Separation of particles has many application fields in the bio-chemical research such as purification of sample for chemical analysis, blood cells or blood plasma separation, separation of infected cell s from normal cells, DNA sorting and so on. The most bio-compatible separating method is radiation force driven separation due to its non-contact feature. However, previous methods of optical separation of particles are limited by their low throughput or non-continuous trait.
In present study, a novel optical particle separating method is investigated theoretically and experimentally, to improve the previous optical separating methods. For the bio-chemical application, transparent particles having size range of 1 ~ 100 μm are considered.
In the fundamental study, the analytic expressions for the radiation force on a sphere in a loosely focused Gaussian beam are derived in suitable form for applications by the photon stream method in ray optics regime. For perfect reflecting spheres, an analytic solution for the scattering force and a simplified expression for gradient force are obtained. And, an in situ separation system, cross-type optical chromatography, is developed theoretically and an analytic solution of the retention distance is derived. Particle trajectories in the cross-type optical chromatography are calculated for various sizes and materials of the particles, and flow velocities. Finally, an optical differential mobility analyzer for separating colloidal particles using a loosely focused beam is proposed theoretically and the concept of optical mobility is introduced. For the proposed optical differential mobility analyzer, particle trajectories and retention behavior are discussed and a transfer function described by operational parameters is derived. The possibility of separating particles of various sizes and different chemical compositions is discussed.
In the experimental study, the fabrication process and performance of the proposed optical particle separator are discussed. The retention distance and trajectories of particles are observed experimentally and well agree with theoretical predictions. To evaluate the performance, the selectivity and resolution of the cross-type optical chromatography were determined. By comparing to conventional optical chromatography, the cross-type optical chromatography has higher selectivity and resolution.
In the application study, biological cells are modeled as concentric sphere having non-uniform index of refraction. To evaluate the radiation forces on a biological cell, analytic expressions of radiation forces on a concentric sphere in the loosely focused beam with arbitrary intensity profile are derived. And, the nuclear enlargement of the cancer cells is considered in numerical calculations. Furthermore, the possibility of the cancer cells separation from normal cells in the optical chromatography is discussed and the mechanism of the cancer cell separation is explained by evaluating the trajectories of the normal and cancer cells in the proposed optical separator.
Finally, in the appendix, the effects of radiation forces on human blood cells are observed experimentally. And, demonstration of blood plasma separation using proposed optical separator in present study is performed. The experimental observation shows that real-time blood plasma separation from whole blood sample can be possible by using proposed optical separating method.
In conclusion, a novel optical separating method is proposed and verified theoretically and experimentally. And, the possibility of the real-time cancer cell separation and blood plasma using proposed optical separating method is validated through theoretical approaches, numerical simulations and experiments.
입자 분리는 혈장 및 혈액 세포 분리, DNA 분리 그리고 암세포의 분리 등의 생화학적인 응용분야에 적용이 가능하다. 생물 세포의 생명성을 유지하면서 분리가 가능한 기술로는 광력을 이용하는 방법이 가장 효과적이다. 광력을 이용한 입자 분리 기술은 현재까지 낮은 효율 혹은 비연속적인 과정이라는 단점을 가지고 있다.
본 연구에서는 기존의 광력을 이용한 입자 분리 기술의 단점을 극복할 수 있는 새로운 방식의 연속적이면서 높은 효율을 가지는 광력을 이용한 입자 분리기를 개발하는 것을 목표로 한다. 특히, 생물 세포 분리 응용을 위해 1㎛∼100㎛의 크기를 가지는 투명한 구형 입자를 대상으로 연구를 수행하였다.
먼저, 기초연구에서 광자흐름법과 기하광학 방법을 이용하여 구형 입자에 작용하는 광산란력과 광구배력의 해석적인 표현 식을 구하였고 완전 반사 구형 입자에 대해서는 광산력의 해석해와 광구배력에 대한 간단화된 표현 식을 구하였다. 그리고 교차형 광크로마토 그래피를 이론적으로 제시하고 발전시켰다. 교차형 광크로마토 그래피에서 입자의 크기 혹은 광학적 특성에 따른 거동 특성을 수치적 계산을 통하여 알아보고 분리 거리에 대한 해석적인 근사식을 유도하였다. 마지막으로 광이동도의 개념을 정립하여 광력을 이용한 광이동도 차이를 이용한 입자 분리기에 대한 이론적인 예측 연구를 수행하였다. 그리고 이론적으로 입자의 크기 분리와 동일한 크기의 입자도 광학적인 특성이 다를 경우 분리가 가능하다는 것을 보였다.
실험적 연구에서는 교차형 광크로마토 그래피를 광리소그래피 방법을 이용하여 제작하여 성능을 알아보았다. 입자의 분리 거리와 이동 경로의 특성은 이론적으로 예측된 것과 잘 일치하였다. 그리고 교차형 광크로마토 그래피의 선택지수와 정밀도의 측정 및 예측을 통하여 기존의 광크로마토 그래피와 비교 분석하여 본 연구에서 제시한 교차형 광크로마토 그래피가 선택지수 및 정밀도에서 성능이 우수함을 확인 하였다.
본 연구의 응용으로 광자흐름법을 이용하여 생물 입자에 작용하는 광력에 대한 해석적인 표현 식을 구하였다. 생물 입자를 세포질과 세포핵을 각각 모사하는 동심 구형 입자로 가정하여 기초 연구에서 수행한 방법을 이용하여 생물 세포에 작용하는 광산란력과 광구배력 식을 구하였다. 그리고 암세포의 특징인 세포핵 팽창 특성에 따른 광력을 계산하여 정상 세포에 비해 암세포에 더 큰 광력이 작용함을 이론적으로 예측하였다. 마지막으로 암세포의 세포핵 팽창 특성을 이용하여 본 연구에서 제시한 교차형 광크로마토 그래피를 이용하여 정상 세포로부터 암세포를 분리할 수 있음을 보였다.
마지막으로, 혈액 세포에 작용하는 광력에 대한 효과를 실험적으로 관찰하였다. 그리고 교차형 광크로마토 그래피를 이용하여 혈장 분리가 가능함을 실험적으로 관찰하였다.
