption band of gold nanorods is highly sensitive to the refractive index of the environment, which can be changed by the events on the gold surface such as molecular adsorption on the surface. By using this feature were the binding interactions on the GMPs monitored. With variation of the concentration of C-reactive protein (CRP), the plasmon absorption peak shift was measured, which allows the detection of CRP, an invaluable biomarker in human bodies. This system, which measures the plasmon absorption peak shift from the UV-Vis spectra of GMPs in a quarts cell, can be considered as a concept of ‘lab-on-a-tube’. The detection limit was 100ng/ml which was 10-fold lower than the typical clinical cutoff concentration. The plasmonic detection of CRP was also conducted on a microfluidic chip.
금 나노막대는 이방성나노구조를 가지는 플라즈모닉 재료로서, 국부 표면 플라즈모닉 공명 현상 (Localized Surface Plasmon Resonance)에 기반한 독특한 광학성질 때문에 바이오센싱, 바이오 이미징 등 다양한 분야에서 응용되고 있다. 본 연구에서는 미세 입자를 매개로 하는 다양한 가로세로비(Aspect Ratio)의 금 나노막대의 합성, 기능화 및 플라즈모닉 바이오 센서로의 응용 연구에 대해 설명하고자 한다. 반응 조건을 조절함으로서 금나노막대의 가로세로비를 조절할 수 있었으며, 특히, 은 이온을 미량 첨가한 경우 고수율의 균일한 짧은 나노 막대의 합성이 가능하였다. 금 나노막대의 성장 단계에서 질산을 첨가제로 사용한 경우 고 수율 및 고 가로세로비의 금 나노막대를 합성할 수 있었으며, 이 때 pH 조건이 가로세로비에 미치는 영향에 대해 살펴 보았다. 또한, 여분의 금 이온이 존재하는 상태에서 금 나노막대를 고온 성장시킴으로써 정밀한 가로세로비의 조절이 가능하였다.
다음 단계에서는, 낮은 가로세로비의 금 나노막대를 분자 단위의 탐색 도구(Molecular Probe)로 사용하기 위해 금 결합 폴리펩타이드(Gold-Binding Polypeptide, GBP)가 융합된 단백질 A (Protein A)를 이용한 새로운 종류의 금나노막대 기능화 방법을 개발하였다. 생물 모방 기술에 기반하는, 금 나노막대에 항체를 고정화 할 수 있는 효율적인 링커로서 융합단백질을 활용할 수 있었다. 융합단백질은 금 표면에 대한 우수한 결합력을 바탕으로 금 나노막대의 표면에 형성되어 있는 CTAB 이중층을 부분적으로 대체하면서 금 표면에 결합 할수 있었고, 이 과정에서 용액 내 CTAB 분자의 추출을 적용시킴으로서 금 나노막대의 기능화를 안정적으로 완성할 수 있었다. 한편, 기능화된 금 나노막대에 존재하는 혼합 단백질의 단백질 A 부위와 항체 비항원결합 부위간의 결합력을 이용하여 손쉽게 C-반응 단백질 (C-Reactive Protein, CRP) 항체를 금 나노막대에 결합하였다.
이렇게 생산된 금 나노막대 분자 단위 탐색 도구는 금 나노막대의 독특한 광학적 성질을 이용한 플라즈모닉 바이오센서로 적용이 가능하였다. 금 표면 주변 환경의 굴절률 변화에 민감하게 반응하는 세로 플라즈몬 흡수 대역 (The Longitudinal Plasmon Absorption Band)은 금 표면에서의 분자 흡착 등이 일어날 경우 파장 편이 (Wavelength Shift)가 일어나는데, 이를 측정함으로서 바이오센싱이 가능하다. 이러한 원리를 이용하여, C-반응 단백질의 농도 변화에 따른 파장변위를 측정함으로서 중요한 인체내 감염 척도인 C-반응 단백질을 검출할 수 있었다. 석영셀 내에서 금 나노막대 분자 단위 탐색도구의 UV-Vis 분광을 분석하는 본 시스템은 ‘랩온어튜브’ 개념으로서 볼 수 있으며, C-반응 단백질의 검출 한계는 100ng/ml에 달하였다. 이는 일반적인 임상 확정 범위보다 10배 이상 낮은 농도에 해당한다. 한편, 이러한 금 나노막대 기반 플라즈모닉 바이오센싱을 마이크로플루이딕 칩 상에서의 검출에도 적용하였다.
