We report the formation of nonbiofouling, polymeric films, and their applications to selective immobilization of biomacromolecules such as protein and cell. Chapter 2 describes the formation of the biocompatible, nonbiofouling pOEMGA films by SIP of oligo(ethylene glycol) methyl ether methacrylate (OEGMEMA) and subsequently click chemistry for bioconjugation. Chapter 3 describes the covalent bonding of a random copolymer, poly(PEGMA-r-NAS), by “grafting onto” approach on amine-terminated monolayers and the application to selective immobilization of biological materials.
In chapter 2 we report the introduction of biotin as bio-ligand on the nonbiofouling pOEGMA films formed by SIP of OEGMA and subsequently the application of click chemistry for selective immobilization of streptavidin on gold substrates. A thin film of a biocompatible and nonbiofouling poly(oligo(ethylene glycol) methyl ether methacrylate) (pOEGMA) was formed on a gold surface by a combination of the formation of self-assembled monolayers terminating in bromoester―an initiator of atom transfer radical polymerization (ATRP)―and surface-initiated, aqueous ATRP. After the formation of the pOEGMA film, the terminal bromide group of pOEGMA was transformed to the azide group by the reaction with $NaN_3$, and the Sharpless “click”chemistry, triazole formation between azide groups and acetylene groups, was investigated as an approach to bioconjugation. We tested five acetylene group-containing compounds bearing different functionalities, such as methyl, hydroxyl, carboxylic acid, ester, and biotin groups. The non-biofouling effect of the pOEGMA thin films against proteins (bovine serum albumin, fibrinogen, lysozyme, and ribonuclease A, and streptavidin) was confirmed at each step of the processes by ellipsometric measurements, and the specific binding of streptavidin to the biotin-presenting surface was investigated by surface plasmon resonance spectroscopy and enzyme-linked immunosorbent assay. The surfaces were characterized by X-ray photoelectron spectroscopy, FT-IR spectroscopy, and contact angle goniometry.
In chapter 3 we report a facile method for the selective immobilization of biomolecules onto a gold surface that was pre-activated by a polymeric adlayer. The polymeric adlayer was designed to perform triple functions: high resistance to nonspecific protein adsorption, efficient surface anchoring, and subsequent covalent attachment of biomolecules. For this purpose, a random copolymer, poly(PEGMA-r-NAS), was synthesized by radical polymerization of poly(ethylene glycol) methyl ether methacrylate (PEGMA) and N-acryloxysuccinimide (NAS). In the first step, the polymeric adlayer was formed onto amine-terminated self-assembled monolayers (SAMs) on gold through covalent bond formation between reactive N-hydroxysuccinimide (NHS) ester of the copolymer and the amine of the SAMs. In the second step, amine-bearing biotin as a model biomolecule was covalently attached onto the polymeric adlayer that still contained unreacted NHS esters. The degrees of the binding sensitivity for a target protein and the non-specific binding for four model proteins on the biotinylated polymeric adlayer were examined by surface plasmon resonance (SPR) spectroscopy. Finally, the specific immobilization of rhodamin (TRITC)-conjugated streptavidin on the biotinylated polymeric adlayer was achieved by a simple microcontact printing (μCP) technique, resulting in well-defined patterns of the protein.
In chapter 4 we performed SRP and QCM studies on functionalization of poly(oligo(ethylene glycol) methacrylate) films on gold and Si/ $SiO_2$ for immobilization of proteins and cells. Thin films of a biocompatible and non-biofouling poly(oligo(ethylene glycol) methacrylate) (pOEGMA) with various thicknesses were formed on gold and Si/ $SiO_2$ substrates by a combination of the formation of self-assembled monolayers (SAMs) terminating in bromoester-an initiator of atom transfer radical polymerization (ATRP)-and surface-initiated ATRP. After the formation of the pOEGMA films, terminal hydroxyl groups of side chains divergent from the methacrylate backbones were activated with N,N’-disuccinimidyl carbonate (DSC), and the DSC-activated pOEGMA films were reacted with (+)-biotinyl-3,6,9-trioxaundecanediamine (Biotin- $NH_2$) to form biotinylated pOEGMA films. By surface plasmon resonance experiments with the target protein (streptavidin) and model proteins (fibrinogen and lysozyme), we verified that the resulting films showed the enhanced signal-to-noise ratio (~10-fold enhancement) for the biospecific binding of streptavidin, compared with the biotinylated substrate prepared from carboxylic acid-terminated SAMs. Quartz crystal microbalance measurements were also carried out to obtain the surface coverage of streptavidin and fibrinogen adsorbed onto the biotinylated pOEGMA films with various thicknesses, and to investigate the effect of film thicknesses on the biospecific binding of streptavidin. Both the binding capacity of streptavidin and the signal-to-noise ratio of streptavidin/fibrinogen were found to be saturated at the 20-nm-thick pOEGMA film. In addition, to demonstrate a wide applicability of the pOEGMA films, we constructed micropatterns of streptavidin and cells by microcontact-printing Biotin- $NH_2$ and poly-L-lysine onto the DSC-activated pOEGMA films, respectively.
최근 소형화 및 고감도화에 대한 관심과 그 실현 가능성은 최근 나노 기술의 비약적인 발전과 밀접하게 연결되어있다. 바이오센서 칩의 소형화 및 고감도화는 현재 인류의 난제로 치부되는 여러 질병들의 근원적인 소형 분자(small molecules)의 진단 및 치료에 일조해 왔다. 의학계에서는 특정 세포 및 단백질 변이에 의한 질병 발현을 쉽고 빠르게 진달할 수 있는 저비용 혹은 고효율의 진단 센서를 개발하는 데 많은 투자가 이루어져 왔으며, DNA 혼성, 항원-항체 반응, 그리고 단백질-단백질 상호작용과 같은 생체 특이적 상호작용을 인식하는 바이오 소자의 개발에 많은 관심을 가지고 있다. 이러한 바이오 소자가 생체 분자를 효율적으로 검출하기 위해서는 소자표면에서 생체분자간 결합의 선택성을 극대화 할 수 있도록 표면을 설계하는 것이 필수적이다. 즉 생체분자간의 선택적인 결합을 유도함과 동시에 검출과정에 결정적인 문제가 될 수 있는 비특이적 흡착(non-specific adsorption)을 완벽하게 방지할 수 있는 표면처리 기술의 개발이 바이오 소자 개발의 관건이다. 이처럼 바이오 소자의 개발에서 소자의 표면을 설계할 때 대두되는 핵심적인 두 가지 문제 ‘생체물질의 고정화’와 ‘비특이적 흡착의 방지’는 소자의 표면에 코팅되는 유기박막의 설계를 통해 그 해결책을 찾을 수 있을 것으로 기대된다. 이러한 관점에서, 본 연구논문에서는 고분자 물질을 기반으로 한 박막을 고체기판의 표면에 형성하여 생체물질의 선택적 고정화 정도를 극대화하는 반면 비특이적 흡착은 억제시킬 수 있는 표면을 설계하고 제작하는 과정에 그 초점을 맞추고 있다.
원하지 않는 생체 물질의 비특이적 흡착을 억제하거나 제거하기 위해서 다양한 물질들(예를 들면, 다당류, 양쪽성이온 화합물(zwitterionic compound), poly(ethylene glycol) (PEG) 혹은 oligo(ethylene glycol) (OEG) 등)이 연구되고 실제 적용되고 있다. 본 실험실에서는 양쪽성이온 화합물과 PEG 혹은 OEG가 포함된 화합물을 바이오소자 개발에 필요한 표면 제작을 위해 사용하였다. 특히 PEG는 세포와 단백질에 대한 비특이적 흡착 억제성질 이외에도 생체 적합성, 저독성, 비면역반응성, 그리고 물에 대한 고용해도와 같은 부가적인 특성으로 인하여 가장 광범위하게 이용되고 있다.
이러한 비특이적 흡착 억제 성질을 가진 에틸렌 글라이콜 (EG)을 표면에 도입을 위해 크게 물리적 흡착(physisorption)과 화학적 결합 (covalent grafting)및 자기 조립 단분자막 (self-assembled monolayers, SAMs) 형성과 같은 화학적 흡착(chemisorption) 방식이 이용되고 있다. 물리적 흡착은 가역적인 방식으로 진행되는데 외부의 자극 즉 온도, pH, 빛 등에 의해서 쉽게 와해 될 가능성이 있다. 즉 화학적 흡착의 방식과 비교했을 때 작은 결합력에 기인한 낮은 박막의 안정도를 가진다고 볼 수 있다. 그러나 이에 반해 화학적 흡착 방식은 공유결합으로써 비가역적인 방식으로 흡착이 진행되어 물리적인 흡착 과정을 통해 형성된 박막에 비해 훨씬 높은 안정도를 지니게 된다.
이에 본 연구논문에서는 에틸렌 글라이콜 (EG) 작용기를 갖는 고분자물질의 박막을 고체표면에 화학적으로 흡착하고 이를 화학적으로 개질하여 생체물질이 선택적으로 결합할 수 있는 표면을 구현하는 과정에 연구의 초점을 맞추었다. 특히 단분자 박막이 아니라 고분자 박막에 초점을 맞춘 것은 고분자 박막이 가지는 장점 때문인데 단분자막에 비해서 박막의 우수한 안정도, 비특이적 흡착방지 특성, 그리고 다수의 반응기 도입 가능성에 기인한 선택적 결합특성의 증대 등 다양한 효과를 기대할 수 있다.
고분자물질의 화학적 흡착과정에는 일반적으로 “grafting-from” 방식과 “grafting-onto” 방식이 적용된다. 'Grafting-from' 방식은 일반적으로 표면 개시 중합(surface-initiated polymerization, SIP)법에 의한 고분자 박막 형성법을 의미하는데 고체 기판에 개시제를 고정화한 후 이 개시제로부터 단량체가 중합되는 과정을 통하여 고분자 박막이 형성되는 과정을 지칭한다. 이 방식은 표면에 중합되는 단량체의 grafting 밀도를 중합시간과 단량체의 농도로써 제어할 수 있는 이점이 있다. 반면 'grafting-onto” 방식은 이미 합성되어 있는 고분자 물질을 자기조립박막의 형성이나 공유결합의 형성을 이용하여 표면에 고정화하는 방식을 말한다. 본 실험실에서는 크게 세 부분으로 실험을 수행하였는데, 대략적인 내용은 다음과 같다.
첫째, Poly(OEGMEMA) 박막을 원자 전이 라디칼 중합법 (surface-initiated, atom transfer radical polymerization, SI-ATRP)을 이용하여 표면에 형성한 후 표면의 생체물질의 비특이적 흡착 방지 특성을 조사하고 “click chemistry”를 이용한 고분자 개질의 과정을 통해 생체물질의 선택적인 결합에 이용하였다.
둘째, Poly(PEGMA-r-NAS) (poly(ethylene glycol)methyl ether methacrylate (PEGMA)와 N-acryloxysuccinimide (NAS) 의 혼성공중합체(copolymer))를 이용한 고분자 표면 코팅 (grafting-onto approach)을 시도하였다. 합성된 고분자 poly(PEGMA-r-NAS)를 공유결합의 형성과정을 이용하여 표면에 박막의 형태로 도입한 후 고분자 개질의 과정을 통해 생체물질의 선택적인 결합 특성을 도입하였다.
셋째, Poly(oligo(ethylene glycol) methacrylate) (poly(OEGMA))와 poly(3-(methacryloylamino)propyl)-dimethyl(3-sulfopropyl)ammonium hydroxide) (poly(MPDSAH)) 박막의 형성 (grafting-from approach)을 시도하였다. Poly(OEGMA)와 poly(MPDSAH) 박막을 원자 전이 라디칼 중합법 (SI-ATRP)을 이용하여 표면에 형성한 후 표면의 생체물질의 비특이적 흡착 방지 특성을 조사하였다. 특히 poly(OEGMA)박막의 경우 고분자 개질의 과정을 통해 생체물질의 선택적인 결합에 이용하였다. 이때 고분자박막의 3차원 구조에 기반을 두어 생체물질의 선택적 결합 정도를 극대화 할 수 있는 시스템을 제작하였다. 위와 같은 세가지 실험을 통해서 다음과 같은 결론에 도달하였다.
바이오 칩 개발을 위해서 고려해야하는 조건으로서, 민감도(sensitivity), 선택성(selectivity), 재생성(reproducibility), 그리고 대량 검출 처리능력(high-throughput screening, HTS)을 거론할 수 있다. 본 연구논문에서는 민감도 및 선택성을 높일 수 있는 표면의 화학적 처리 방식을 개발하고 유기 박막을 형성하는 것을 연구 목표로 삼아 연구를 수행하였다. 금속이나 무기 물질 표면 위에 생체 물질의 비특이적 흡착을 막을 수 있으면서 우리가 원하는 리간드 혹은 탐침 생체 분자를 도입할 수 있는 고분자를 화학적방법으로 박막을 형성한 후에, 그 표면의 특성을 IR, XPS, ellisometry, contact angle goniometry, SPR 분광기, QCM-D 장비로 분석하였다. 두 가지 기능을 동시에 가지는 고분자 박막은, (1) 단량체 OEGMEMA를 SI-ATRP법을 이용하여 표면 합성한 뒤 'click chemistry' 반응을 통해서 제조하였고, (2) 랜덤 공중합체 poly(PEGMA-r-NAS)를 이용하여 형성하였고, (3) 단량체 OEGMA를 SI-ATRP법으로 표면 합성한 뒤 DSC/DMAP 화합물을 사용하여 제조하였다. 위의 세가지 방법을 통해 형성된 박막의 공통점은 ethylene glycol (EG)기를 사용하였다는 것으로서, EG 기능기를 통해서 생체 물질의 비특이적 흡착을 성공적으로 억제할 수 있었다. 그리고 탄소 삼중 결합과 아지드기의 결합을 가능하게 하는 click 반응은 여러 가지 기능기 즉 메틸 기, 하이드록실기, 페닐기, 카르복실기, biotin 리간드의 도입을 가능하게 하여 생체 물질의 표면 도입을 위한 유용한 반응임을 알 수 있었다. 공중합체에 의한 표면 합성된 고분자 표면은 NHS기가 표면에 노출되어 있기 때문에 생체 물질을 도입하기 위한 리간드를 부착할 수 있었다. 세 번째 방식은 첫 번째와 비교했을 때 표면 합성법에서 큰 차이는 없지만, 고분자 탄소 골격의 수직으로 달려 있는 -OH기를 사용하여 고분자 탄소 골격의 평행한 기능기를 이용한 생체 도입 기능기의 수를 상대적으로 늘릴 수 있었다. 즉 단랑체를 OEGMA로 교체함으로써, 고분자 탄소 골격에 단 하나의 생체 물질을 도입할 수밖에 없었던 poly(OEGMEMA) 고분자 박막과는 다르게 고분자 탄소 골격에 상대적으로 많은 양의 생체 물질 도입을 위한 기능기를 가진 고분자 박막을 형성할 수 있었다. 이와 같은 방법을 통하여, 생체 물질 도입을 위한 기능기의 수가 증가했을 뿐만 아니라 EG기도 동시에 증가했기 때문에 민감도 및 선택성을 동시에 높일 수 있었고, 이 결과는 SPR 및 QCM 실험을 통하여 증명하였다. 반면, 공중합체로 만들어진 고분자 박막은 SI-ATRP 법으로 만들어진 두 고분자 박막에 비하여 민감도가 떨어지는 결과를 보여주었는데, 이것은 비특이적 흡착량이 높게 나와 S/N 비가 두 기판에 비해 낮게 나왔기 때문이다. 결론적으로, 'grafting-onto' 방식을 사용할 경우에 화학적 흡착은 쉽고 빠르게 할 수 있다는 장점은 있지만, 공중합체 고분자의 표면 밀도가 낮고 엉성한 표면을 구성하여 비특이적 흡착량이 높아지게 되었으며, 고분자 표면의 EG기의 낮은 밀도로 인해 생체 물질의 비특이적 흡착 방지 성질이 제대로 기능하지 못한 것으로 사료된다. 본 연구논문를 통해서 EG 작용기를 갖는 고분자 박막과 양쪽성 이온 화합물의 박막은 우수한 안정도와 탁월한 비특이적 흡착방지 특성을 가짐을 알 수 있었다. 특히 EG작용기를 갖는 고분자 박막의 경우에 다수의 반응기 도입 가능성에 기인한 선택적 결합특성의 증대 등 다양한 특성을 조절할 수 있었다. 본 연구 결과를 기반으로 바이오 소자의 개발에서 소자의 표면을 설계할 때 대두되는 핵심적인 문제인 ‘생체물질의 고정화’와 ‘비특이적 흡착의 방지’에 해결책을 제시할 수 있을 것으로 사료된다.
