Sialic acid, the terminal sugar in N-linked complex glycans, is usually found in glycoproteins. Sialic acid residues play a major role in determining the circulatory lifespan of glycoproteins. Therefore, it is often desirable to maximize the sialic acid content of glycoproteins used as therapeutic agents to ensure their quality and consistency. In this study, we tried to enhance the sialylation of recombinant erythropoietin (EPO) in Chinese hamster ovary (CHO) cells.
N-Acetylcysteine (NAC) is one of mild thiol reducing agent. We have previously shown that the anti-apoptotic reagent, NAC not only blocks apoptosis but also increases the production of recombinant erythropoietin. We also found that the productive effect of NAC obtained without the apoptotic condition. However, NAC resulted in decreasing the glycosylation performance, sialylation. Human α2,3-sialyltransferase (α2,3-ST) was introduced into CHO cells, which produce recombinant human EPO, in order to compensate for the reduced sialylation under supplementation of NAC. Sialyltransferase is responsible for terminal sialylation. When NAC was treated in culture media, EPO sialylation was reduced. However, when α2,3-ST was expressed under the supplementation of NAC, reduced sialylation was restored and even more sialylated glycans were produced. Thus, our study is significant in that it offers the possibility of preventing decreased sialylation of EPO while still allowing increased EPO production.
Sialylation requires the metabolic generation of the nucleotide sugar, CMP-sialic acid, followed by the transfer of the sialic acid to an acceptor oligosaccharide in the Golgi apparatus by sialyltransferases. In the case of mammalian cells, CMP-sialic acid is generated from sialic acid by CMP-sialic acid synthase (CMP-SAS). To enhance EPO sialylation, we introduced human α2,3-ST and CMP-SAS into recombinant human EPO-producing CHO cells. The sialylation of EPO was increased by the expression of α2,3-ST alone. However, more sialylation could not be achieved by the co-expression of α2,3-ST and CMP-SAS together, although the intracellular pool of CMP-sialic acid increased. These results imply that sialylation was enhanced only by the expression of α2,3-ST, and the expression of CMP-SAS did not contribute the augmentation of sialylation but increased the CMP-sialic acid level.
Based on the previous observation, it was postulated that CMP-sialic acid transport capacity into the Golgi lumen was limited and thereby causing the lack of CMP-sialic acid substrate for sialylation. Therefore, we co-expressed human α2,3-ST and CMP-SAS, and also over-expressed Chinese hamster CMP-sialic acid transporter (CMP-SAT) in CHO cells, which produce recombinant human EPO. Our hypothesis was that over-expression of CMP-SAT would facilitate the translocation of increased CMP-sialic acid into the Golgi, leading to increase the sialylation in CHO cells. When α2,3-ST, CMP-SAS, and CMP-SAT were over-expressed in CHO cells, the sialic acid content and the relative sialylation of the N-glycans increased compared to our previous results. In particular, we have shown the augmentation of tetra-sialylated glycans, which could not be achieved in our previous study. Over-expression of α2,3-ST, CMP-SAS, and CMP-SAT did not affect CHO cell growth and EPO production. Thus, Over-expression of α2,3-ST, CMP-SAS, and CMP-SAT may be beneficial for producing therapeutic glycoproteins with enhanced sialylation in CHO cells.
시알산(sialic acid)은 당단백질의 당쇄 말단에서 발견되며 당단백질의 생체 내 반감기에 중요한 역할을 한다. 따라서 치료용 당단백질의 품질과 동등성을 보장하기 위해서 시알산의 함량을 늘리는 것이 요구되고 있다. 이러한 이유로 본 연구에서는 CHO(중국 햄스터 난소) 세포에서 생산되는 재조합 에리스로포이에틴 (erythropoietin, EPO)의 시알산을 증가시키고자 하였다.
선행 연구에서 우리는 황환원제인 NAC(N-acetylcysteine)가 아포토시스(apoptosis)를 억제할 뿐만 아니라, 재조합 EPO의 생산성을 향상시킨다는 것을 보고하였다. 또한 아포토시스 조건이 아닌 경우에서도 NAC에 의해 생산성이 향상되었다. 그러나 NAC에 의해 당단백질의 시알산이 감소되었다. 이러한 NAC에 의한 시알산 감소를 극복하고자, 인간 시알산전이효소(α2,3-sialyltransferase, α2,3-ST)를 인간 EPO를 생산하는 CHO 세포에 도입하였다. 시알산전이효소는 시알산을 당쇄말단에 연결시켜주는 효소이다. NAC를 세포 배양시 배지에 첨가하였을 때, 예상했던 것처럼 EPO의 당쇄에서 시알산이 감소하였다. 그러나 NAC 처리시 α2,3-ST를 발현시켜주면, 감소되었던 시알산이 다시 회복되고 오히려 처음보다 더 증가하였다. 이 연구를 통해 EPO의 생산성을 증가시키면서 동시에 시알산의 감소를 막아주는 세포 배양 방법을 확립할 수 있었다.
당쇄의 말단에 시알산이 연결되는 과정은 시알산전이 효소에 의해 골지체에서 이루어지는데, 이때 기질로서 CMP-시알산이 사용된다. 포유류 세포의 경우 CMP-시알산은 CMP-시알산 합성효소(CMP-sialic acid synthase, CMP-SAS)에 의해 합성된다. 인간 α2,3-ST와 CMP-SAS를 재조합 인간 EPO 생산 세포에 도입함으로써 세포 내 CMP-시알산 함량을 증가시키고, 이로 인한 EPO의 시알산 증가를 기대하였다. α2,3-ST만 발현되었을 때, EPO의 시알산은 증가하였다. 그러나 α2,3-ST와 CMP-SAS를 같이 발현하였을 경우, 세포 내 CMP-시알산 함량은 증가하였으나 α2,3-ST 만 발현되었을 때와 비교하여 EPO의 시알산은 더 증가하지 않았다.
이러한 결과에서 당쇄에 시알산이 연결되는 장소인 골지체로의 CMP-시알산 수송 능력이 충분치 않아 세포 내 CMP-시알산이 증가되었지만 EPO의 시알산은 더 이상 증가하지 않았다고 가정할 수 있었다. 이러한 가정을 바탕으로 인간 α2,3-ST와 CMP-SAS의 발현과 더불어 중국 햄스터 CMP-시알산 수송단백질(CMP-sialic acid transporter, CMP-SAT)을 EPO 생산 세포에 과발현 시켰다. CMP-SAT의 과발현을 통해 골지체로 CMP-시알산 수송의 증진과 최종적으로 시알산의 증가를 기대하였다. α2,3-ST, CMP-SAS, CMP-SAT가 과발현 되었을 때 EPO의 시알산은 선행연구에 비해 증가하였다. 특히 α2,3-ST, CMP-SAS의 발현만으로는 증가하지 않았던 시알산 4개가 붙어있는 당쇄의 양이 CMP-SAT의 과발현을 통해 증가하였다.α2,3-ST, CMP-SAS, CMP-SAT 과발현은 세포의 성장과 EPO 생산량에는 영향을 미치지 않았다. 따라서 본 연구에서 구축된 방법은 치료용 당단백질 생산에서 시알산을 증가시키는 데 효과적으로 이용될 수 있을 것이다.
