This thesis presents the results of a study on fabrication techniques of bio-compatible microneedles with a shape thickened gradually from sharp 3-D tip to needle shank and base which is needed for the biomedical applications like painless and local drug delivery, sample extraction, and selective treatment.
Fabrication process is divided into two parts: fabrication of microneedle mold and molding process using the molds. Three novel but simple fabrication methods of microneedle mold are developed. Fabrication of PMMA (PolyMethylMetaAcrylic) positive microneedle mold using inclined deep X-ray lithography has been reported by Sang Jun Moon in 2006. No shape limitation when using this method and sharp 3-D tip microneedle mold is fabricated. However, in time and cost aspect it is inefficient to fabricate microneedle array with large space between microneedles which is needed due to flexibility of skin. In this study, a space control method is suggested to fabricate microneedle array with large space between microneedles with high efficiency.
Fabrication method of SU-8 negative microneedle mold using inclined UV lithography is developed. This method has high accessibility because UV is a light source which is commonly used in MEMS process and take short time because little exposure dose is needed to make SU-8 cross linked. SU-8 negative microneedle mold is also fabricated using inclined deep X-ray lithography. Due to short wave length of deep X-ray microneedle mold with shaper tip and smoother surface is fabricated. These microneedle molds fabricated are used as a mass production mold base. By applying PDMS (Poly Di-Methyl Siloxane) molding, hot pressing, and Ni electroplating processes on those microneedle molds, bio-compatible PC (Poly Carbonate) microneedles with sharp 3-D tip are fabricated finally.
In order to test the microneedle’s effectiveness as a tool for transdermal drug delivery, experiment of OVA (Ovalbumin) delivery to the mouse through the microneedles is done. The experiment result shows that the microneedles have sufficient mechanical strength, height, and sharpness to penetrate the skin of mouse and deliver OVA.
미세바늘은 무통증 삽입, 정밀 주사, 그리고 삽입 시 생물조직을 파괴하지 않는 장점을 갖고 있어 의학과 생물학 분야 특히 경피 약물전달, 샘플 추출, 그리고 선택적 치료 분야에서 크게 필요로 하고 있다.
본 논문에서는 이런 분야에 응용 가능한 생체적합성 미세바늘 제작기술에 대해 연구하였으며 경피 약물전달용으로의 제작된 미세바늘의 효용성을 테스트하였다.
생체적합성 미세바늘 제작은 미세바늘 몰드제작과 몰딩공정을 이용한 생체적합성 미세바늘 제작 두 단계로 이루어진다. 몰드 제작에 있어 세가지 방법을 개발하였다. Deep X-ray 경사 노광을 이용한 PMMA 몰드 제작 방법은 2006년에 Sang Jun Moon에 의해 발표되었으며 3차원 형상의 뾰족한 팁을 갖는 PMMA 미세바늘 몰드를 제작하였다. 하지만 시간과 제작비 면에서 볼 때 이 방법을 이용하여서는 보다 넓은 바늘 사이 간격을 갖는 미세바늘 어레이를 제작하기 힘들다는 단점이 있다. 본 연구에서는 바늘 사이 간격을 쉽게 조절할 수 있는 방법을 제시하여 보다 넓은 바늘 사이 간격을 갖는 미세바늘 어레이를 제작하였다.
MEMS 공정에서 보편적으로 사용하는 광원 UV을 이용하여 SU-8 미세바늘 몰드 제작방법을 개발하였다. SU-8는 cross linking되는데 필요로 하는 광량이 적어 노광시간을 현저히 단축시킬 수 있었다. 하지만 자외선의 파장이 deep -X-ray에 비해 길어 미세바늘 몰드의 팁의 뾰족함과 표면의 매끈함 면에서 deep X-ray 경사 노광을 이용하여 제작한 PMMA 미세바늘 몰드에 비해 떨어졌다. 위 두 방법의 장점을 모두 갖는 미세바늘 몰드 제작 방법으로 deep X-ray를 이용한 SU-8 몰드 제작 공정을 개발하였다. Deep X-ray를 사용하므로 예리한 모서리 그리고 매끈한 표면을 갖는 몰드 제작이 가능하였으며 SU-8를 감광재로 사용하므로 노광시간을 단축시킬 수 있었다
경피 약물전달용으로의 미세바늘의 효용성을 테스트 하기 위하여 마우스를 이용한 경피 약물전달 실험을 진행하였다. 실험 결과 약물을 그냥 발랐을 때보다 미세바늘을 사용하여 약물을 전달하였을 때 약물이 더욱 효과적으로 전달됨을 확인할 수 있었다.
